A well-defined functional star polymer nanoparticle was prepared using an “ arm first” approach via RAFT polymerization. In this work, N-adamantylacrylamide and N-succinimidyl acrylate were copolymerized with N-isopropylacrylamide sucessfully. The obtained diblock copolymer regarded as an “ arm” endowed the star polymer with multiple conjugate functions. By adjusting the amount of crosslinking agent, the size and polydispersity of the star polymer were controlled. With the star polymer as the scaffold, a sequential multi-enzyme system was constructed, where HRP was placed in the inner layer through covalent conjugation, and b-cyclodextrin-modified GOx was assembled in the outer layer through hostguest recognition. More interestingly, the activity of the enzyme modified by b-cyclodextrin was improved. In addition to the enhanced thermal stability, this multienzyme system also exhibited a reduced Km value (from 2.18 mM to 0.39 mM) and an excellent substrate affinity. The specificity constant (Kcat/Km) for GOx in the star polymer@ HRP@ GOx system was 1.7-fold higher than that of the free HRP with free GOx system. This strategy will promote the development of biocatalysis reactions and biosensors as a promising method.

New bench-stable reagents with two diazonium sites were designed and synthesized for protein crosslinking. Because of the faster diazonium-tyrosine coupling reaction, hydrogels from the crosslinking of tobacco mosaic virus and the reagent DDA-3 could be prepared within 1 min at room temperature. Furthermore, hydrogels with the introduction of disulfide bonds viaDDA-4 could be chemically degraded by dithiothreitol. Our results provided a facile approach for the direct construction of virus-based hydrogels.

To achieve a high stability in physiological environment and rapid intracellular drug release, a biodegradable zwitterionic triblock copolymer with a disulfide-linked poly-epsilon-caprolactone and polycarboxybetaine methacrylate (PCBMA-SS-PCL-SS-PCBMA) was prepared for micellar carrier to delivery doxorubicin (DOX) into tumor cells. PCBMA-SS-PCL-SS-PCBMA was obtained by following steps: i) introducing disulfide bonds through end-group modification of PCL diol with cystamine dihydrochloride; ii) preparing PCL-RAFT macromolecular chain transfer agent by EDC/NHS chemistry; iii) RAFT polymerization of zwitterionic monomer. Self-assembling from PCBMA-SS-PCL-SS-PCBMA, polymeric micelles had many advantages, such as ultra-low protein absorption in serum and obvious reduction-responsiveness in the presence of DTT. Furthermore, DOX-loaded micelles exhibited high stability upon centrifugation and lyophilization, a fast intracellular drug release and enhanced drug efficacy due to GSH-triggered PCBMA shell shedding and micellar reassembling. Thus, the polymeric micelles integrated several functions and properties could be prospectively utilized as valuable nanocarriers in cancer chemotherapeutics.

Among the numerous homogeneous electrochemical CO2 reduction catalysts, [Ni(cyclam)](2+) is known as one of the most potent catalysts. Likewise, [Ni(isocyclam)](2+) was reported to enable electrochemical CO2 conversion but has received significantly less attention. However, for both catalysts, a purposeful substitution of a single nitrogen donor group by chalcogen atoms was never reported. In this work, we report a series of isocyclam-based Ni complexes with {ON3}, {SN3}, {SeN3}, and {N-4} moieties and investigated the influence of nitrogen/chalcogen substitution on electrochemical CO2 reduction. While [Ni(isocyclam)](2+) showed the highest selectivity toward CO2 reduction within this series with a Faradaic efficiency of 86% for the generation of CO at an overpotential of -1.20 V and acts as a homogeneous catalyst, the O- and S-containing Ni complexes revealed comparable catalytic activities at ca. 0.3 V milder overpotential but tend to form deposits on the electrode, acting as precursors for a heterogeneous catalysis. Moreover, the heterogeneous species generated from the O- and S-containing complexes enable a catalytic hydride transfer to acetonitrile, resulting in the generation of acetaldehyde. The incorporation of selenium, however, resulted in loss of CO2 reduction activity, mainly leading to hydrogen generation that is also catalyzed by a heterogeneous electrodeposit.

Nitrogen-doped MoS2 quantum dots (N-MoS2 QDs) were synthesized via a facile hydrothermal approach, and exhibited high fluorescence quantum yield (QY, 14.9%), excellent photostability, biocompatibility and water solubility. A novel method with good selectivity and sensitivity was established to assay hematin using N-MoS2 QDs as a fluorescent probe based on inner filter effect (IFE). Fluorescent quenching of N-MoS2 QDs has a fine linear dependence with the concentration of hematin in the range of 0.5-15 mu mol/L and a limit of detection of 0.32 mu mol/L (S/N = 3). By the detection method, average concentration of hematin in real health human erythrocytes was measured as 22.5 +/- 3.9 mu mol/L. And, recoveries range varied from 94 to 108% through standard recovery experiment. The N-MoS2 QDs probe shows excellent photostability, low cytotoxicity and anti-interference ability for hematin assay, which may become a promising method for the test of hematin in human blood.

A new method of supramolecular emulsion interfacial polymerization is developed, which can be used to fabricate supramolecular polymeric nanospheres. We designed a water-soluble monomer containing two maleimide end groups, acting as both building block and surfactant, and an oil-soluble supramonomer bearing two thiol groups connected by quadruple hydrogen bonds. With the assist of ultrasonication, hollow nanospheres can be controllably prepared by thiol-maleimide reaction of two monomers at the emulsion interface, which exhibit good stability and dynamic property. In addition, the encapsulated guest molecules could be controllably released from the supramolecular polymeric nanospheres, owing to their stimuli-responsiveness. It is anticipated that this approach will enrich the methodology of supramolecular polymerization and can be applied to constructing supramolecular materials with controllable structures and functions.

Real-time monitor of drug-release from drug formulations in a noninvasive way can provide spatio-temporal information for drug activation and guide further clinical rational administration. In this work, a molecular shuttle, as a typical nanosized artificial molecular machine, was managed to act as a conceptually-new nanotheranostics for oxaliplatin. A post-recognition strategy was utilized, where a default supramolecular-dye couple was pre-blocked. The rational design, synthesis, characterization and proof-of-concept of this strategy were described in detail. The drug-release upon reducing environment can be translated into near-infrared (NIR) fluorescence signal (OFF-to-ON), allowing to track the drug-release procedure by multi-modal images including IVIS, FLECT and photoacoustic imaging. The versatile nanotheranostics system can target to triple negative breast tumor via conjugated F3 peptide, and show an improved anti-tumor efficacy with much lower side effect. The intelligent nanotheranostics system based on molecular shuttle provides new reference for precision medicine in preclinical trial and postclinical evaluation.

Volumetric muscle loss (VML) injuries characterized by critical loss of skeletal muscle tissues result in severe functional impairment. Current treatments involving use of muscle grafts are limited by tissue availability and donor site morbidity. In this study, we designed and synthesized an implantable glycosaminoglycan-based hydrogel system consisting of thiolated hyaluronic acid (HA) and thiolated chondroitin sulfate (CS) crosslinked with poly(ethylene glycol) diacrylate to promote skeletal muscle regeneration of VML injuries in mice. The HA-CS hydrogels were optimized with suitable biophysical properties by fine-tuning degree of thiol group substitution to support C2C12 myoblast proliferation, myogenic differentiation and expression of myogenic markers MyoD, MyoG and MYH8. Furthermore, in vivo studies using a murine quadriceps VML model demonstrated that the HA-CS hydrogels supported integration of implants with the surrounding host tissue and facilitated migration of Pax7(+) satellite cells, de novo myofiber formation, angiogenesis, and innervation with minimized scar tissue formation during 4-week implantation. The hydrogel-treated and autograft-treated mice showed similar functional improvements in treadmill performance as early as 1-week post-implantation compared to the untreated groups. Taken together, our results demonstrate the promise of HA-CS hydrogels as regenerative engineering matrices to accelerate healing of skeletal muscle injuries.

Drugs, including small molecule anticancer drugs, have to be delivered and released into the cytoplasm or nucleus in order to elicit a therapeutic effect. In the present study, a novel corona core-shell nanoparticle was proposed for enhanced cellular uptake and light-responsive intracellular release. Light-responsive core-shell nanoparticles, i.e., gold nanoparticles-coated liposomes (CS), were prepared and further modified with hyaluronic acid (HA) (CCS). HA modification enhances the endocytosis of the nanoparticles by HA receptor-expressing cells, while the plasmonic properties of gold nanoparticles enable their light-triggered drug release. The results demonstrated that the uptake of CCS in HA receptor-expressing MDA-MB-231R cells was significantly enhanced compared with unmodified CS. Nanosecond pulsed laser irradiation (700 nm; 100 mJ/cm(2); 5 pulses) rapidly triggered the intracellular release of a fluorescent dye, 6-carboxyfluorescein (6-CF), from CCS and CS trapped in endolysosomes. The released 6-CF was evenly distributed throughout the entire cytosol and nucleus. Finally, the cytotoxicity of doxorubicin towards MDA-MB-231 cells was significantly increased by CCS delivery upon pulsed laser irradiation. In conclusion, with enhanced cellular uptake and light-triggered intracellular release, CCS significantly enhanced the therapeutic effects of doxorubicin, and may serve as a promising avenue for intracellular drug delivery.

PURPOSE This report describes outcomes of an ongoing quality-improvement project (VitalSign6) in a large US metropolitan area to improve recognition, treatment, and outcomes of depressed patients in 16 primary care clinics (6 charity clinics, 6 federally qualified health care centers, 2 private clinics serving low-income populations, and 2 private clinics serving patients with either Medicare or private insurance). METHODS Inclusion in this retrospective analysis was restricted to the first 25,000 patients (aged >= 12 years) screened with the 2-item Patient Health Questionnaire (PHQ-2) in the aforementioned quality-improvement project. Further evaluations with self-reports and clinician assessments were recorded for those with positive screen (PHQ-2 >2). Data collected from August 2014 though November 2016 were available at 3 levels: (1) initial PHQ-2 (n = 25,000), (2) positive screen (n = 4,325), and (3) clinician-diagnosed depressive disorder with 18 or more weeks of enrollment (n = 2,160). RESULTS Overall, 17.3% (4,325/25,000) of patients screened positive for depression. Of positive screens, 56.1% (2,426/4,325) had clinician-diagnosed depressive disorder. Of those enrolled for 18 or more weeks, 64.8% were started on measurement-based pharmacotherapy and 8.9% referred externally. Of the 1,400 patients started on pharmacotherapy, 45.5%, 30.2%, 12.6%, and 11.6% had 0, 1, 2, and 3 or more follow-up visits, respectively. Remission rates were 20.3% (86/423), 31.6% (56/177), and 41.7% (68/163) for those with 1, 2, and 3 or more follow-up visits, respectively. Baseline characteristics associated with higher attrition were: non-white, positive drug-abuse screen, lower depression/anxiety symptom severity, and younger age. CONCLUSION Although remission rates are high in those with 3 or more followup visits after routine screening and treatment of depression, attrition from care is a significant issue adversely affecting outcomes.
