Three biorthogonal click reactions, a photoinitiated thiol-yne reaction, an azide-alkyne cycloaddition, and a methyltetrazine-transcyclooctene Diels Alder, were used to independently control the presentation of several bioactive proteins to valvular interstitial cells (VICs) in hydrogel scaffolds. Tethered fibroblast growth factor (FGF-2) was found to suppress myofibroblast activation (from 48 +/- 7% to 17 +/- 6%) and promote proliferation (from 10 +/- 2% to 54 +/- 3%) at a concentration of 10 ng/mL. In the presence of the pro-fibrotic cytokine transforming growth factor-beta (TGF-beta 1), FGF-2 could protect the VIC fibroblast phenotype, even at much higher concentrations of TGF-beta 1 than that of FGF-2. With respect to the fibrocalcific VIC phenotype, TGF-beta 1 and bone-morphogenic protein-2 (BMP-2) were found to synergistically promote calcific nodule formation (a five-fold increase in nodules compared to TGF-beta 1 or BMP-2 alone). Exploiting the orthogonal click reactions, FGF-2, TGF-beta 1 and BMP-2 combinations were patterned into distinct regions on a hydrogel to control VIC activation and nodule formation. Cellular crosstalk between separate regions of the same scaffold was affected by the size of each region as well as the interfacial area between different regions. Collectively, these results demonstrate the versatility and robustness of a photoinitiated thiol-yne reaction to template pendant functionalities that allow for the bioconjugation of multiple proteins. This approach maintains protein bioactivity, providing an in vitro platform capable of achieving a better understanding of the complex mechanisms involved in tissue fibrosis.

The utilization of nanotechnology to deliver vaccines and modulate immunity has shown great potential in cancer therapy. Peptide-based supramolecular hydrogels as novel vaccine adjuvants have been found to effectively improve the immune response and tumor curative effect. In this study, we designed a set of reduction-responsive self-assembled peptide precursors (Fbp-G(D)F(D)F(D)Y(D)(E, S, or K)-ss-ERGD), which can be reduced by glutathione (GSH) into Fbp-G(D)F(D)F(D)Y(D)(E, S or K)-SH for forming of hydrogel with different surface properties (E-gel, S-gel, and K-gel, respectively). Using the same method, co-assembled hydrogel vaccines (E-vac, S-vac, and K-vac, respectively) can also be prepared by mixing different precursors with antigens before GSH reduction. Through TEM observation of the nanostructure, we found that all the co-assembled hydrogels, especially K-vac, possessed much denser and more unified nanofiber networks as compared with antigen-free hydrogels, which were very suitable for antigen storage and vaccine delivery. Although the three peptides adopted similar beta-sheet secondary structures, the mechanical properties of their resulted co-assembled hydrogel vaccines were obviously different. Compared to E-vac, S-vac had a much weaker mechanical property, while K-vac had a much higher.In vivoexperiments, co-assembled hydrogel vaccines, especially K-vac, also promoted antibody production and anti-tumor immune responses more significantly than the other two vaccines. Our results demonstrated that co-assembled hydrogels formed by peptides and antigens co-assembly could act as effective vaccine delivery systems for boosting antibody production, and different immune effects can be acquired by tuning the surface properties of the involved self-assembling peptides.

Cell encapsulation within 3D hydrogels is an attractive approach to develop effective cell-based therapies. However, little is known about how cells respond to the dynamic microenvironment resulting from hydrogel gelation-based cell encapsulation. Here, a tunable biomimetic hydrogel system that possesses alterable gelation kinetics and biologically relevant matrix stiffness is developed to study 3D dynamic cellular responses during encapsulation. Hydrogels are synthesized by crosslinking thiolated hyaluronic acid and thiolated chondroitin sulfate with poly(ethylene glycol) diacrylate under cell-compatible conditions. Hydrogel properties are tailored by altering thiol substitution degrees of glycosaminoglycans or molecular weights of crosslinkers. Encapsulation of human mesenchymal stem cells through hydrogel gelation reveals high cell viability as well as a three-stage gelation-dependent cellular response in real-time focal adhesion kinase (FAK) phosphorylation in live single cells. Furthermore, stiffer hydrogels result in higher equilibrium FAK activity and enhanced actin protrusions. Carbon quantum dots (CQDs) show promising potential for tumor imaging owing to their unique superior fluorescent properties. However, the small particle size limits their practical application. Here, pH/reduction dual-responsive comet-shaped PEGylated CQD-DOX conjugate prodrug, DOX-Hy-CQD-SS-PEG with DOX content of 28.5%, was designed with the hydrophobic acid-labile DOX conjugated CQDs as comet nucleus and the few hydrophilic bioreducible detachable PEG brushes as comet tails. The comet-shaped DOX-Hy-CQD-SS-PEG prodrug could self-assemble into unique micelles with mean diameter of 127 nm. The DOX-Hy-CQD-SS-PEG micelles possessed excellent pH/reduction dual-responsive drug release with low drug leakage of 9% in 150 h. Furthermore, the fluorescent CQDs was recovered after DOX release and de-PEGylation, demonstrating their potential application for real-time response of therapy.

For next-generation electronics, it is crucial to be able to accurately mimic the nature of human skin and develop skin-inspired materials and devices for electronic skin (e-skin) applications. However, it is a great challenge to design such a materials because they require the complex integration of stretchability, skin adhesiveness, transparency, healability, and tactile sensing. Herein, we present the first dual covalently reversible dynamic bond-based polymer that can be integrated with various functional materials and used as a skin-like sensor. A series of polymers was designed using polypropylene glycol (PPG)-based polyurethane (PU) as the polymer backbone, and these polymers could integrate two types of covalent reversible dynamic bonds, Diels-Alder and disulfide bonds, in one polymer for the first time. Based on our design, a polymer was created with the advantages of high stretchability, fast healing abilities (60 degrees C, 5 min), skin adhesion, recyclability, high transparency, and tactile sensing abilities. We provide an alternative strategy for the construction of skin-inspired multifunctional materials for e-skin applications.

Molecular probes featuring promising capabilities including specific targeting, high signal-to-noise ratio, and in situ visualization of deep tissues are in great demand for tumor diagnosis and therapy. F-19 magnetic resonance imaging (MRI) techniques incorporating stimuli-responsive probes are anticipated to be highly beneficial for specific detection and imaging of tumors because of negligible background and deep tissue penetration. Herein, we report a cascaded multi-responsive self-assembled nanoprobe, which enables sequential redox-triggered and near-infrared (NIR) irradiation-induced F-19 MR signal activation/amplification for sensing and imaging. Specifically, we designed and synthesized a cascaded multiresponsive F-19-bearing nanoprobe based on the self-assembly of amphiphilic redox-responsive F-19-containing polymers and NIR-absorbing indocyanine green (ICG) molecules. It could realize the activation of F-19 signals in the reducing tumor microenvironment and subsequent signal amplification via the photothermal process. This stepwise two-stage activation/amplification of F-19 signals was validated by F-19 NMR and MRI both in vitro and in vivo. The multiresponsive F-19 nanoprobes capable of cascaded F-19 signal activation/amplification and photothermal effect exertion can provide accurate sensing and imaging of tumors.

Polymers of intrinsic microporosity (PIMs) are a class of microporous polymers with rigid and contorted molecular structures. The special structures lead to incomplete space occupation, and the pores of PIMs originate from the formed void. In this work, we made attempts to expand the applications of PIMs to the aspect of heavy metal removal. First, PIM-1 was synthesized using tetrafluoroterephthalonitrile (TFTPN) and 5,5′,6,6′-tetrahydroxy-3,3,3′,3′-tetramethyl-1,1′-spirobisindane (TTSBI) as precursors. The primary PIM-1 was quite hydrophobic, and thus could not be evenly dispersed in water, resulting in low adsorption capacity for mercury ions (Hg2+). A conversion of nitrile group in PIM-1 to thiol group was then carried out by two steps, namely carboxylation and introduction of thiol groups. The carboxylation made the polymers more active, and the final thiol-functionalization provided the polymers with hydrophilicity and affinity for Hg2+. The thiolethyl modified PIM-1 (assigned as PIM-G) and thiophenyl modified (assigned as PIM-B) possessed maximum Hg2+ adsorption capacity of 136 mg g(-1) and 127 mg g(-1) at pH 5, respectively. Besides, the thiol-functionalized PIMs had fluorescence property and showed potential in sensing for Hg2+.

The carboxylation made the polymers more active, and the final thiol-functionalization provided the polymers with hydrophilicity and affinity for Hg2+. The thiolethyl modified PIM-1 (assigned as PIM-G) and thiophenyl modified (assigned as PIM-B) possessed maximum Hg2+ adsorption capacity of 136 mg g(-1) and 127 mg g(-1) at pH 5, respectively. Besides, the thiol-functionalized PIMs had fluorescence property and showed potential in sensing for Hg2+.
