Objective The development of a new tracer based on the cyclic sulfonamides (sultams) was investigated. Methods 3-(Methoxy-phenyl-methyl)-1,6-dimethyl-1H benzo[c][1,2] thiazine 2,2-dioxide (benzo-delta-sultam) was synthesized and characterized by elemental analysis, FT-IR spectroscopy and single crystal X-ray structure determination. The prepared cyclic sulfonamide was labeled with non-commercial Zn-62 radioisotope for fast in vivo targeting and Coincidence imaging purposes (radiochemical purity 97 % ITLC, 96 % HPLC, specific activity 20-23 GBq/mmol). In vivo biodistribution of the final complex was investigated in Sprague Dawley (R) rats bearing fibro sarcoma tumor after 2, 4 and 8 h post injection and compared with free Zn+2 cation. Results Using instant paper chromatography method, the physicochemical properties of labeled compounds were found sufficiently stable in organic phases, e. g. a human serum, to be reliably used in bioapplications. Conclusions The complex exhibited a rapid as well as high tumor uptake (tumor to blood ratio 4.38 and tumor to muscle ratio 9.63) resulting in an efficient tumor targeting agent.

Methylene Green (MG), a thiazine dye, is catalytically reduced by N2H4 in presence of palladium nanoparticles (Pdn) in buffer media. The observed rate, k(o) increases with increase in [Pdn], [N2H4] and pH but decrease with increase in ionic strength (maintained with NaNO3). Unlike other thiazine dyes, the catalyzed reduction of MG shows an induction period which is caused by the restructuring of Pdn surface. The rate of adsorbate-induced surface restructuring which increase with increase in [N2H4] but independent of [MG], suggest that only N2H4 is adsorbed on Pdn surface during the catalyzed reaction. Interestingly, NO3 ions are also adsorbed on the Pdn surface and turn the surface negatively charged. The reductant, N2H4 is adsorbed on this negatively charged Pdn surface before it reacts with MG. The surface restructuring energy of Pdn, though is constant (78.8 +/- 1.6 kJ M-1) over the temperature range of 288-313 K but the activation energy for the catalyzed reduction varies with temperatures. The convex Arrhenius plot illustrates that activation energy is different in lower (288-304 K, 123.8 +/- 12.6 kJ M-1) and higher temperature range (304-313 K, 69.2 +/- 2.1 kJ M-1) and around the inversion temperature, Tinv (304 K) a change in the nature of transition state takes place. ANTICONVULSANT EVALUATION OF SOME NOVEL 1, 3-THIAZINE DERIVATIVES

In the present study, a series of novel biologically active 8-benzylidene-6-tert-butyl-4-phenyl-5, 6, 7, 8-tetrahydro-benzo-1, 3-thiazin – 2 -imines (TB1-TB12) were synthesized and evaluated for their biological activities. Initially, 2,6-dibenzylidene-4-tert-butylcyclohexanones were synthesized by Claisen-Schmidt condensation of 4-tert-butylcyclohexanone with various aromatic aldehydes in the presence of dilute sodium hydroxide. Further these compounds were subjected to cyclocondensation with thiourea, in isopropyl alcohol, catalyzed by aqueous potassium hydroxide to form 4-aryl-8-arylidene-5,6,7,8-tetrahydro-1H-benzo[d][1,3] thiazin-2(4H)-imines (TB1-TB12). The structures of the newly synthesized compounds have been established on the basis of their spectral data and elemental analysis. Anticonvulsant activity was performed by Maximal electroshocks (MES) method by using Diazepam as standard reference. These compounds were subjected to molecular properties prediction, drug-likeness, lipophilicity and solubility parameters determination using Molinspiration, Osiris program was used for prediction of the toxicity, and also Molsoft and ALOGPS 2.1 softwares. Among all compounds TB5 and TB8 containing lipophilic methoxyl and isopropyl group were more potent whereas TB12 containing hydroxyl groups were least potent among the series.

An efficient two-step procedure of the preparation of (2,2,3,3-tetrafluoropropyl)sulfenyl chloride from 2,2,3,3-tetrafluoropropyl tosylate has been developed. The oxidation of the major regioisomer with hydrogen peroxide afforded ethyl 2-chloro-3-((2,2,3,3-tetrafluoropropyl)sulfonyl)propanoate. The latter compound was found to react with ammonia and aliphatic primary amines under basic conditions with the formation of the acyclic enamines whereas its reactions with aromatic amines afforded ethyl N-aryl-5-(difluoromethyl)-3,4-dihydro-2H-1,4-thiazine-3-carboxylate-1,1-dioxides. Synthesis, Characterization and Molecular Docking of Novel Bioactive Thiazolyl-Thiazole Derivatives as Promising Cytotoxic Antitumor Drug

Reactions of ethylidenethiocarbohydrazide with hydrazonoyl halides gave 1,3-thiazole or 1,3,4-thiadiazole derivatives according to the type of hydrazonoyl halides. Treatment of ethylidenethiosemicarbazide with hydrazonoyl halides and dimethylacetylene dicarboxylate (DMAD) afforded the corresponding arylazothiazoles and 1,3-thiazolidin-4-one derivatives, respectively. The structures of the synthesized products were confirmed by IR, H-1-NMR, C-13-NMR and mass spectral techniques. The cytotoxic activity of the selected products against the Hepatic carcinoma cell line (Hepg-2) was determined by MTT assay indicating a concentration dependent cellular growth inhibitory effect, especially for compounds 14c and 14e. The dose response curves indicated the IC50 (the concentration of test compounds required to kill 50% of cell population) were 0.54 M and 0.50 M, respectively. Confocal laser scanning imaging of the treated cells stained by Rhodamin 123 and Acridine orange dyes confirmed that the selected compounds inhibit the mitochondrial lactate dehydrogenase enzymes. The binding mode of the active compounds was interpreted by a molecular docking study. The obtained results revealed promising cytotoxic activity.
