We present a sensitive single step nanobiosensor for thyroid disease diagnosis. Nanoparticles modified screen printed carbon electrode catalyzes the formation of peptide bond between anti-TSH antibody and amino coated gold nanoparticles rendering a covalently coupled antibody. The nanobiosensor detects and quantifies the thyroid stimulating hormone in the sample by sensing the effective resistance offered by electrode. As the TSH concentration increases in serum sample, more is the immunocomplex formed and higher is the resistance offered by electrode. Gold nanoparticles functionalization with cystamine dihydrochloride facilitates a covalent bonding between surface amino group and carboxylic Fc ensuring maximizing available active antibody. This strategy ensures a much lower limit of detection in addition to improved detection range due to increased loading capacity as a result of larger effective surface area offered by gold nanoparticles. These two aspects of immunosensor fabrication resulted in limit of detection as low as 0.001 mu IU/mL and an enhanced detection range of 0.001-150 mu IU/mL. This makes the developed immunosensor suitable for diagnostic purpose covering the clinically relevant range and a simple detection technique makes it potential candidate for fabrication of a Point-of-Care device for detection of Thyroid dysfunctioning. A dumbbell-shaped copolymer (PDMAEMA)(4)-PCL-SS-PCL-(PDMAEMA)(4) with a lower critical solution temperature (LCST) and redox responses was first synthesized by the combination of ring opening polymerization (ROP), click chemistry and atom transfer radical polymerization (ATRP). Then a dumbbell-shaped copolymer (PDMAPS)(4)-PCL-SS-PCL-(PDMAPS)(4) with an upper critical solution temperature (UCST) and redox responses was obtained through the quaternization reaction between DMAEMA and excess 1,3-propane sultone. Both (PDMAEMA)(4)-PCL-SS-PCL-(PDMAEMA)(4) and (PDMAPS)(4)-PCL-SS-PCL-(PDMAPS)(4) were amphiphilic, and they could self-assemble into spherical micelles under certain conditions. Benefitting from the LCST-type thermoresponse of PDMAEMA and UCST-type thermoresponse of PDMAPS, both dumbbell-shaped copolymers (PDMAEMA)(4)-PCL-SS-PCL-(PDMAEMA)(4) and (PDMAPS)(4)-PCL-SS-PCL-(PDMAPS)(4) presented temperature-responsive properties. The morphology and size of their assemblies would be changed when the temperature is altered. Meanwhile, due to the redox-response of disulfide bonds, both (PDMAEMA)(4)-PCL-SS-PCL-(PDMAEMA)(4) and (PDMAPS)(4)-PCL-SS-PCL-(PDMAPS)(4) showed redox-responsive properties. When dl-dithiothreitol (DTT) was added into the systems, disulfide bonds were broken, causing the change of (PDMAEMA)(4)-PCL-SS-PCL-(PDMAEMA)(4) to PCL-(PDMAEMA)(4), while (PDMAPS)(4)-PCL-SS-PCL-(PDMAPS)(4) changed into PCL-(PDMAPS)(4). The distributions of R-h for both (PDMAEMA)(4)-PCL-SS-PCL-(PDMAEMA)(4) and (PDMAPS)(4)-PCL-SS-PCL-(PDMAPS)(4) assemblies broadened with asymmetric peaks, giving rise to curves with double peaks. After 24 h, DLS curves became symmetric with a single peak, and the R-h was about half that of the original assemblies. In the meantime, dispersed spherical micelles could be observed by TEM.

Multifunctional nanosystems that can transport therapeutic and diagnostic agents into tumor sites and activate their respective functions via tumor-microenvironment recognition are highly desirable for clinical applications. We fabricated pH and redox dual-activatable self-assembled nanotheranostics (named as DA-SNs) via coordination-driven self-assembly of chlorin e6 (Ce6) disulfide-linked pH sensitive polymer ligand, poly (isobutylene-alt-maleic anhydride-graft-methoxy-poly (ethyleneglycol)-graft-imidazole-graft-Cystamine-Ce6) [PIMA-mPEG-API-SS-Ce6], and gadolinium ions (Gd3+). DA-SNs exhibited uniform particle size of similar to 48 nm, excellent stability, and inherent biosafety. Negatively charged DA-SNs could prolong blood circulation time (t(1/2) = 2.91 h) and improve tumor accumulation. Moreover, DA-SNs could undergo surface charge switch from negative charge to positive one in a slightly acidic tumor extracellular environment (pH 6.8), thus enhancing cellular uptake. After entering tumor cells, fluorescence, photodynamic therapeutic activity, and T1MR contrast from DA-SNs could be activated within this intracellular environment with lowered pH and high level of GSH. Importantly, human tumors implanted in mice could be successfully visualized via distinct pH and redox dual-sensitive T1MR contrast and fluorescence imaging, indicating that DA-SNs could serve as a dual-modal MR/fluorescence imaging probe for tumor-targeting diagnosis. In addition, DA-SNs exhibited superior photodynamic therapeutic efficiency with negligible side effects. Therefore, this DA-SN shows great promise for synergistic photodynamic therapy and diagnostic imaging.

Isotactic polypropylene (PP) is one of the most popular plastics. However, the remarkably low surface energy of PP prevents the surface functionalization of PP. We studied the surface functionalization of PP by dip-coating with a maleic anhydride-grafted chlorinated polypropylene (MPO)/methacrylate-based terpolymer mixture. A methacrylate-based terpolymer (PMFP) was synthesized, which contained perfluoroalkyl (R-f)-conjugated monomers and poly(ethylene glycol)-conjugated monomers. Tape-peeling tests revealed that MPO successfully immobilized PMFP on a PP surface, although PMFP was less adhesive to PP. X-ray photoelectron spectroscopy (XPS), contact angle, and protein adsorption measurements revealed that the R-f groups and PEG chains in PMFP were segregated to the outermost surface of the dip-coated layer. The surface segregation of these moieties produced a low-fouling surface on the PP substrate. In addition, we synthesized a terpolymer that contained R-f groups and PEG chains with carboxy groups at their termini (PMFB) and used it to dip-coat a PP substrate. The surface segregation of side chains in PMFB induced the presentation of carboxy groups at the outermost surface, which were used as reactive sites for enzyme immobilization.

Soft poly(N-vinylcaprolactam) (PNVCL) based nanogels were prepared and surface-decorated with gold nanoparticles (AuNPs). The applicability of the hybrid nanogels (PNVCL-AuNPs) as carriers for low molar mass substances was of special interest. AuNPs protected with a mixture of 11-azidoundecanothiol and 11-mercaptoundecanoic acid were bound to propargyl functionalized PNVCL based nanogels. Acidic groups on the surfaces of AuNPs and PNVCL based nanogels stabilize the particle dispersions against precipitation above the phase transition temperature of PNVCL. Both the neat PNVCL nanogels and the PVCL-AuNPs shrink upon heating the dispersions. Even though the AuNPs are mainly located in the soft surface layer of the nanogels, the PNVCL-AuNPs respond to visible light as well as to radio-frequency (RF) irradiation by shrinking due to the AuNPs acting as nanoheaters. Interactions of linear PNVCL, PNVCL nanogels and PNVCL-AuNPs with two fluorescent probes were studied as function of increasing temperature. Once bound to the polymer the fluorescent probe may or may not be released from it, depending on its polarity and water solubility. Presence of AuNPs changed the release behavior of the water soluble charged fluorescent probe from the nanogels.

Background Turf toe injuries, though most common in athletes, can also occur in non-athletes. No study exists in the current literature investigating operative outcomes in non-athlete patients with chronic turf toe injury. In this study, we present our outcomes on operatively treated turf toe injuries in non-athletes in the only cohort yet studied. Methods Using ICD-10 codes, we assembled a cohort of 12 patients who underwent operative repair of chronic turf toe injury from January 2012 through January 2018 at the investigating institution. These 12 patients were evaluated to determine demographic information, method of injury, length of time from injury to surgery, clinical and radiologic characteristics of the injury, and operative outcomes including mean preoperative and postoperative VAS (Visual Analog Scale) scores, preoperative and postoperative FFI (Foot Function Index) scores, and postoperative complications. Results On initial clinical presentation, all 12 patients had local tenderness with associated painful range of motion. Four patients had restricted range of motion, all patients had a positive Lachman test, two had local edema, and eight had hallux valgus deformity. Mean VAS improved from 4.6 (range 2-9) to 1 (range 0-4). Mean FFI improved from 102.5 (range 56-177) to 61.75 (range 23-144). All patients had a negative Lachman test at final follow-up. No patients developed major complications or required revision surgery. Conclusions Our study is the first to investigate operative outcomes following chronic turf toe injury in non-athlete patients. Based on our study, surgeons and patients can expect significant improvement in overall pain and function following surgery.

Melphalan (MEL) is an effective chemotherapeutic agent for treatment of retinoblastoma (Rb) which is the most common childhood malignancy. However, the inherent cardiopulmonary toxicity and hazardous integration limit its therapeutic effect on RB. N-Acetylheparosan (AH), a natural heparin-like polysaccharide in mammals with long circulation effect and good biocompatibility, was linked by d-alpha-tocopherol acid succinate (VES) via and cystamine (CYS) to synthesize reduction-responsive N-acetylheparosan-CYS-Vitamin E succinate (AHV) copolymers. In addition, CYS was replaced by adipic acid dihydrazide (ADH) to obtain a control of non-reduction-responsive polymers N-acetylheparosan-ADH-Vitamin E succinate (ADV). MEL-loaded AHV micelles (MEL/AHV) as well as ADV micelles (MEL/ADV) were prepared with small particle size and high drug loading content. In vitro drug release showed that MEL/AHV micelles presented obvious reduction-triggered release behavior compared with MEL/ADV. In vitro antitumor effects were investigated using WERI-Rb-1 retinoblastoma cells. Cytotoxicity experiments showed that the IC50 of MEL/AHV was significantly lower than that of free MEL and MEL/ADV, suggesting that MEL/AHV enhanced the cytotoxicity against retinoblastoma cells. Furthermore, MEL/AHV micelles were more easily uptaken by multiple pathways compared with MEL/ADV and free MEL. Cancer cells have reductive and acidic environments as compared to the normal body cells. Development of reductive/acidic responsive polymersomes will play a major role in cancer therapy to trigger the release of the loaded drug. In our work, we synthesized three different reductive/acidic dual-responsive polymers, poly[(mPEG-SS-amino) (N,N-diisopropylethylenediamino)phosphazenes] (PPDPs) in different mole ratios of side groups. These PPDPs were characterized by H-1 NMR, P-31 NMR, FT-IR and GPC. After that, the PPDPs were allowed to self-assemble into drug-loaded polymersomes with high loading content and encapsulation efficiency of hydrophilic/hydrophobic anticancer drugs. The hydrophilic anticancer drug doxorubicin hydrochloride (DOX center dot HCl) and hydrophobic drug doxorubicin were used. These PPDPs-based polymersomes showed reductive/acidic stimuli-responsive release of anticancer drugs. Moreover, these polymersomes also exhibited suitable hydrodynamic diameters, which will facilitate the longtime circulation in bloodstream due to avoiding renal clearance and close contact to the tumor cells in vascular sections due to enhanced permeability and retention effect. Collectively, these developed polymersomes may provide an effective platform for anticancer drugs delivery.

To mimic the helical structure and function of biopolymers, shell cross-linked nanoparticle (P4) composed of left-handed helical poly(phenylborate isocyanide) in core and hydrophilic polyisocyanide in shell was prepared. The phenylborate in the core and the disulfide bonds in the cross-linkage render the nanoparticle with excellent dual stimuli-responsiveness to glutathione (GSH) and H2O2. Nevertheless, it has good stability in normal physiological conditions. Because of the helicity and borate pendants of the core, such nanoparticle has high capacity for anticancer drug loading, for example, the loading capacity of doxorubicin (DOX) was up to 68%. Moreover, the DOX-loaded DOX@P4 showed excellent tumor cell penetration potency and fast drug release. More than 78% of murine breast cancer cell (4T1) can be killed within 48 h, supporting this material with great potential in antitumor drug nanocarriers.
