A synthetic method is presented for S-N bond formation starting from cheap and affordable materials. We show that (un)substituted N-protected cyclic eight-membered C-2-symmetric sulfenamides have been prepared in a few steps using this procedure. The synthetic utility of these ambipolar derivatives was demonstrated in a variety of synthetic transformations affording different S,N-heterocyles of pharmaceutical relevance in one or two steps from simple starting materials.

Drugs, including small molecule anticancer drugs, have to be delivered and released into the cytoplasm or nucleus in order to elicit a therapeutic effect. In the present study, a novel corona core-shell nanoparticle was proposed for enhanced cellular uptake and light-responsive intracellular release. Light-responsive core-shell nanoparticles, i.e., gold nanoparticles-coated liposomes (CS), were prepared and further modified with hyaluronic acid (HA) (CCS). HA modification enhances the endocytosis of the nanoparticles by HA receptor-expressing cells, while the plasmonic properties of gold nanoparticles enable their light-triggered drug release. The results demonstrated that the uptake of CCS in HA receptor-expressing MDA-MB-231R cells was significantly enhanced compared with unmodified CS. Nanosecond pulsed laser irradiation (700 nm; 100 mJ/cm(2); 5 pulses) rapidly triggered the intracellular release of a fluorescent dye, 6-carboxyfluorescein (6-CF), from CCS and CS trapped in endolysosomes. The released 6-CF was evenly distributed throughout the entire cytosol and nucleus. Finally, the cytotoxicity of doxorubicin towards MDA-MB-231 cells was significantly increased by CCS delivery upon pulsed laser irradiation. In conclusion, with enhanced cellular uptake and light-triggered intracellular release, CCS significantly enhanced the therapeutic effects of doxorubicin, and may serve as a promising avenue for intracellular drug delivery.

Volumetric muscle loss (VML) injuries characterized by critical loss of skeletal muscle tissues result in severe functional impairment. Current treatments involving use of muscle grafts are limited by tissue availability and donor site morbidity. In this study, we designed and synthesized an implantable glycosaminoglycan-based hydrogel system consisting of thiolated hyaluronic acid (HA) and thiolated chondroitin sulfate (CS) crosslinked with poly(ethylene glycol) diacrylate to promote skeletal muscle regeneration of VML injuries in mice. The HA-CS hydrogels were optimized with suitable biophysical properties by fine-tuning degree of thiol group substitution to support C2C12 myoblast proliferation, myogenic differentiation and expression of myogenic markers MyoD, MyoG and MYH8. Furthermore, in vivo studies using a murine quadriceps VML model demonstrated that the HA-CS hydrogels supported integration of implants with the surrounding host tissue and facilitated migration of Pax7(+) satellite cells, de novo myofiber formation, angiogenesis, and innervation with minimized scar tissue formation during 4-week implantation. The hydrogel-treated and autograft-treated mice showed similar functional improvements in treadmill performance as early as 1-week post-implantation compared to the untreated groups. Taken together, our results demonstrate the promise of HA-CS hydrogels as regenerative engineering matrices to accelerate healing of skeletal muscle injuries.

Sequence specific molecules with high folding ability (i.e., foldamers) can be used to precisely control the distribution and projection of side chains in space and have recently been introduced as tailored systems for delivering nucleic acids into cells. Designed oligourea sequences with an amphipathic distribution of Arg- and His-type residues were shown to form tight complexes with plasmid DNA, and to effectively promote the release of DNA from the endosomes. Herein, we report the synthesis of new cell-penetrating foldamer sequences in which the foldamer segment is conjugated via a reducible disulfide bond to a ligand that binds cell-surface expressed nucleoproteins with the idea that this system could facilitate both assemblies with nucleic acids and cell entry. This new system was evaluated for delivery of DNA in several cell lines and was found to compare favorably with all comparators tested (DOTAP and b-PEI as well as a number of known cell penetrating peptides) in various cell lines and particularly in culture medium containing up to 50% of serum. These results suggest that this dual molecular platform which is long lasting and noncytotoxic could be of practical use for in vivo applications.

Among the numerous homogeneous electrochemical CO2 reduction catalysts, [Ni(cyclam)](2+) is known as one of the most potent catalysts. Likewise, [Ni(isocyclam)](2+) was reported to enable electrochemical CO2 conversion but has received significantly less attention. However, for both catalysts, a purposeful substitution of a single nitrogen donor group by chalcogen atoms was never reported. In this work, we report a series of isocyclam-based Ni complexes with {ON3}, {SN3}, {SeN3}, and {N-4} moieties and investigated the influence of nitrogen/chalcogen substitution on electrochemical CO2 reduction. While [Ni(isocyclam)](2+) showed the highest selectivity toward CO2 reduction within this series with a Faradaic efficiency of 86% for the generation of CO at an overpotential of -1.20 V and acts as a homogeneous catalyst, the O- and S-containing Ni complexes revealed comparable catalytic activities at ca. 0.3 V milder overpotential but tend to form deposits on the electrode, acting as precursors for a heterogeneous catalysis. Moreover, the heterogeneous species generated from the O- and S-containing complexes enable a catalytic hydride transfer to acetonitrile, resulting in the generation of acetaldehyde. The incorporation of selenium, however, resulted in loss of CO2 reduction activity, mainly leading to hydrogen generation that is also catalyzed by a heterogeneous electrodeposit.

A dumbbell-shaped copolymer (PDMAEMA)(4)-PCL-SS-PCL-(PDMAEMA)(4) with a lower critical solution temperature (LCST) and redox responses was first synthesized by the combination of ring opening polymerization (ROP), click chemistry and atom transfer radical polymerization (ATRP). Then a dumbbell-shaped copolymer (PDMAPS)(4)-PCL-SS-PCL-(PDMAPS)(4) with an upper critical solution temperature (UCST) and redox responses was obtained through the quaternization reaction between DMAEMA and excess 1,3-propane sultone. Both (PDMAEMA)(4)-PCL-SS-PCL-(PDMAEMA)(4) and (PDMAPS)(4)-PCL-SS-PCL-(PDMAPS)(4) were amphiphilic, and they could self-assemble into spherical micelles under certain conditions. Benefitting from the LCST-type thermoresponse of PDMAEMA and UCST-type thermoresponse of PDMAPS, both dumbbell-shaped copolymers (PDMAEMA)(4)-PCL-SS-PCL-(PDMAEMA)(4) and (PDMAPS)(4)-PCL-SS-PCL-(PDMAPS)(4) presented temperature-responsive properties. The morphology and size of their assemblies would be changed when the temperature is altered. Meanwhile, due to the redox-response of disulfide bonds, both (PDMAEMA)(4)-PCL-SS-PCL-(PDMAEMA)(4) and (PDMAPS)(4)-PCL-SS-PCL-(PDMAPS)(4) showed redox-responsive properties. When dl-dithiothreitol (DTT) was added into the systems, disulfide bonds were broken, causing the change of (PDMAEMA)(4)-PCL-SS-PCL-(PDMAEMA)(4) to PCL-(PDMAEMA)(4), while (PDMAPS)(4)-PCL-SS-PCL-(PDMAPS)(4) changed into PCL-(PDMAPS)(4). Background: Black patients have higher rates of hospitalization for acute heart failure than other race/ethnic groups. We sought to determine whether diuretic efficiency is associated with racial differences in risk for rehospitalization after acute heart failure. Methods: A post hoc analysis was performed on 721 subjects (age, 68 +/- 13 years; 22% black) enrolled in 3 acute heart failure clinical trials: ROSE-AHF (Renal Optimization Strategies Evaluation in Acute Heart Failure), DOSE-AHF (Diuretic Optimization Strategy Evaluation in Acute Decompensated Heart Failure), and CARRESS-HF (Cardiorenal Rescue Study in Acute Decompensated Heart Failure). Repeated-measures ANOVA was used to test for a racextime effect on measures of decongestion. Diuretic efficiency was calculated as net fluid balance per total furosemide equivalents. In a subset of subjects, Cox regression was used to examine the association between race and rehospitalization according to plasma renin activity (PRA). Results: Compared with nonblack patients, black patients were younger and more likely to have nonischemic heart failure. During the first 72 to 96 hours, there was greater fluid loss (P=0.001), decrease in NT-proBNP (N-terminal pro-B-type natriuretic peptide;P=0.002), and lower levels of PRA (P<0.0001) in black patients. Diuretic efficiency was higher in black than in nonblack patients (403 [interquartile range, 221-795] versus 325 [interquartile range, 154-698];P=0.014). However, adjustment for baseline PRA attenuated the association between black race and diuretic efficiency. Over a median follow-up of 68 (interquartile range, 56-177) days, there was an increased risk of all-cause and heart failure-specific rehospitalization in nonblack patients with increasing levels of PRA, while the risk of rehospitalization was relatively constant across levels of PRA in black patients. This study aimed to develop an efficient step-by-step osteosarcoma (OS)-targeting liposome system functionalized with a redox-cleavable, bone- and cluster of differentiation 44 (CD44)-dual-targeting polymer. Furthermore, the effect of coadministration of a tumor-penetrating peptide, internalizing RGD (iRGD), was investigated. First, a bone-targeting moiety, alendronate (ALN), was conjugated with hyaluronic acid (HA), a ligand for CD44. This ALN-HA conjugate was coupled with DSPE PEG(2000)-COOH through a bioreducible disulfide linker (-SS-) to obtain a functionalized lipid, ALN-HA-SS-L, to be postinserted into preformed liposomes loaded with doxorubicin (DOX). The roles of ALN, HA, and the redox sensitivity of the ALN-HA-SS-L liposomes (ALN-HA-SS-L-L) in the anti-OS effect were critically evaluated against various reference liposomal formulations (with only ALN, HA, or redox sensitivity). ALN-HA-SS-L-L displayed a zeta potential of -26.07 +/- 0.32 mV and selectively disassembled in the presence of a reducing agent, 10 mM glutathione, which can be found in cancer cells. cells. The antitumor effects of various liposomes were ranked in the same order as the degree of tumor biodistribution shown by in vivo/ex vivo imaging: ALN-HA-SS-L-L coadministered with iRGD > ALN-HA-SS-L-L > HA-SS-L-L > HA-L-L > PEG-L> free drug. ALN-HA-SS-L-L/DOX also reduced the cardiotoxicity of DOX and lung metastases. Overall, this study demonstrated that ALN-HA-SS-L-L/DOX, equipped with bone- and CD44-dual-targeting abilities and redox sensitivity, could be a promising OS-targeted therapy. The efficacy could also be augmented by coadministration of iRGD.

Hypoxia plays vital roles in the development of tumor resistance against typical anticancer therapies and local reoxygenation has proved effective to overcome the hypoxia-induced chemoresistance. Perfluorocarbon (PFC) is an FDA approved oxygen carrier and currently vigorously investigated for oxygen delivery to tumors. This study reports a perfluorocarbon and etoposide (EP) loaded porous hollow Fe3O4-based theranostic nanoplatform capable of delivering oxygen to solid tumors to enhance their susceptibility against EP. Results show that oxygen could be released at a moderate rate from the porous hollow magnetic Fe3O4 nanoparticles (PHMNPs) over an extended period of time, therefore effectively reducing the hypoxia-induced EP resistance of tumor cells. Moreover, the surface of PHMNPs was modified with lactobionic acid (LA)-containing amphiphilic polymers via hydrophobic interaction, which could provide targeting effect against certain types of tumors. The hydrophilic moiety would be subsequently shed by the intratumoral GSH after cellular internalization and result in the agglomeration of nanocarriers inside tumor cells, consequently impeding the nanoparticle exocytosis to enhance their intracellular retention. The enhanced retention could elevate the intracellular EP level and effectively boost the tumor cell killing effect. In addition to the therapeutic benefits, the Fe3O4 nanocage could also be used for the magnetic resonance imaging of the tumor area. The assorted benefits of the composite nanosystem are anticipated to be advantageous for the treatment of drug-resistant hypoxic tumors.
