Nitrogen-doped MoS2 quantum dots (N-MoS2 QDs) were synthesized via a facile hydrothermal approach, and exhibited high fluorescence quantum yield (QY, 14.9%), excellent photostability, biocompatibility and water solubility. A novel method with good selectivity and sensitivity was established to assay hematin using N-MoS2 QDs as a fluorescent probe based on inner filter effect (IFE). Fluorescent quenching of N-MoS2 QDs has a fine linear dependence with the concentration of hematin in the range of 0.5-15 mu mol/L and a limit of detection of 0.32 mu mol/L (S/N = 3). By the detection method, average concentration of hematin in real health human erythrocytes was measured as 22.5 +/- 3.9 mu mol/L. And, recoveries range varied from 94 to 108% through standard recovery experiment. The N-MoS2 QDs probe shows excellent photostability, low cytotoxicity and anti-interference ability for hematin assay, which may become a promising method for the test of hematin in human blood.

Tedious polymeric modifications, laborious syntheses, low water solubility, high cytotoxicity and low quantum yields are still the major concerns related to fluorescent materials used for biosensing applications. Here we report one-pot facile synthesis of polyethylene glycol (PEG)-modified cystamine-based multifunctional fluorescent dyes named as CPEG-168, CPEG-1000 and CPEG-2000, which can be used for biosensing and detection of both cations and anions in pure aqueous medium. Rapid response toward thiols could be achieved due to the presence of disulfide functionality and reversible redox property between thiol and disulfide. A cyclic structure is formed by (NH-H) intramolecular hydrogen bonding to produce electron dense region for strong fluorescence emission. The presence of -NH group made them sensitive toward Cu2+ ions, while excellent sensitivity toward pH and F- ions was provided by intramolecular hydrogen bonding between -NH groups. The increase in chain length of PEGs enhanced the fluorescence emission intensities due to the formation of more rigid structure, and hence, the desired quantum yields can be tuned by changing the chain length of PEG. The aforementioned properties along with high brightness in aqueous solution, at biological pH and temperature range, make these fluorescent dyes potential candidates for biosensing applications and detection of cations and anions.

Combined blockades of CTLA-4 and PD-1 can yield better overall complementary clinical outcomes than individual blockades, but the response rates are still relatively low. To investigate the anti-tumor effects of various combined strategies, we designed various spherical nucleotide nanoparticles (SNPs) loaded with CTLA-4 aptamer (cSNPs), PD-1 siRNA (pSNPs) or both (hybrid SNPs, or hSNPs). The results demonstrated that hSNPs could promote significantly stronger anti-tumor immune responses in a nonredundant fashion than the mixture of pSNPs and cSNPs (pSNPs & cSNPs). We reasoned that this is because all individual immune cells could receive both CTLA-4 and PD-1 blockades when they engulfed hSNPs, but it is much less likely that individual immune cells could receive both CTLA-4 and PD-1 blockades as many of them may not take both pSNPs and cSNPS from pSNPs & cSNPs. Further results revealed that the synergistic immune stimulatory effects of CTLA-4 and PD-1 blockades in the form of hSNPs were at least partly through regulating the immune suppressive function of both Tregs and TIM3(+)exhausted-like CD8 T cells and allowing effector T cells to expand. This mechanism is not identical to earlier reported mechanisms of CTLA-4 and PD-1 blockades.

A compressive strain applied to bilayer films (e.g., a thin film adhered to a thick substrate) can lead to buckled or wrinkled morphologies, which has many important applications in stretchable electronics, anticounterfeit technology, and high-precision micrometrology and nano-metrology. A number of buckling-based metrology methods have been developed to quantify the residual stress and viscoelastic properties of polymer thin films. However, in some systems (e.g., solvent-induced swelling or thermal strain), the compressive strain is unknown or difficult to measure. We present a quantitative method of measuring the compressive strain of wrinkled polymer films and coatings with knowledge of the skin thickness, wrinkle wavelength, and wrinkle amplitude. The derived analytical expression is validated with a well-studied model system, e.g., a stiff, thin film bonded to a thick, compliant substrate. After validation, we use our expression to quantify the applied swelling strain of previously reported wrinkled poly(styrene-alt-maleic anhydride) brush surfaces. Finally, the applied strain is used to rationalize the observed persistence length of aligned wrinkles created during atomic force microscopy lithography and subsequent solvent exposure.

Modified silica-based isoprene rubber (IR) composite has been designed and prepared by using a multi-functional silane, 2-aminoethyl 2-(3-triethoxysilylpropyl)aminoethyl disulfide (ATD), as coupling agent. Such modification significantly improved the dispersity of silica in the corresponding composites, as verified by SEM observation. And the hardness, tensile strength, stress at definite elongation, tear strength and temperature rise as well as the value of dynamic loss coefficient ranging from 0 degrees C to 80 degrees C of silica/IR vulcanized composites, are significantly improved, especially with low ATD dosage (2-4 phr). Isotactic polypropylene (PP) is one of the most popular plastics. However, the remarkably low surface energy of PP prevents the surface functionalization of PP. We studied the surface functionalization of PP by dip-coating with a maleic anhydride-grafted chlorinated polypropylene (MPO)/methacrylate-based terpolymer mixture. A methacrylate-based terpolymer (PMFP) was synthesized, which contained perfluoroalkyl (R-f)-conjugated monomers and poly(ethylene glycol)-conjugated monomers. Tape-peeling tests revealed that MPO successfully immobilized PMFP on a PP surface, although PMFP was less adhesive to PP. X-ray photoelectron spectroscopy (XPS), contact angle, and protein adsorption measurements revealed that the R-f groups and PEG chains in PMFP were segregated to the outermost surface of the dip-coated layer. The surface segregation of these moieties produced a low-fouling surface on the PP substrate. In addition, we synthesized a terpolymer that contained R-f groups and PEG chains with carboxy groups at their termini (PMFB) and used it to dip-coat a PP substrate. The surface segregation of side chains in PMFB induced the presentation of carboxy groups at the outermost surface, which were used as reactive sites for enzyme immobilization.

PURPOSE This report describes outcomes of an ongoing quality-improvement project (VitalSign6) in a large US metropolitan area to improve recognition, treatment, and outcomes of depressed patients in 16 primary care clinics (6 charity clinics, 6 federally qualified health care centers, 2 private clinics serving low-income populations, and 2 private clinics serving patients with either Medicare or private insurance). METHODS Inclusion in this retrospective analysis was restricted to the first 25,000 patients (aged >= 12 years) screened with the 2-item Patient Health Questionnaire (PHQ-2) in the aforementioned quality-improvement project. Further evaluations with self-reports and clinician assessments were recorded for those with positive screen (PHQ-2 >2). Data collected from August 2014 though November 2016 were available at 3 levels: (1) initial PHQ-2 (n = 25,000), (2) positive screen (n = 4,325), and (3) clinician-diagnosed depressive disorder with 18 or more weeks of enrollment (n = 2,160). RESULTS Overall, 17.3% (4,325/25,000) of patients screened positive for depression. Of positive screens, 56.1% (2,426/4,325) had clinician-diagnosed depressive disorder. Of those enrolled for 18 or more weeks, 64.8% were started on measurement-based pharmacotherapy and 8.9% referred externally. Of the 1,400 patients started on pharmacotherapy, 45.5%, 30.2%, 12.6%, and 11.6% had 0, 1, 2, and 3 or more follow-up visits, respectively. Remission rates were 20.3% (86/423), 31.6% (56/177), and 41.7% (68/163) for those with 1, 2, and 3 or more follow-up visits, respectively. Baseline characteristics associated with higher attrition were: non-white, positive drug-abuse screen, lower depression/anxiety symptom severity, and younger age. CONCLUSION Although remission rates are high in those with 3 or more followup visits after routine screening and treatment of depression, attrition from care is a significant issue adversely affecting outcomes.

We fabricated pH and redox dual-activatable self-assembled nanotheranostics (named as DA-SNs) via coordination-driven self-assembly of chlorin e6 (Ce6) disulfide-linked pH sensitive polymer ligand, poly (isobutylene-alt-maleic anhydride-graft-methoxy-poly (ethyleneglycol)-graft-imidazole-graft-Cystamine-Ce6) [PIMA-mPEG-API-SS-Ce6], and gadolinium ions (Gd3+). DA-SNs exhibited uniform particle size of similar to 48 nm, excellent stability, and inherent biosafety. Negatively charged DA-SNs could prolong blood circulation time (t(1/2) = 2.91 h) and improve tumor accumulation. Moreover, DA-SNs could undergo surface charge switch from negative charge to positive one in a slightly acidic tumor extracellular environment (pH 6.8), thus enhancing cellular uptake. After entering tumor cells, fluorescence, photodynamic therapeutic activity, and T1MR contrast from DA-SNs could be activated within this intracellular environment with lowered pH and high level of GSH. Importantly, human tumors implanted in mice could be successfully visualized via distinct pH and redox dual-sensitive T1MR contrast and fluorescence imaging, indicating that DA-SNs could serve as a dual-modal MR/fluorescence imaging probe for tumor-targeting diagnosis. In addition, DA-SNs exhibited superior photodynamic therapeutic efficiency with negligible side effects. Therefore, this DA-SN shows great promise for synergistic photodynamic therapy and diagnostic imaging.

Stimulus-responsive materials have great potential in advanced controllable oil/water separation applications. Here, a novel, cost-effective, and green approach is developed to produce a pH-responsive smart fabric with switchable wettability. The approach first involves grafting polydopamine (PDA) and cystamine dihydrochloride (cystamine) on a fabric surface to obtain thiol-functionalized fabric (Fabric-SH). Hydrophobic stearyl methacrylate (SMA) and pH-responsive undecylenic acid are then decorated on the Fabric-SH surface through efficient and green photoinduced thiol-ene click coupling chemistry. The obtained fabric exhibits rapidly switchable wettability between superhydrophobicity and superhydrophilicity depending on the contacting liquid pH value and can be applied in controllable separation of various mixtures of water and oil with high efficiency up to 99%. More importantly, the as prepared fabric is able to realize the separation of oil/water/oil ternary mixtures and can self-clean and repel oil fouling during the separation process. Its superhydrophobicity is robust, showing no significant change after a 500 cycle peeling test. This novel and cost-effective smart cotton fabric exhibits significant potential in satisfying different separation purposes under complicated conditions.
