The application of combination immune-chemotherapy makes up for the limitation of monotherapy and achieves superior antitumor activity against cancer. However, combinational therapy is always restricted by poor tumor targeted drug delivery efficacy. Herein, novel T cell membrane cloaking tumor microenvi-ronment-responsive nanoparticles (PBA modified T cell membrane cloaking hyaluronic acid (HA)-disulfide bond-vitamin E succinate/curcumin, shortened as RCM@T) were developed. T cell membrane cloaking not only serves as a protection shell for sufficient drug delivery but also acts as a programmed cell death-1(PD-1) antibody to selectively bind the PD-Li of tumor cells. When RCM@T is intravenously administrated into the blood stream, it accumulates at tumor sites and responds to an acidic pH to achieve a ‘membrane escape effect and expose the HA residues of RCM for tumor targeted drug delivery. RCM accumulates in the cytoplasm via CD44 receptor mediated endocytosis and intracellularly releases antitumor drug in the intracellular redox microenvironment for tumor chemotherapy. T cell membrane debris targets the PD-Llof tumor cells for tumor immunotherapy, which not only directly kills tumor cells, but also improves the CD8(+) T cell level and facilitates effector cytokine release. Taken together, the as-constructed RCM@T creates a new way for the rational design of a drug delivery system via the combination of stimuli-responsive drug release, chemotherapeutical agent delivery and cell membrane based immune checkpoint blockade immunotherapy.

Injectable hydrogels have been widely applied in the field of regenerative medicine. However, current techniques for injectable hydrogels are facing a challenge when trying to generate a biomimetic, porous architecture that is well-acknowledged to facilitate cell behaviors. In this study, an injectable, interconnected, porous hyaluronic acid (HA) hydrogel based on an in-situ bubble self-generation and entrapment process was developed. Through an amide reaction between HA and cystamine dihydrochloride activated by EDC/NHS, CO2 bubbles were generated and were subsequently entrapped inside the substrate due to a rapid gelation-induced retention effect. HA hydrogels with different molecular weights and concentrations were prepared and the effects of the hydrogel precursor solution’s concentration and viscosity on the properties of hydrogels were investigated. The results showed that HA10-10 (10 wt.%, MW 100,000 Da) and HA20-2.5 (2.5 wt.%, MW 200,000 Da) exhibited desirable gelation and obvious porous structure. Moreover, HA10-10 represented a high elastic modulus (32 kPa). According to the further in vitro and in vivo studies, all the hydrogels prepared in this study show favorable biocompatibility for desirable cell behaviors and mild host response. Overall, such an in-situ hydrogel with a self-forming bubble and entrapment strategy is believed to provide a robust and versatile platform to engineer injectable hydrogels for a variety of applications in tissue engineering, regenerative medicine, and personalized therapeutics.

Background: Black patients have higher rates of hospitalization for acute heart failure than other race/ethnic groups. We sought to determine whether diuretic efficiency is associated with racial differences in risk for rehospitalization after acute heart failure. Methods: A post hoc analysis was performed on 721 subjects (age, 68 +/- 13 years; 22% black) enrolled in 3 acute heart failure clinical trials: ROSE-AHF (Renal Optimization Strategies Evaluation in Acute Heart Failure), DOSE-AHF (Diuretic Optimization Strategy Evaluation in Acute Decompensated Heart Failure), and CARRESS-HF (Cardiorenal Rescue Study in Acute Decompensated Heart Failure). Repeated-measures ANOVA was used to test for a racextime effect on measures of decongestion. Diuretic efficiency was calculated as net fluid balance per total furosemide equivalents. In a subset of subjects, Cox regression was used to examine the association between race and rehospitalization according to plasma renin activity (PRA). Results: Compared with nonblack patients, black patients were younger and more likely to have nonischemic heart failure. During the first 72 to 96 hours, there was greater fluid loss (P=0.001), decrease in NT-proBNP (N-terminal pro-B-type natriuretic peptide;P=0.002), and lower levels of PRA (P<0.0001) in black patients. Diuretic efficiency was higher in black than in nonblack patients (403 [interquartile range, 221-795] versus 325 [interquartile range, 154-698];P=0.014). However, adjustment for baseline PRA attenuated the association between black race and diuretic efficiency. Over a median follow-up of 68 (interquartile range, 56-177) days, there was an increased risk of all-cause and heart failure-specific rehospitalization in nonblack patients with increasing levels of PRA, while the risk of rehospitalization was relatively constant across levels of PRA in black patients. Conclusions: Higher diuretic efficiency in black patients with acute heart failure may be related to racial differences in activity of the renin-angiotensin-aldosterone system. A simple method to fabricate redox-responsive composite hydrogel films via incorporating chitosan (CS) micro spheres into carboxymethyl cellulose (CMC) hydrogel using a di-sulphide cross-linker cystamine dihydrochloride (CYS) have done in this paper. Dual drugs, an anticancer, 5-Fluorouracil (5-FU) and an antibiotic, Tetracycline Hydrochloride (TH) were used as model drugs. The prepared composite hydrogel films were characterized using FTIR, SEM, rheology measurement, mechanical and swelling tests. Our results showed that the well dispersed spherical CS microspheres endowed hydrogel film increased mechanical properties and swellability. In vitro drug release displayed an accelerated release under weak acidic or reducing media. The in vitro cytotoxicity and cell apoptosis studies showed the drug-loaded composite film caused enhanced inhibition to HepG2 cells in presence of glutathione (GSH). Moreover, the film has exhibited excellent antimicrobial efficacy against E.coli and Staphylococcus aureus. Therefore, our simple and effective dual drug-loaded system has the potential to be used for cancer therapy in the future. (C) 2019 Elsevier B.V. All rights reserved.

A facile method via grafting to approach was used to synthesize hybrid gold-dendrimer nanoparticles. To this end, gold nanoparticles (GNPs) were synthesized via Turkevich method and 5th-generation cystamine-cored poly (propylene imine) (PPI) dendrimer was synthesized by iterative Michael addition and hydrogenation reactions. To prepare hybrid nanoparticles, aqueous solution of dendrimer was poured into colloidal solution of GNPs to form gold-S interactions which resulted in hybrid gold-dendrimer nanoparticles. UV-VIS-NIR and Raman spectroscopies, dynamic light scattering (DLS), thermogravimetric analysis (TGA) and field-emission scanning electron microscopy (FE-SEM) were utilized to confirm the surface modification of GNPs by PPI dendrimer. Cytotoxicity study through MIT assay against human fibroblast (FBS) cells showed appropriate proliferation of cells in presence of hybrid nanoparticles whereas higher grafting ratio of dendrimers induced more toxicity due to existence of peripheral amine groups. Also, hybrid gold-dendrimer nanoparticles were used as DOX nano-carriers. Results showed that carriers did not release the drug at pH = 7.4 significantly while up to 92.8% of drug release was measured at pH = 5.3. Also, higher grafting ratio limited the drug release due to shielding effect of grafted dendrimers.

Molecular machines are ubiquitous in nature and function away from equilibrium by consuming fuels to produce appropriate work. 5-Carboxylcytosine (5caC) plays a vital role in the dynamics of DNA demethylation, and sequencing of its sites will help us dig out more biological functions of 5caC. Herein, we present a novel chemical method to efficiently label 5caC distinguished from other bases in DNA. Combined with bisulfite sequencing, 5caC sites can be located at single-base resolution, and the efficiency of 5caC labeling is 92% based on the Sanger sequencing data. Furthermore, dot blot assays have confirmed that 5caC-containing DNA isolated from HeLa cells was successfully labeled using our method. We expect that our strategy can be further applied to selectively tagging other carboxyl-modified bases and mapping their sites in RNA.

A water-soluble cyclophane dimer having two disulfide groups as a reduction-responsive cleavable bond as well as several acidic and basic functional groups as a pH-responsive ionizable group 1 was successfully synthesized. It was found that 1 showed pH-dependent guest-binding behavior. That is, 1 strongly bound an anionic guest, 6-p-toluidinonaphthalene-2-sulfonate (TNS) with binding constant (K/M-1) for 1:1 host-guest complexes of 9.6 x 10(4) M-1 at pH 3.8, which was larger than those at pH 7.4 and 10.7 (6.0 x 10(4) and 2.4 x 10(4) M-1, respectively), indicating a favorable electrostatic interaction between anionic guest and net cationic 1. What is more, release of the entrapped guest molecules by 1 was easily controlled by pH stimulus. Large favorable enthalpies (Delta H) for formation of host-guest complexes were obtained under the pH conditions employed, suggesting that electrostatic interaction between anionic TNS and 1 was the most important driving force for host-guest complexation. Such contributions of Delta H for formation of host-guest complexes decreased along with increased pH values from acidic to basic solutions. Upon addition of dithiothreitol (DTT) as a reducing reagent to an aqueous PBS buffer (pH 7.4) containing 1 and TNS, the fluorescence intensity originating from the bound guest molecules decreased gradually. A treatment of 1 with DTT gave 2, having less guest-binding affinity by the cleavage of disulfide bonds of 1. Consequently, almost all entrapped guest molecules by 1 were released from the host. Moreover, such reduction-responsive cleavage of 1 and release of bound guest molecules was performed more rapidly in aqueous buffer at pH 10.7.

Chemically crosslinked recyclable photodeformable azobenzene (azo) polymer actuators with good stability (toward organic solvents and higher temperatures) and high processability and reconstruction ability hold great promise in many applications, but their development remains a challenging task. Herein, we report on for the first time a facile and highly efficient post-crosslinking method for addressing this issue. It involves first the synthesis of side-chain polymers bearing N-hydroxysuccinimide (NHS) carboxylate-substituted azo mesogens, fabrication of uniaxially oriented fibers from these azo polymers by the simple melt spinning method, and their subsequent post-crosslinking with cystamine (a diamine containing a disulfide bond) under mild conditions. The resulting chemically crosslinked fibers not only showed rapid and reversible photoinduced bending and unbending at ambient temperature as well as high mechanical strength and good solvent/heating stability, but also could be easily recycled into processable azo polymers in the presence of a reducing agent that can cleave the disulfide bond into thiol groups (i.e., tributylphosphine). In particular, the occurrence of the post-crosslinking reaction only on the thin surface layers of the azo polymer fibers afforded recycled polymers with large amounts of NHS carboxylate-substituted azo mesogens (together with a small amount of oxygen/heating-sensitive thiolsubstituted ones) in the first several (at least 5) recycling processes, thus allowing highly efficient reconstruction of photodeformable fibers with excellent photomobile properties by applying melt spinning and post-cross-linking (by using cystamine) methods. The strategy presented here opens the new possibility to the facile and efficient development of various advanced chemically crosslinked recyclable photodriven actuators.

Development of spatially restricted liquid-liquid phase separation (LLPS) systems emulating biomolecular condensates is a major challenge in coacervating materials science, due to lack of approaches for spatially restricted coacervation of sequence-defined zwitterionic segments resembling intrinsically disordered proteins. Herein, we present interfacial LLPS-driven polymerization-induced electrostatic self-assembly, namely, interfacial LLPS-PIESA. The asymmetric charge sequence patterning of zwitterionic growing segments is achieved via spontaneous polymerization of ion-pair monomers within a nanopartide core-shell interface. We show that charge sequence can profoundly affect the self-coacervation, leading to droplet dispersions, dense coacervates, and free-standing hydrogels. Moreover, the interfacial LLPS-PIESA shows a programmed hierarchical condensation self-assembly mechanism involving vesicles-to-lamellae transition, interfacial self-coacervation, lamellae-to-sheets transition, layer-by-layer sheet self-assembly, spatially restricted condensation and agglomeration, and redispersing into fibril network condensates under dynamic evolving surface charge regulation. The spatially restricted asymmetric charge sequence patterning, interfacial self-coacervation, and programmed hierarchical condensation self-assembly, all these elements can serve as the primary principles for the asymmetric charge sequence patterning design of hierarchically nanostructured condensates that emulate cellular biomolecular condensates.
