Cancer cells have reductive and acidic environments as compared to the normal body cells. Development of reductive/acidic responsive polymersomes will play a major role in cancer therapy to trigger the release of the loaded drug. In our work, we synthesized three different reductive/acidic dual-responsive polymers, poly[(mPEG-SS-amino) (N,N-diisopropylethylenediamino)phosphazenes] (PPDPs) in different mole ratios of side groups. These PPDPs were characterized by H-1 NMR, P-31 NMR, FT-IR and GPC. After that, the PPDPs were allowed to self-assemble into drug-loaded polymersomes with high loading content and encapsulation efficiency of hydrophilic/hydrophobic anticancer drugs. The hydrophilic anticancer drug doxorubicin hydrochloride (DOX center dot HCl) and hydrophobic drug doxorubicin were used. These PPDPs-based polymersomes showed reductive/acidic stimuli-responsive release of anticancer drugs. Moreover, these polymersomes also exhibited suitable hydrodynamic diameters, which will facilitate the longtime circulation in bloodstream due to avoiding renal clearance and close contact to the tumor cells in vascular sections due to enhanced permeability and retention effect. Collectively, these developed polymersomes may provide an effective platform for anticancer drugs delivery.

Natural hydrogels are widely investigated for biomedical applications because of their structures similar to extracellular matrix of native tissues, possessing excellent biocompatibility and biodegradability. However, they are often susceptible to mechanical disruption. In this study, self-healing hyaluronic acid (HA) hydrogels are fabricated through a facile dynamic covalent Schiff base reaction. Dialdehyde-modified HA (AHA) precursor was synthesized, and then the AHA/cystamine dihydrochloride (AHA/Cys) hydrogels were formed by blending AHA and Cys at acidic pH levels. By varying Cys to AHA ratio, the hydrogel morphology, swelling and kinetics of gelation could be controlled. Gelation occurred fast, which was predominantly attributed to Schiff base reaction between the dialdehyde groups on AHA and amimo groups on Cys. The hydrogel exhibited improved mechanical properties with increase in Cys content. Furthermore, due to dynamic imine bonds, this hydrogel demonstrated excellent self-healing ability based on the stress after mechanical disruption. Also, it was found to be pH-responsive and injectable. Taken together, this kind of hyaluronic acid hydrogel can provide promising future for various biomedical applications in drug delivery, bioprinting, smart robots and tissue regeneration.

The application of combination immune-chemotherapy makes up for the limitation of monotherapy and achieves superior antitumor activity against cancer. However, combinational therapy is always restricted by poor tumor targeted drug delivery efficacy. Herein, novel T cell membrane cloaking tumor microenvi-ronment-responsive nanoparticles (PBA modified T cell membrane cloaking hyaluronic acid (HA)-disulfide bond-vitamin E succinate/curcumin, shortened as RCM@T) were developed. T cell membrane cloaking not only serves as a protection shell for sufficient drug delivery but also acts as a programmed cell death-1(PD-1) antibody to selectively bind the PD-Li of tumor cells. When RCM@T is intravenously administrated into the blood stream, it accumulates at tumor sites and responds to an acidic pH to achieve a ‘membrane escape effect and expose the HA residues of RCM for tumor targeted drug delivery. RCM accumulates in the cytoplasm via CD44 receptor mediated endocytosis and intracellularly releases antitumor drug in the intracellular redox microenvironment for tumor chemotherapy. T cell membrane debris targets the PD-Llof tumor cells for tumor immunotherapy, which not only directly kills tumor cells, but also improves the CD8(+) T cell level and facilitates effector cytokine release. Taken together, the as-constructed RCM@T creates a new way for the rational design of a drug delivery system via the combination of stimuli-responsive drug release, chemotherapeutical agent delivery and cell membrane based immune checkpoint blockade immunotherapy.

Crosslinked polymer nanomaterials have attracted great attention due to their stability and highly controllable drug delivery; herein, a series of well-defined amphiphilic PDPA-b-P(NMS-co-OEG) diblock terpolymers (P1-P3) were designed and prepared via RAFT polymerization and were self-assembled into non-cross-linked (NCL) nanomicelles, which were further prepared into shell-cross-linked (SCL) micelles via cystamine-based in situ shell cross-linking. Using P3 as an optimized polymer, SCL-P3 micelles were prepared, which demonstrated remarkable pH/redox-dual responsive behaviour. For drug delivery, camptothecin (CPT)-loaded SCL-P3 micelles were prepared and showed much higher CPT-loading capability than their NCL-P3 counterparts. Notably, the SCL-P3 micelles showed good synergistic pH/redox-dual responsive CPT release properties, making them potential smart nanocarriers for drug delivery.

Introducing clickable groups onto polymer microspheres allows the microspheres to be facilely functionalized via a click reaction. In this report, we have synthesized monodisperse PMMA microspheres with clickable double bonds on the surface by one-step RAFT-assisted dispersion photopolymerization using a cyclohexyl-containing macro-RAFT agent as a stabilizer. This macro-RAFT agent, named P(THPAA-b-AA)-TTC, was synthesized by RAFT-mediated block copolymerization of a cyclohexyl-containing monomer (THPAA, obtained by esterification of cis-1,2,3,6-tetrahydrophthalic with 2-hydroxyethyl acrylate) and acrylic acid. The obtained microspheres were analyzed by H-1 NMR and Iodometric method, indicating that the content of carbon double bonds on the microspheres increased with increasing the amount of P(THPAA-b-AA)-TTC in the polymerization feed. We then used a thiolated biotin to perform the photo-click reaction with the cyclohexene double bond on the micro spheres, thereby attaching the biotin unit onto the microspheres. The biotinylated PMMA microspheres were found to be able to effectively immobilize fluorescein isothiocyanate streptavidin (FITC-SAv) via the specific adsorption of biotin to streptavidin, and thus exhibited clear fluorescence under confocal microscope. This study provides a convenient and feasible way to prepare surface-functionalized microspheres, which has great potential in biological applications as well as other fields.

The aim of this work was to obtain a novel onco-theranostic system for early diagnosis and improved chemotherapeutic outcome. Hybrid nanogels with magnetic and dual responsive properties were fabricated by covalently attaching superparamagnetic iron oxide nanoparticles (SPIONs) with a disulfide-modified alginate derivative, while simultaneously encapsulating the anticancer drug doxorubicin. BACKGROUND Decontamination of water by means of photocatalysis is seen as a sustainable solution. However, catalysts with low recovery from liquid suspension remain the main predicament that obstructs their industrial applications. To overcome this shortcoming, a methodology is reported here for the functionalization of nanospinel nickel zinc ferrite by cystamine ligand to develop Ni0.5Zn0.5Fe2O4@cystamine magnetic recyclable catalyst for water decontamination applications. RESULTS The diffraction patterns of bare and functionalized nanoferrites indicated that the cubic inverse spinel structure of the particles was not changed after cystamine functionalization. X-ray photoelectron spectroscopy (XPS) and Raman spectroscopy analyses confirmed the chemical bonding between cystamine and spinel ferrites. The spherical morphology and elemental composition were studied through high-resolution transmission electron spectroscopy (HRTEM) images and energy-dispersive X-ray (EDX) analysis respectively. Deeper understanding of the catalyst’s optical behavior and calculation of the energy band gap were achieved by means of photoluminescence spectroscopy. Magnetic saturation values were calculated from vibrating sample magnetometer (VSM) analysis, and almost negligible change was found. A possible photocatalytic mechanism for the removal of organic dye is proposed. CONCLUSION The present findings imply that the presence of amine groups on the catalyst surface, good dispersibility stability and outstanding catalytic activity make functionalized nanoferrite a suitable candidate for industrial wastewater treatment. Easy synthesis, easy work-up and short reaction times are some of the advantages of the methodology. (c) 2019 Society of Chemical Industry
