A dumbbell-shaped copolymer (PDMAEMA)(4)-PCL-SS-PCL-(PDMAEMA)(4) with a lower critical solution temperature (LCST) and redox responses was first synthesized by the combination of ring opening polymerization (ROP), click chemistry and atom transfer radical polymerization (ATRP). Then a dumbbell-shaped copolymer (PDMAPS)(4)-PCL-SS-PCL-(PDMAPS)(4) with an upper critical solution temperature (UCST) and redox responses was obtained through the quaternization reaction between DMAEMA and excess 1,3-propane sultone. Both (PDMAEMA)(4)-PCL-SS-PCL-(PDMAEMA)(4) and (PDMAPS)(4)-PCL-SS-PCL-(PDMAPS)(4) were amphiphilic, and they could self-assemble into spherical micelles under certain conditions. Benefitting from the LCST-type thermoresponse of PDMAEMA and UCST-type thermoresponse of PDMAPS, both dumbbell-shaped copolymers (PDMAEMA)(4)-PCL-SS-PCL-(PDMAEMA)(4) and (PDMAPS)(4)-PCL-SS-PCL-(PDMAPS)(4) presented temperature-responsive properties. The morphology and size of their assemblies would be changed when the temperature is altered. Meanwhile, due to the redox-response of disulfide bonds, both (PDMAEMA)(4)-PCL-SS-PCL-(PDMAEMA)(4) and (PDMAPS)(4)-PCL-SS-PCL-(PDMAPS)(4) showed redox-responsive properties. When dl-dithiothreitol (DTT) was added into the systems, disulfide bonds were broken, causing the change of (PDMAEMA)(4)-PCL-SS-PCL-(PDMAEMA)(4) to PCL-(PDMAEMA)(4), while (PDMAPS)(4)-PCL-SS-PCL-(PDMAPS)(4) changed into PCL-(PDMAPS)(4). The distributions of R-h for both (PDMAEMA)(4)-PCL-SS-PCL-(PDMAEMA)(4) and (PDMAPS)(4)-PCL-SS-PCL-(PDMAPS)(4) assemblies broadened with asymmetric peaks, giving rise to curves with double peaks. After 24 h, DLS curves became symmetric with a single peak, and the R-h was about half that of the original assemblies. In the meantime, dispersed spherical micelles could be observed by TEM.

The quantification of proteins and peptides becomes more important besides mere identification in modern life sciences. Therefore, we have developed a new reagent that adds to the known metall-coded affinity tagging strategy employed in molecular and elemental mass spectrometry containing a photocleavable linker. A synthesis route was developed that provides the new reagent in good yields. The stability of the synthesized reagents was assessed under different temperature and illumination conditions. Labeling reactions were carried out at peptide and protein level, while also the fragmentation behavior of labeled peptides was assessed. In additional experiments, the photocleavability of the new reagent was examined. Upon irradiation with ultraviolet light, the photoproducts were liberated and could be used for quantification of labeled peptides.

Active targeting and overcoming multi-drug resistance (MDR) can be some of the important attributes of targeted therapy for metastatic breast cancer (MBC) and triple-negative breast cancer (TNBC) treatment. In this study, we constructed a hyaluronic acid (HA)-decorated mixed nanomicelles-encapsulating chemotherapeutic agent paclitaxel (PTX) and P-glycoprotein inhibitor ritonavir (RTV). HA was conjugated to poly (lactide) co-(glycolide) (PLGA) polymer by disulfide bonds (HA-ss-PLGA). HA is a natural ligand for CD44 receptors overexpressed in breast cancer cells. Disulfide bonds undergo rapid reduction in the presence of glutathione, present in breast cancer cells. The addition of RTV can inhibit the P-gp and CYP3A4-mediated metabolism of PTX, thus aiding in reversing MDR and sensitizing the cells toward PTX. An in vitro uptake and cytotoxicity study in MBC MCF-7 and TNBC MDA-MB-231 cell lines demonstrated the effective uptake of the nanomicelles and drug PTX compared to non-neoplastic breast epithelium MCF-12A cells. Interestingly, in vitro potency determination showed a reduction in mitochondrial membrane potential and reactive oxygen species in breast cancer cell lines, indicating effective apoptosis of cancer cells. Thus, stimuli-sensitive nanomicelles along with HA targeting and RTV addition can effectively serve as a chemotherapeutic drug delivery agent for MBC and TNBC.

Molecular machines are ubiquitous in nature and function away from equilibrium by consuming fuels to produce appropriate work. Chemists have recently excelled at mimicking the fantastic job performed by natural molecular machines with synthetic systems soluble in organic solvents. In efforts toward analogous systems working in water, we show that guest molecules can be exchanged in the synthetic macrocycle cucurbit[7]uril by involving kinetic traps, and in such a way as modulating energy wells and kinetic barriers using pH, light, and redox stimuli. Ditolyl-viologen can also be exchanged using the best kinetic trap and interfaced with alginate, thus affording pH-responsive blue, fluorescent hydrogels. With tunable rate and binding constants toward relevant guests, cucurbiturils may become excellent ring molecules for the construction of advanced molecular machines working in water.

The reduced graphene oxide (rGO) modified natural rubber composite (NR) filled with high contents of silica was prepared by a wet compounding and latex mixing process using a novel interface modifier cystamine dihydrochloride (CDHC) with coagulation ability. CDHC acts as a coagulation agent through electrostatic interaction with rGO, SiO2, and latex rubber particles during the latex-based preparation process, while in the obtained silica/graphene/natural rubber composites, CDHC acts as an interface modifier. Compared with the composites prepared by the conventional mechanical mixing method, the dispersion of both rGO and SiO2 in the composites made by a wet compounding and latex mixing process is improved. As a result, the obtained silica/graphene/natural rubber composite prepared by this new method has good comprehensive properties. A Dynamic Mechanical Test suggests that the tan delta values of the composites at 60 degrees C decrease, indicating a low rolling resistance with increasing the graphene content at a low strain, but it increases at a higher strain. This unique feature for this material provides an advantage in the rubber tire application.

Recent advances in the development of protein-based vaccines have expanded the opportunities for preventing and treating both infectious diseases as well as cancer. However, the development of readily and efficient antigen delivery systems capable of stimulating strong cytotoxic T-lymphocyte (CTL) responses remains a challenge. With the attempt to closely mimic the properties of viruses in terms of their size and molecular organization, we constructed RNA (which is a ligand for Toll-like receptor 7 (TLR7) and TLR8) and antigen-loaded nanoparticles resembling the structural organization of viruses. Cationic polymers containing either azide or bicyclo[6.1.0]nonyne (BCN) groups were synthesized as electrostatic glue that binds negatively charged single stranded RNA (PolyU) to form a self-crosslinked polyplex core. An azide-modified model antigen (ovalbumin, OVA) and a BCN-modified mannosylated or galactosylated polymer were sequentially conjugated to the RNA core via disulfide bonds using copper free click chemistry to form the shell of the polyplexes. The generated reducible virus mimicking particles (VMPs) with a diameter of 200 nm and negatively surface charge (-14 mV) were colloidally stable in physiological conditions. The immunogenicity of these VMP vaccines was evaluated both in vitro and in vivo. The surface mannosylated VMPs (VMP-Man) showed 5 times higher cellular uptake by bone marrow derived DCs (BMDCs) compared to galactosylated VMP (VMP-Gal) counterpart. Moreover, VMP-Man efficiently activated DCs and greatly facilitated MHC I Ag presentation in vitro. Vaccination of mice with VMP-Man elicited strong OVA-specific CTL responses as well as humoral immune responses. These results demonstrate that the modular core-shell polymeric nanoparticles described in this paper are superior in inducing strong and durable immune responses compared to adjuvanted protein subunit vaccines and offer therefore a flexible platform for personalized vaccines.

Among the numerous homogeneous electrochemical CO2 reduction catalysts, [Ni(cyclam)](2+) is known as one of the most potent catalysts. Likewise, [Ni(isocyclam)](2+) was reported to enable electrochemical CO2 conversion but has received significantly less attention. However, for both catalysts, a purposeful substitution of a single nitrogen donor group by chalcogen atoms was never reported. In this work, we report a series of isocyclam-based Ni complexes with {ON3}, {SN3}, {SeN3}, and {N-4} moieties and investigated the influence of nitrogen/chalcogen substitution on electrochemical CO2 reduction. While [Ni(isocyclam)](2+) showed the highest selectivity toward CO2 reduction within this series with a Faradaic efficiency of 86% for the generation of CO at an overpotential of -1.20 V and acts as a homogeneous catalyst, the O- and S-containing Ni complexes revealed comparable catalytic activities at ca. 0.3 V milder overpotential but tend to form deposits on the electrode, acting as precursors for a heterogeneous catalysis. Moreover, the heterogeneous species generated from the O- and S-containing complexes enable a catalytic hydride transfer to acetonitrile, resulting in the generation of acetaldehyde. The incorporation of selenium, however, resulted in loss of CO2 reduction activity, mainly leading to hydrogen generation that is also catalyzed by a heterogeneous electrodeposit.

Herein, we report a cascaded multi-responsive self-assembled nanoprobe, which enables sequential redox-triggered and near-infrared (NIR) irradiation-induced F-19 MR signal activation/amplification for sensing and imaging. Specifically, we designed and synthesized a cascaded multiresponsive F-19-bearing nanoprobe based on the self-assembly of amphiphilic redox-responsive F-19-containing polymers and NIR-absorbing indocyanine green (ICG) molecules. It could realize the activation of F-19 signals in the reducing tumor microenvironment and subsequent signal amplification via the photothermal process. This stepwise two-stage activation/amplification of F-19 signals was validated by F-19 NMR and MRI both in vitro and in vivo. The multiresponsive F-19 nanoprobes capable of cascaded F-19 signal activation/amplification and photothermal effect exertion can provide accurate sensing and imaging of tumors.
