Glutathione (GSH) plays a vital role in maintaining biological redox homeostasis. Accordingly, accurate imaging of glutathione in vivo is of great significance. Herein, we propose a magnetic resonance energy transfer (MRET) strategy based on a distance-dependent magnetic exchange coupling effect (MECE), which can realize GSH detection within tumors in vivo by susceptibility weighted imaging (SWI). Fe3O4 nanoparticles (NPs) and CoFe2O4 NPs linked with cystamine (Fe3O4-S-S-CoFe2O4) have been successfully designed as SWI nanoprobes. After the disulfide bonds are broken by excess GSH in the tumor, the increase in the distance between Fe3O4 NPs and CoFe2O4 NPs will induce a decrease of MECE and magnetic susceptibility. As a result, the changes in the SWI signals are used for tumor GSH detection in vivo. Experimental results in vitro and in vivo demonstrate that the Fe3O4-S-S-CoFe2O4 SWI nanoprobe can sensitively detect concentrations of GSH in tumors. Hence, this strategy not only improves the sensitivity of the GSH response in SWI but also provides a powerful basis for the design of other responsive functional MRI nanoprobes.

Multidrug resistance (MDR) of cancer cells is a significant challenge in chemotherapy, highlighting the urgent medical need for simple and reproducible strategies to reverse this process. Here, we report the development of an active tumor-targeting and redox-responsive nanoplatform (PA-ss-NP) using hyaluronic acid-g-cystamine dihydrochloride-poly-epsilon-(benzyloxycarbonyl)-L-lysine (HA-ss-PLLZ) to co-deliver paclitaxel (PTX) and apatinib (APA) for effective reversal of MDR. This smart nanoplatform specifically bound to CD44 receptors, leading to selective accumulation at the tumor site and uptake by MCF-7/ADR cells. Under high concentrations of cellular glutathione (GSH), the nanocarrier was degraded rapidly with complete release of its encapsulated drugs. Released APA effectively inhibited the function of the P-glycoprotein (P-gp) drug pump and improved the sensitivity of MDR cells to chemotherapeutic agents, leading to the recovery of PTX chemosensitivity in MDR cells. As expected, this newly developed intelligent drug delivery system could effectively control MDR, both in vitro and in vivo.

Therapeutic delivery selectively to lymph nodes has the potential to address a variety of unmet clinical needs. However, owing to the unique structure of the lymphatics and the size-restrictive nature of the lymph node reticular network, delivering cargo to specific cells in the lymph node cortex and paracortex is difficult. Here, we describe a delivery system to overcome lymphatic and intra-lymph node transport barriers by combining nanoparticles that are rapidly conveyed to draining lymph nodes after administration in peripheral tissues with programmable degradable linkers. This platform enables the controlled release of intra-lymph-mobile small-molecular cargo, which can reach vastly more immune cells throughout the lymph node than either the particles or free compounds alone. The release rate can be programmed, allowing access to different lymph node structures and therefore specific lymphocyte subpopulations. We are thus able to alter the subtypes of drugged lymph node cells to improve immunotherapeutic effects. Nanoparticles that access lymphatic vessels and are functionalized with degradable linkers, whose half-lives can be programmed, enable the controlled release of therapeutic cargo in different regions of the lymph nodes, allowing the targeting of otherwise difficult-to-reach lymphocyte subpopulations.

Polymers of intrinsic microporosity (PIMs) are a class of microporous polymers with rigid and contorted molecular structures. The special structures lead to incomplete space occupation, and the pores of PIMs originate from the formed void. In this work, we made attempts to expand the applications of PIMs to the aspect of heavy metal removal. First, PIM-1 was synthesized using tetrafluoroterephthalonitrile (TFTPN) and 5,5′,6,6′-tetrahydroxy-3,3,3′,3′-tetramethyl-1,1′-spirobisindane (TTSBI) as precursors. The primary PIM-1 was quite hydrophobic, and thus could not be evenly dispersed in water, resulting in low adsorption capacity for mercury ions (Hg2+). A conversion of nitrile group in PIM-1 to thiol group was then carried out by two steps, namely carboxylation and introduction of thiol groups. The carboxylation made the polymers more active, and the final thiol-functionalization provided the polymers with hydrophilicity and affinity for Hg2+. The thiolethyl modified PIM-1 (assigned as PIM-G) and thiophenyl modified (assigned as PIM-B) possessed maximum Hg2+ adsorption capacity of 136 mg g(-1) and 127 mg g(-1) at pH 5, respectively. Besides, the thiol-functionalized PIMs had fluorescence property and showed potential in sensing for Hg2+.

We present a sensitive single step nanobiosensor for thyroid disease diagnosis. Nanoparticles modified screen printed carbon electrode catalyzes the formation of peptide bond between anti-TSH antibody and amino coated gold nanoparticles rendering a covalently coupled antibody. The nanobiosensor detects and quantifies the thyroid stimulating hormone in the sample by sensing the effective resistance offered by electrode. As the TSH concentration increases in serum sample, more is the immunocomplex formed and higher is the resistance offered by electrode. Gold nanoparticles functionalization with cystamine dihydrochloride facilitates a covalent bonding between surface amino group and carboxylic Fc ensuring maximizing available active antibody. This strategy ensures a much lower limit of detection in addition to improved detection range due to increased loading capacity as a result of larger effective surface area offered by gold nanoparticles. These two aspects of immunosensor fabrication resulted in limit of detection as low as 0.001 mu IU/mL and an enhanced detection range of 0.001-150 mu IU/mL. This makes the developed immunosensor suitable for diagnostic purpose covering the clinically relevant range and a simple detection technique makes it potential candidate for fabrication of a Point-of-Care device for detection of Thyroid dysfunctioning. A series of isocyanate-free polyurethanes (NIPUs), showing an intrinsic thermo-healing performance, have been synthesized from sustainable vegetable oil-based cyclic carbonates and bio-based amines. The bio-based contents of the NIPUs are higher than 78%. Their thermo-mechanical and self-healing performances are tailored simply by varying the feed ratios of the amines employed. The biomass linoleic acid dimer-based diamine plays a positive role in maintaining the higher thermal stability and good elasticity of the prepared NIPUs, the cycloaliphatic isophorone diamine contributes to high tensile strength, and cysteine-derived disulfide-containing cystamine (CA) enhances the self-healing efficiency. The combination of intrinsic hydrogen bonds existed in the NIPU matrix with the dynamic exchange reactions of disulfide bonds and the enhanced flexibility of NIPU networks results in 98.1% self-healing efficiency at 25 degrees C. The contributions of disulfide bonds and hydrogen bonds to the healing efficiency were estimated by adjusting the composition of NIPUs, showing that the contribution of disulfide bonds to the healing ability is about 45.3% even at a low amount of CA (6.0 wt%).

PURPOSE This report describes outcomes of an ongoing quality-improvement project (VitalSign6) in a large US metropolitan area to improve recognition, treatment, and outcomes of depressed patients in 16 primary care clinics (6 charity clinics, 6 federally qualified health care centers, 2 private clinics serving low-income populations, and 2 private clinics serving patients with either Medicare or private insurance). METHODS Inclusion in this retrospective analysis was restricted to the first 25,000 patients (aged >= 12 years) screened with the 2-item Patient Health Questionnaire (PHQ-2) in the aforementioned quality-improvement project. Further evaluations with self-reports and clinician assessments were recorded for those with positive screen (PHQ-2 >2). Data collected from August 2014 though November 2016 were available at 3 levels: (1) initial PHQ-2 (n = 25,000), (2) positive screen (n = 4,325), and (3) clinician-diagnosed depressive disorder with 18 or more weeks of enrollment (n = 2,160). RESULTS Overall, 17.3% (4,325/25,000) of patients screened positive for depression. Of positive screens, 56.1% (2,426/4,325) had clinician-diagnosed depressive disorder. Of those enrolled for 18 or more weeks, 64.8% were started on measurement-based pharmacotherapy and 8.9% referred externally. Of the 1,400 patients started on pharmacotherapy, 45.5%, 30.2%, 12.6%, and 11.6% had 0, 1, 2, and 3 or more follow-up visits, respectively. Remission rates were 20.3% (86/423), 31.6% (56/177), and 41.7% (68/163) for those with 1, 2, and 3 or more follow-up visits, respectively. Baseline characteristics associated with higher attrition were: non-white, positive drug-abuse screen, lower depression/anxiety symptom severity, and younger age. CONCLUSION Although remission rates are high in those with 3 or more followup visits after routine screening and treatment of depression, attrition from care is a significant issue adversely affecting outcomes.

Cancer cells have reductive and acidic environments as compared to the normal body cells. Development of reductive/acidic responsive polymersomes will play a major role in cancer therapy to trigger the release of the loaded drug. In our work, we synthesized three different reductive/acidic dual-responsive polymers, poly[(mPEG-SS-amino) (N,N-diisopropylethylenediamino)phosphazenes] (PPDPs) in different mole ratios of side groups. These PPDPs were characterized by H-1 NMR, P-31 NMR, FT-IR and GPC. After that, the PPDPs were allowed to self-assemble into drug-loaded polymersomes with high loading content and encapsulation efficiency of hydrophilic/hydrophobic anticancer drugs. The hydrophilic anticancer drug doxorubicin hydrochloride (DOX center dot HCl) and hydrophobic drug doxorubicin were used. These PPDPs-based polymersomes showed reductive/acidic stimuli-responsive release of anticancer drugs. Moreover, these polymersomes also exhibited suitable hydrodynamic diameters, which will facilitate the longtime circulation in bloodstream due to avoiding renal clearance and close contact to the tumor cells in vascular sections due to enhanced permeability and retention effect. BACKGROUND Decontamination of water by means of photocatalysis is seen as a sustainable solution. However, catalysts with low recovery from liquid suspension remain the main predicament that obstructs their industrial applications. To overcome this shortcoming, a methodology is reported here for the functionalization of nanospinel nickel zinc ferrite by cystamine ligand to develop Ni0.5Zn0.5Fe2O4@cystamine magnetic recyclable catalyst for water decontamination applications. RESULTS The diffraction patterns of bare and functionalized nanoferrites indicated that the cubic inverse spinel structure of the particles was not changed after cystamine functionalization. X-ray photoelectron spectroscopy (XPS) and Raman spectroscopy analyses confirmed the chemical bonding between cystamine and spinel ferrites. The spherical morphology and elemental composition were studied through high-resolution transmission electron spectroscopy (HRTEM) images and energy-dispersive X-ray (EDX) analysis respectively. Deeper understanding of the catalyst’s optical behavior and calculation of the energy band gap were achieved by means of photoluminescence spectroscopy. Magnetic saturation values were calculated from vibrating sample magnetometer (VSM) analysis, and almost negligible change was found. A possible photocatalytic mechanism for the removal of organic dye is proposed. CONCLUSION The present findings imply that the presence of amine groups on the catalyst surface, good dispersibility stability and outstanding catalytic activity make functionalized nanoferrite a suitable candidate for industrial wastewater treatment. Easy synthesis, easy work-up and short reaction times are some of the advantages of the methodology. (c) 2019 Society of Chemical Industry
