Nitrogen-doped MoS2 quantum dots (N-MoS2 QDs) were synthesized via a facile hydrothermal approach, and exhibited high fluorescence quantum yield (QY, 14.9%), excellent photostability, biocompatibility and water solubility. A novel method with good selectivity and sensitivity was established to assay hematin using N-MoS2 QDs as a fluorescent probe based on inner filter effect (IFE). Fluorescent quenching of N-MoS2 QDs has a fine linear dependence with the concentration of hematin in the range of 0.5-15 mu mol/L and a limit of detection of 0.32 mu mol/L (S/N = 3). By the detection method, average concentration of hematin in real health human erythrocytes was measured as 22.5 +/- 3.9 mu mol/L. And, recoveries range varied from 94 to 108% through standard recovery experiment. The N-MoS2 QDs probe shows excellent photostability, low cytotoxicity and anti-interference ability for hematin assay, which may become a promising method for the test of hematin in human blood.

Molecular probes featuring promising capabilities including specific targeting, high signal-to-noise ratio, and in situ visualization of deep tissues are in great demand for tumor diagnosis and therapy. F-19 magnetic resonance imaging (MRI) techniques incorporating stimuli-responsive probes are anticipated to be highly beneficial for specific detection and imaging of tumors because of negligible background and deep tissue penetration. Herein, we report a cascaded multi-responsive self-assembled nanoprobe, which enables sequential redox-triggered and near-infrared (NIR) irradiation-induced F-19 MR signal activation/amplification for sensing and imaging. Specifically, we designed and synthesized a cascaded multiresponsive F-19-bearing nanoprobe based on the self-assembly of amphiphilic redox-responsive F-19-containing polymers and NIR-absorbing indocyanine green (ICG) molecules. It could realize the activation of F-19 signals in the reducing tumor microenvironment and subsequent signal amplification via the photothermal process. This stepwise two-stage activation/amplification of F-19 signals was validated by F-19 NMR and MRI both in vitro and in vivo. The multiresponsive F-19 nanoprobes capable of cascaded F-19 signal activation/amplification and photothermal effect exertion can provide accurate sensing and imaging of tumors.

Objective: To investigate chest computed tomography (CT) findings associated with severe COVID-19 pneumonia in the early recovery period. Methods: We retrospectively analyzed the cases of patients diagnosed with severe COVID-19 pneumonia at a single center between January 12, 2020, and March 16, 2020. The twelve ICU patients studied had been diagnosed SARS-CoV-2 (COVID-19) nucleic acid positive. Patient clinical symptoms were relieved or disappeared, and basic clinical information and laboratory test results were collected. The study focused on the most recent CT imaging characteristics. Results: The average age of the 12 patients was 58.8 +/- 16.2 years. The most prevalent symptoms were fever (100%), dyspnea (100%), and cough (83.3%). All patients experienced acute respiratory distress syndrome (ARDS), of which 9 were moderate to severe. Six patients used noninvasive ventilators, and 4 patients used mechanical ventilation. One patient was treated with extracorporeal membrane oxygenation (ECMO). The lymphocyte count decreased to 0.67 +/- 0.3 (x 10 /L). The average day from illness onset to the last follow-up CT was 56.1 +/- 7.7 d. The CT results showed a decrease in ground glass opacities (GGO), whereas fibrosis gradually increased. The common CT features included GGO (10/12, 83.3%), subpleural line (10/12, 83.3%), fibrous stripes (12/12, 100%), and traction bronchiectasis (10/12, 83.3%). Eight patients (66.7%) showed predominant reticulation and interlobular thickening. Four patients (33.3%) showed predominant GGO. Lung segments involved were 174/216 (80.6%). Conclusions: Fibrous stripes and GGO are common CT signs in critically ill patients with COVID-19 pneumonia in the early recovery period. Signs of pulmonary fibrosis in survivors should be carefully monitored. (C) 2020 Elsevier Inc. All rights reserved.

In this work, two novel boronate affinity monolithic materials able to extract glycopeptides within a polypropylene syringe are described and compared. The first material was synthesized from glycidyl methacrylate (GMA)-based monoliths modified with poly-3-mercaptopropyl-methylsiloxane (PMPMS) followed by attachment of 4-vinylphenylboronic acid (VPBA) via thiol-ene click reaction. The second material was prepared by using gold nanoparticle (AuNP)-modified monoliths as substrate followed by subsequent attachment of PMPMS and VPBA. The resulting materials were used as sorbents for solid-phase extraction (SPE) to selectively preconcentrate glycopeptides from horseradish peroxidase (HRP) digests. The material that gave the superior performance was that prepared with AuNPs due to the presence of abundant boronic acid groups, being its practical applicability also examined. The hybrid material exhibited a satisfactory efficiency of glycopeptide enrichment (identifying 24 glycopeptides from a total of 27) in mixture of tryptic digests of HRP and bovine serum albumin (BSA) (1:100, w/w). The sorbent shows low sensitivity (0.5 fmol/?L), good adsorption capacity (25 mg g-1) and suitable reusability (over 10 times). Moreover, the hybrid monolith was successfully applied to the selective enrichment of glycopeptides from human serum digests, without any pretreatment, in which 85 glycopeptides were identified by nano-LC-MS/MS, suggesting a great potential for application in glycoproteome field.

Poly(ethylene glycol) (PEG) diacrylate copolymer networks containing pentafluorophenyl active esters can be quantitatively substituted with a wide variety of primary amines, enabling the development of a versatile synthetic platform for the preparation of polymer hydrogel and membrane libraries. By tuning the starting network, a high degree of control over cross-linking density, water uptake, and functional group incorporation can be reproducibly achieved, which is vital for elucidating structure-property relationships in ion transporting membranes. From the same cross-linked scaffold, a diversity of basic, acidic, and solute-chelating moieties were obtained through functionalization, which allows for tailored uptake of basic and acidic organic dyes and metal chloride salts. Ion permeation and sorption measurements for a series of polymer networks with controlled cross-linking density and varied imidazole grafting densities illustrate the ability of this platform to isolate the effect of chemical functionality on ion transport from the effects of cross-linking density and water content.

The efficient selectivity of heavy metal ions from wastewater is still challenging but gains great public attention in water treatment on a world scale. In this study, the novel disulfide cross-linked poly(methacrylic acid) iron oxide (Fe3O4@S-S/PMAA) nanoparticles with selective adsorption, improved adsorption capability, and economic reusability were designed and prepared for selective adsorption of Pb(II) ions in aqueous solution. In this study, nuclear magnetic resonance, dynamic light scattering, scanning electron microscopy, X-ray diffraction, vibrating sample magnetometry, and thermogravimetric analysis were utilized to study the chemophysical properties of Fe3O4@S-S/PMAA. The effect of different factors on adsorption properties of the Fe3O4@S-S/PMAA nanoparticles for Co(II) and Pb(II) ions in aqueous solution was explored by batch adsorption experiments. For adsorption mechanism investigation, the adsorption of Fe3O4@S-S/PMAA for Co(II) and Pb(II) ions can be better fitted by a pseudo-second-order model, and the adsorption process of Fe3O4@S-S/PMAA for Co(II) and Pb(II) matches well with the Freundlich isotherm equation. Notably, in the adsorption experiments, the Fe3O4@S-S/PMAA nanoparticles were demonstrated to have a maximum adsorption capacity of 48.7 mg.g(-1) on Pb(II) ions with a selective adsorption order of Pb2+ > Co2+ > Cd2+ > Ni2+ > Cu2+ > Zn2+ > K+ > Na+ > Mg2+ > Ca2+ in the selective experiments. In the regeneration experiments, the Fe3O4@S-S/PMAA nanoparticles could be easily recovered by desorbing heavy metal ions from the adsorbents with eluents and showed good adsorption capacity for Co(II) and Pb(II) after eight recycles. In brief, compared to other traditional nanoadsorbents, the as-prepared Fe3O4@S-S/PMAA with improved adsorption capability and high regeneration efficiency demonstrated remarkable affinity for adsorption of Pb(II) ions, which will provide a novel technical platform for selective removal of heavy metal ions from actual polluted water.

We present a sensitive single step nanobiosensor for thyroid disease diagnosis. Nanoparticles modified screen printed carbon electrode catalyzes the formation of peptide bond between anti-TSH antibody and amino coated gold nanoparticles rendering a covalently coupled antibody. The nanobiosensor detects and quantifies the thyroid stimulating hormone in the sample by sensing the effective resistance offered by electrode. As the TSH concentration increases in serum sample, more is the immunocomplex formed and higher is the resistance offered by electrode. Gold nanoparticles functionalization with cystamine dihydrochloride facilitates a covalent bonding between surface amino group and carboxylic Fc ensuring maximizing available active antibody. This strategy ensures a much lower limit of detection in addition to improved detection range due to increased loading capacity as a result of larger effective surface area offered by gold nanoparticles. These two aspects of immunosensor fabrication resulted in limit of detection as low as 0.001 mu IU/mL and an enhanced detection range of 0.001-150 mu IU/mL. This makes the developed immunosensor suitable for diagnostic purpose covering the clinically relevant range and a simple detection technique makes it potential candidate for fabrication of a Point-of-Care device for detection of Thyroid dysfunctioning. For next-generation electronics, it is crucial to be able to accurately mimic the nature of human skin and develop skin-inspired materials and devices for electronic skin (e-skin) applications. However, it is a great challenge to design such a materials because they require the complex integration of stretchability, skin adhesiveness, transparency, healability, and tactile sensing. Herein, we present the first dual covalently reversible dynamic bond-based polymer that can be integrated with various functional materials and used as a skin-like sensor. A series of polymers was designed using polypropylene glycol (PPG)-based polyurethane (PU) as the polymer backbone, and these polymers could integrate two types of covalent reversible dynamic bonds, Diels-Alder and disulfide bonds, in one polymer for the first time. Based on our design, a polymer was created with the advantages of high stretchability, fast healing abilities (60 degrees C, 5 min), skin adhesion, recyclability, high transparency, and tactile sensing abilities. We provide an alternative strategy for the construction of skin-inspired multifunctional materials for e-skin applications.

Galectin-3 (gal-3) plays a crucial role in various cellular events associated to tumor metastasis and progression. In this direction, gal-3 binding core-shell glyconanoparticles based on citrus pectin (CP) have been designed for targeted, trigger-responsive combination drug delivery. Depolymerization via periodate oxidation in heterogeneous medium yielded low-molecular weight dialdehyde oligomers (CPDA) of CP with a gal-3 binding property (K-d = 160.90 mu M). CPDA-based core-shell nanoparticles prepared to enhance the gal-3 binding specificity via a multivalent ligand presentation have shown to reduce homotypic cellular aggregation, tumor cell binding with endothelial cells, and endothelial tube formation, the major steps involved in the progression of cancer. Immune-fluorescence and flow cytometric analysis confirmed significant reduction in gal-3 expression on MDA-MB 231 cancer cells upon incubation with nanoparticles. An on-demand tumor microenvironment-responsive release of drugs at low pH and high concentrations of glucose and glutathione prevailing in tumor milieu was achieved by introducing a cleavable Schiff’s base, a boronate ester, and disulfide linkages within the shell of the nanoparticles. Nanoparticles with encapsulated sulindac in the core and doxorubicin (DOX) in the shell demonstrated target specificity and enhanced internalization with synergistic cytotoxic effects with a 30-fold reduction in IC50 in DOX-resistant, triple-negative MDA-MB 231 breast cancer cells. Nanoparticles were radiolabeled with 131I radioisotopes with >= 80% efficiency while retaining its gal-3 binding property. Biodistribution studies of radiolabeled placebo nanoparticles and drug-loaded CPDA nanoparticles demonstrated proof of concept of gal-3 targeting seen as preferential accumulation in the gal-3-expressing tissues of the gastric tract. The CPDA core-shell nanoparticles are thus promising platforms for gal-3 targeting and inhibition of gal-3-mediated processes involved in cancer progression with a potential of radiolabeling for in vivo monitoring or delivering therapeutic doses of radiation and on-demand triggered, target-specific drug release.
