Combined blockades of CTLA-4 and PD-1 can yield better overall complementary clinical outcomes than individual blockades, but the response rates are still relatively low. To investigate the anti-tumor effects of various combined strategies, we designed various spherical nucleotide nanoparticles (SNPs) loaded with CTLA-4 aptamer (cSNPs), PD-1 siRNA (pSNPs) or both (hybrid SNPs, or hSNPs). The results demonstrated that hSNPs could promote significantly stronger anti-tumor immune responses in a nonredundant fashion than the mixture of pSNPs and cSNPs (pSNPs & cSNPs). We reasoned that this is because all individual immune cells could receive both CTLA-4 and PD-1 blockades when they engulfed hSNPs, but it is much less likely that individual immune cells could receive both CTLA-4 and PD-1 blockades as many of them may not take both pSNPs and cSNPS from pSNPs & cSNPs. Further results revealed that the synergistic immune stimulatory effects of CTLA-4 and PD-1 blockades in the form of hSNPs were at least partly through regulating the immune suppressive function of both Tregs and TIM3(+)exhausted-like CD8 T cells and allowing effector T cells to expand. This mechanism is not identical to earlier reported mechanisms of CTLA-4 and PD-1 blockades.

Background: Recent data have suggested a substantial incidence of atrial arrhythmias (AAs) in cardiac sarcoidosis (CS). Our study aims were to first assess how often AAs are the presenting feature of previously undiagnosed CS. Second, we used prospective follow-up data from implanted devices to investigate AA incidence, burden, predictors, and response to immunosuppression. Methods and Results: This project is a substudy of the CHASM-CS (Cardiac Sarcoidosis Multicenter Prospective Cohort Study; NCT01477359). Inclusion criteria were presentation with clinically manifest cardiac sarcoidosis, treatment-naive status, and implanted with a device that reported accurate AA burden. Data were collected at each device interrogation visit for all patients and all potential episodes of AA were adjudicated. For each intervisit period, the total AA burden was obtained. A total of 33 patients met the inclusion criteria (aged 56.1 +/- 7.7 years, 45.5% women). Only 1 patient had important AAs as a part of the initial CS presentation. During a median follow-up of 49.1 months, 11 of 33 patients (33.3%) had device-detected AAs, and only 2 (6.1%) had a clinically significant AA burden. Both patients had reduced burden after CS was successfully treated and there was no residual fluorodeoxyglucose uptake on positron emission tomography scan. Conclusions: First, we found that AAs are a rare presenting feature of clinically manifest cardiac sarcoidosis. Second, AAs occurred in a minority of patients at follow-up; the burden was very low in most patients. Development of novel activable dual-mode T-1/T-2-weighted magnetic resonance (MR) contrast agents with the same composition for dynamic precision imaging of tumors has been a challenging task. Here, we demonstrated a strategy to prepare clustered Fe3O4 nanoparticles (NPs) with redox-responsiveness to tumor microenvironment to achieve switchable T-2/T-1-weighted dual-mode MR imaging applications. In this study, we first synthesized ultrasmall Fe3O4 NPs with an average size of 3.3 nm and an r, relaxivity of 4.3 mM(-1) s(-1), and then cross-linked the single Fe3O4 NPs using cystamine dihydrochloride (Cys) to form clustered Fe3O4/Cys NPs. The Fe3O4 nanoclusters (NCs) possess desirable colloidal stability, cytocompatibility, high r(2) relaxivity (26.4 mM(-1) s(-1)), and improved cellular uptake efficiency. Importantly, with the redox-responsiveness of the disulfide bond of Cys, the Fe3O4 NCs can be dissociated to form single particles under a reducing condition, hence displaying a switchable T-2/T-1-weighted MR imaging property that can be utilized for dynamic precision imaging of cancer cells in vitro and a subcutaneous tumor model in vivo due to the reductive intracellular environment of cancer cells and the tumor microenvironment. With the tumor reductive microenvironment-mediated switching of T-2 to T-1 MR effect and the ultrasmall size of the single particles that can pass through the kidney filter, the developed Fe3O4 NCs may be used as a promising switchable T-2/T-1 dual-mode MR contrast agent for precision imaging of different biosystems.

Molecular machines are ubiquitous in nature and function away from equilibrium by consuming fuels to produce appropriate work. With tunable rate and binding constants toward relevant guests, cucurbiturils may become excellent ring molecules for the construction of advanced molecular machines working in water.

The aim of this work was to obtain a novel onco-theranostic system for early diagnosis and improved chemotherapeutic outcome. Hybrid nanogels with magnetic and dual responsive properties were fabricated by covalently attaching superparamagnetic iron oxide nanoparticles (SPIONs) with a disulfide-modified alginate derivative, while simultaneously encapsulating the anticancer drug doxorubicin. The resulting nanogels exhibited magnetic-targeted characteristics, high drug loading content, co-triggered release behavior, high toxicity to tumor cells, low side effects to normal cells, and magnetic resonance imaging (MRI) functions. These findings proved that the hybrid nanogels have great potential as novel tumor-targeting nano-theranostic agents for simultaneous MRI imaging and efficient anti-tumor treatment.

A new method of supramolecular emulsion interfacial polymerization is developed, which can be used to fabricate supramolecular polymeric nanospheres. We designed a water-soluble monomer containing two maleimide end groups, acting as both building block and surfactant, and an oil-soluble supramonomer bearing two thiol groups connected by quadruple hydrogen bonds. With the assist of ultrasonication, hollow nanospheres can be controllably prepared by thiol-maleimide reaction of two monomers at the emulsion interface, which exhibit good stability and dynamic property. In addition, the encapsulated guest molecules could be controllably released from the supramolecular polymeric nanospheres, owing to their stimuli-responsiveness. It is anticipated that this approach will enrich the methodology of supramolecular polymerization and can be applied to constructing supramolecular materials with controllable structures and functions.

Classic dendrimers are not used in many specific applications such as simultaneous loading of different drugs due to the existence of identical end groups. The possibility of designing the structure in dendrimers paves the way to synthesize more complicated dendrimers. This includes the creation of various types of peripheral groups on the surface of dendrimers to synthesize for example Janus dendrimers. Nowadays, three main methods are proposed for the synthesis of Janus dendrimers in which the high amount of conventional dendrimers is produced. The goal of this work is to develop a new method for synthesis of Janus dendrimers without production of conventional dendrimers. The present method has three main stages for the synthesis of Janus dendrimers including synthesis of 5th generation PPI dendrimer with cystamine core and hydrophobic surface, conversion of disulfide bonds to thiol group using a structure scission approach, and the formation of PAMAM hydrophilic dendrons with amine end groups. Different analyses including FT-IR, NMR, DLS, and GPC are used to prove the success of the synthesis route. Also, due to application of dendrimers in drug delivery including solubility improvement of hydrophobic drugs, Janus dendrimers are used to improve solubility of tetracycline and dexamethasone in presence of different generations of Janus dendrimers. Since the Janus dendrimers have both hydrophilic and hydrophobic ends, as well as having numerous terminal groups and numerous internal cavities, they have resulted in significant improvements in drug solubility. Also, solubility of the two hydrophobic drugs in water was increased by increasing concentration and generation of dendrimer.

The study of multifunctional polymer micelles combined with chemotherapy due to reduced systemic toxicity and enhanced efficacy has attracted intensive attention. Herein, a multifunctional core-cross-linked hybrid micelle system based on mPEG-b-PGu(BA-TPE) and OCT-PEG-b-PGu(DA-TPE) with pH- and redox-triggered drug release and aggregation-induced emission (AIE) active imaging has been developed for active targeting of neuroendocrine neoplasms (NENs), especially neuroendocrine carcinomas (NECs) with poor prognosis. These micelles showed excellent biocompatibility and stability. After the formation of borate ester bonds, core-cross-linked micelles (CCLMs) showed enhanced emission properties. In addition, etoposide (ETO), one of the most important anticancer drugs of NECs, was loaded into the hydrophobic core of micelles by self-assembly with an average diameter of 274.6 nm and spherical morphology. Octreotide (OCT) conjugated onto the micelles enhanced cellular uptake by receptor-mediated endocytosis. The CAS is 56-17-7. Through research, I have a further understanding and discovery of 2,2′-Disulfanediyldiethanamine dihydrochloride.

A water-soluble cyclophane dimer having two disulfide groups as a reduction-responsive cleavable bond as well as several acidic and basic functional groups as a pH-responsive ionizable group 1 was successfully synthesized. It was found that 1 showed pH-dependent guest-binding behavior. That is, 1 strongly bound an anionic guest, 6-p-toluidinonaphthalene-2-sulfonate (TNS) with binding constant (K/M-1) for 1:1 host-guest complexes of 9.6 x 10(4) M-1 at pH 3.8, which was larger than those at pH 7.4 and 10.7 (6.0 x 10(4) and 2.4 x 10(4) M-1, respectively), indicating a favorable electrostatic interaction between anionic guest and net cationic 1. What is more, release of the entrapped guest molecules by 1 was easily controlled by pH stimulus. Large favorable enthalpies (Delta H) for formation of host-guest complexes were obtained under the pH conditions employed, suggesting that electrostatic interaction between anionic TNS and 1 was the most important driving force for host-guest complexation. Such contributions of Delta H for formation of host-guest complexes decreased along with increased pH values from acidic to basic solutions. Upon addition of dithiothreitol (DTT) as a reducing reagent to an aqueous PBS buffer (pH 7.4) containing 1 and TNS, the fluorescence intensity originating from the bound guest molecules decreased gradually. A treatment of 1 with DTT gave 2, having less guest-binding affinity by the cleavage of disulfide bonds of 1. Consequently, almost all entrapped guest molecules by 1 were released from the host. Moreover, such reduction-responsive cleavage of 1 and release of bound guest molecules was performed more rapidly in aqueous buffer at pH 10.7.
