In the present study, a series of novel biologically active 8-benzylidene-6-tert-butyl-4-phenyl-5, 6, 7, 8-tetrahydro-benzo-1, 3-thiazin – 2 -imines (TB1-TB12) were synthesized and evaluated for their biological activities. Initially, 2,6-dibenzylidene-4-tert-butylcyclohexanones were synthesized by Claisen-Schmidt condensation of 4-tert-butylcyclohexanone with various aromatic aldehydes in the presence of dilute sodium hydroxide. Further these compounds were subjected to cyclocondensation with thiourea, in isopropyl alcohol, catalyzed by aqueous potassium hydroxide to form 4-aryl-8-arylidene-5,6,7,8-tetrahydro-1H-benzo[d][1,3] thiazin-2(4H)-imines (TB1-TB12). The structures of the newly synthesized compounds have been established on the basis of their spectral data and elemental analysis. Synthesis, Characterization, and Molecular Docking of Novel bis-thiazolyl Thienothiophene Derivatives as Promising Cytotoxic Antitumor Drug

A novel, facile reaction for the synthesis of series of bis-thiazole derivatives has been developed from the reaction of the appropriate thiosemicarbazone derivatives and bis-2-bromoacetylthieno[2,3-b]thiophene derivatives in ethanol under reflux. The structures of the newly synthesized products were established on the basis of spectral data (mass, IR, and H-1 and C-13 NMR) and elemental analyses. Fifteen compounds of the synthesized compounds were evaluated for their anticancer activity against human liver hepatocellular carcinoma cell line (HepG2). The saturated and two-photon absorption of various classes of heterocyclic dyes [thiazine (thionine), xanthene (erythrosine), and carbocyanine (3,3′-di-(gamma-sulfopropy1)-4,4′,5,5′-dibenzo-9-ethylthiacarbocyaninebetaine pyridinium salt, DEC)] possessing optical absorption in the 500-680 nm range were measured at the wavelengths of 1064 and 532 nm using nanosecond (10 ns) and picosecond (40 ps) pulses and z-scan technique. It was shown that reverse saturated absorption unlikely plays important role in those dyes. We defined the two-photon absorption coefficients of thionine and erythrosine at 1064nm using 40 ps pulses (0.8 x 10(-11) and 1.0 x 10(-11) cmW(-1) respectively). The concurrence of saturated absorption and two-photon absorption was analyzed in the case of thionine and erythrosine using various models. It was suggested that dimerization of those dyes significantly suppresses the nonlinear optical response in the case of both 10 ns and 40 ps pulses of 532 nm radiation. At the same time it was found that, in the case of 532 nm pulses, the monomers of thionine participate in the processes of saturated absorption and two-photon absorption, while J-aggregates of DEC show predominantly two-photon absorption. And for pure solutions, the reverse saturated absorption is preferred nonlinear absorption mechanism. The nonlinear absorption coefficients for MB and AzA in the hybrid associates were found. They are beta approximate to 8.4 x 10(-9) cm W-1 (sic) approximate to 1.34 x 10(-8) cm W-1, respectively. Notably, there have been several attempts to introduce MB for attenuating pain in the last decade. Some clinical studies reported remarkable results, which, however, have been much debated. In addition, accumulating evidence have revealed that MB diminishes voltage-gated sodium channel currents. Accordingly, in the present study, we conducted in vivo experiments, including in vivo single nerve recording and behavioral test, to investigate whether MB dampens neural firing rates and ultimately contributes to pain relief. As a result, neural firing rates significantly decreased and finally converged to zero after MB administration. This event lasted longer than that of lidocaine and was dose-dependently modulated. Furthermore, there was a marked improvement in pain behaviors. The withdrawal threshold and latency of hind paws significantly rose post-MB administration. Therefore, these results demonstrate that MB lessens pain by significantly weakening neural excitability, which implies a strong possibility that this dye may be developed as a pain-relieving medication in the future. Strong evidence shows an association between cerebral vascular diseases and Alzheimer’s disease (AD). In order to study the interaction of beta-amyloid (A beta) plaques with brain vessels, we crossbred an AD mouse model (overexpressing amyloid precursor protein with the Swedish-Dutch-Iowa mutations, APP_SweDI) with mice expressing green fluorescent protein (GFP) under the flt-1/VEGFR1 promoter in vessels (GFP_FLT1). Our data show, that only very few A beta plaques were seen in 4-months old mice, focused in the mammillary body and in the lateral septal nucleus. The number of plaques markedly increased with age being most prominent in 12-months old mice. Thiazine Red was used to verify the plaques. Several Thiazine Red(+) inclusions were found in GFP(+) vessels, but only in non-perfused 4-months old mice. These inclusions were verified by Resorufin stainings possibly representing cerebral amyloid angiopathy. The inclusions were also seen in non-crossbred APP_SweDI but not in wildtype and GFP_FLT1 mice. In order to characterize these inclusions Flow Cytometry (FACS) analysis demonstrated that platelets were specifically stained by Thiazine Red(+), more pronounced when aggregated. In conclusion, our data show that Thiazine Red(+) inclusions representing aggregated platelets are a first pathological sign in AD before plaque development and may become important therapeutic targets in early AD.
