Molecular Docking and Receptor-Based 3D-QSAR Studies on Aromatic Thiazine Derivatives as Selective Aldose Reductase Inhibitors

Aromatic thiazine derivatives were proved to be potent aldose reductase inhibitors (ARIs) with high selectivity for aldose reductase (ALR2) over aldehyde reductase (ALR1). Molecular docking and three-dimensional quantitative structure-activity relationship (3D-QSAR) studies are conducted on a dataset of 44 molecules to explore the interactions between aromatic thiazine derivatives and ALR2. The superposition of ALR2 and ALR1 active sites indicate that residues Leu 300 and Cys 298 from ALR2 may explain the good selectivity of the most active compound 1m. The comparative molecular field analysis (CoMFA) model (q(2) = 0.649, r(2) = 0.934; q(2): cross-validated correlation coefficient, r(2): non-cross-validated correlation coefficient) and comparative molecular similarity indices analysis (CoMSIA) model (q(2) = 0.746, r(2) = 0.971), based on the docking conformations of these compounds, are obtained to identify the key structures impacting their inhibitory potencies. The predictive power of the developed models is further validated by a test set of seven compounds, resulting in predictive r(Pred)(2) values of 0.748 for CoMFA and 0.828 for CoMSIA. 3D contour maps, drawn from 3D-QSAR models, reveal that future modifications of substituents at the C3 and C4 positions of the benzyl ring and the C5 and C7 positions of the benzothiazine-1,1-dioxide core might be favorable for improving the biological activity, which are in good accordance with the C7 modification results reported in our earlier work. The information rendered by 3D-QSAR models could be helpful in the rational design of novel ARIs with good inhibitory activity to treat diabetic complications in the future.

Background Epilepsy is a common neurological condition characterised by recurrent seizures. Most patients respond to conventional antiepileptic drugs, however, around 30% will continue to experience seizures despite multiple antiepileptic drugs. Sulthiame, also known as sultiame, is a widely used antiepileptic drug in Europe and Israel. We present a summary of the evidence for the use of sulthiame as add-on therapy in epilepsy. Objectives To compare the efficacy and side-effect profile of sulthiame as add-on therapy compared with placebo or another antiepileptic drug. Search methods We searched the Cochrane Epilepsy Group’s Specialised Register, the Cochrane Central Register of Controlled Trials (CENTRAL), MEDLINE, ClinicalTrials.gov and the WHO ICTRP Search Portal on 11 August 2015. No language restrictions were imposed. We contacted the manufacturers of sulthiame and researchers in the field to seek any ongoing or unpublished studies. Selection criteria Randomised controlled add-on trials of sulthiame in people of any age with epilepsy of any aetiology. Data collection and analysis Two review authors independently selected trials for inclusion and extracted relevant data. The following outcomes were assessed: 1) reduction in seizure frequency of 50% or greater between baseline and end of follow-up; 2) complete cessation of seizures during follow-up; 3) mean seizure frequency; 4) time to treatment withdrawal; 5) adverse drug effects; and 6) quality of life scoring. Primary analyses were intention-to-treat. We present a narrative analysis. Main results We included one trial with 37 participants with a new diagnosis of West syndrome. Sulthiame was given as an add-on therapy to pyridoxine. No data were reported for outcomes 1), 3) or 6). Overall risk ratio with 95% confidence intervals (CI) for complete cessation of seizures during a nine-day follow-up period versus placebo was 0.71 (95% CI 0.53 to 0.96). Meaningful analysis of time to treatment withdrawal and adverse drug effects was not possible due to incomplete data. Authors’ conclusions Sulthiame may lead to a cessation of seizures when used as an add-on therapy to pyridoxine in patients with West syndrome. The included study was small and had a significant risk of bias which limits the impact of the evidence. CONSPECTUS: Over the past decade, the field of photoredox catalysis has gained increasing attention in synthetic organic chemistry because of its wide applicability in sustainable free-radical-mediated processes. Numerous examples have shown that under carefully optimized conditions, efficient and highly selective processes can be developed through excitation of a photosensitizer using inexpensive, readily available light sources. However, despite all of these recent advancements, some generalizations and/or misconceptions have become part of the photoredox culture, and often many of these discoveries lack in-depth investigations into the excited-state kinetics and underlying mechanisms. In this Account, we begin with a tutorial for understanding both the redox properties of excited states and how to measure the kinetics of excited-state processes. We discuss the generalization of direct excitation of closed-shell species to generate more potent reductive or oxidative excited states, using the helium atom as a quantitative example. We also outline how to apply redox potentials to calculate whether the proposed electron transfer events are thermodynamically feasible. In the second half of our tutorial, we discuss how to measure the kinetics of excited-state processes using techniques such as steady-state and time-resolved fluorescence and transient spectroscopy and how to apply the data using Stem-Volmer and kinetic analysis. Then we shift gears to discuss our recent contributions to the field of photoredox catalysis. Our lab focuses on developing transition metal-free alternatives to ruthenium and iridium bipyridyl complexes for these transformations, with the goal of developing systems in which the reaction kinetics is more favorable. We have found that methylene blue, a member of the thiazine dye family, can be employed in photoredox processes such as oxidative hydroxylations of arylboronic acids to phenols. Interestingly, we were able to demonstrate that methylene blue is more efficient for this reaction than Ru(bpy)(3)Cl-2, which upon further examination using transient spectroscopic techniques we were able to relate to the reductive quenching ability of the aliphatic amine. Recently we were also successful in applying methylene blue for radical trifluoromethylation reactions, which is discussed in detail. Finally, we have also demonstrated that common organic electron donors, such as alpha-sexithiophene, can be used in photoredox processes, which we demonstrate using the dehalogenation of vic-dibromides as a model system. This is a particularly interesting system because well-defined, long-lived intermediates allowed us to fully characterize the catalytic cycle. Once again, through an in-depth kinetic analysis we were able to gain valuable insights into our reaction mechanism, which demonstrates how powerful a tool proper kinetic analysis can be in the design and optimization of photoredox processes.

Maximum activity of enzyme was observed at 50 degrees C and pH 7, which did not change in the alkaline pH and temperature range of 20-70 degrees C. Results indicated ions, inhibitors and solvent stability of the enzyme and its activity were stimulated by Co2+, Mn2+, PMSF, acetone, acetonitrile, ethanol, and methanol. The spore laccase could decolorize synthetic dyes from various chemical groups including azo (acid orange, amaranth, trypan blue, congo red, and amido black), indigo (indigo carmine), thiazine (methylene blue, and toluidine blue), and triarylmethane (malachite green) with ABTS/syringaldazine mediators after 5 h. Degradation products were not toxic against Sorghum vulgare and Artemia sauna model organisms.
