Crystal structure, Hirshfeld surface analysis and interaction energy, DFT and antibacterial activity studies of ethyl 2-[(2Z)-2-(2-chlorobenzylidene)-3-oxo-3,4-dihydro-2H-1,4-benzothiazin-4-yl]acetate

The title compound, C19H16ClNO3S, consists of chlorophenyl methylidene and dihydrobenzothiazine units linked to an acetate moiety, where the thiazine ring adopts a screw-boat conformation. In the crystal, two sets of weak C-HPh center dot center dot center dot ODbt (Ph = phenyl and Dbt = dihydrobenzothiazine) hydrogen bonds form layers of molecules parallel to the bc plane. The layers stack along the aaxis direction with intercalation of the ester chains. The crystal studied was a two component twin with a refined BASF of 0.34961 (5). The Hirshfeld surface analysis of the crystal structure indicates that the most important contributions to the crystal packing are from H center dot center dot center dot H (37.5%), H center dot center dot center dot C/C center dot center dot center dot H (24.6%) and H center dot center dot center dot O/O center dot center dot center dot H (16.7%) interactions. Hydrogen-bonding and van der Waals interactions are the dominant interactions in the crystal packing. A Rapid One-Pot Five Component Sequential Access to Novel Imidazo[2,1-b] thiazinyl-alpha-aminophosphonates

Synthesis and antioxidant activity of some 1-aryl/aralkyl piperazine derivatives with xanthine moiety at N4

Piperazine nucleus is one of the most important heterocyclic systems exhibiting remarkable pharmacological activities. Thus, in the current study six new aryl/aralkyl substituted piperazine derivatives, containing methylxanthine moiety were synthesized and their structures were confirmed by IR and H-1 NMR analysis. All compounds were in vitro screened for their activity as antioxidants using DPPH (2,2 ‘-Diphenyl-1-picrylhydrazyl), ABTS (2,2 ‘-azinobis-(3-ethylbenzo thiazine-6-sulfonic acid)) and FRAP (ferric reducing/antioxidant power) methods. The antioxidant activity of the studied compounds against lipid peroxidation was also measured. The highest antioxidant activity was demonstrated by compound 3c. It is obvious that the presence of a hydroxyl group in the structure is essential for the antioxidant properties and should be taken into consideration in further design of structures with potential antioxidant properties.

Stereoselective Synthesis and Antiproliferative Activity of Monoterpene-Fused 2-Imino-1,3-oxazines

Background: In the recent years the 2-imino-1,3-thiazine and 2-iminothiazolidine ring systems can be found as moieties in biologically relevant compounds, including BACE1 inhibitors, or cannabinoid receptor agonists, while monoterpene-based 2-imino-1,3-thiazines, prepared from chiral 1,3-amino alcohols exhibiting pronounced antiproliferative activity. Methods: The antiproliferative activities of the prepared compounds were determined in vitro against a panel of human adherent cancer cell lines including HeLa, MCF7 and A431 by MTT assay. Results: Starting from pinane-, apopinane-and carane-based beta-amino acid derivatives, 1,3-amino alcohols were prepared via two-step syntheses. The reactions of the product 1,3-amino alcohols and aryl isothiocyanates yielded gamma-hydroxythioureas, which were transformed to monoterpene-fused 2-imino-1,3-oxazines via base-catalysed ring closure. The antiproliferative activities of these 2-imino-1,3-oxazines were examined and the structure-activity relationships were studied from the aspects of the type and stereochemistry of the monoterpene ring and the substituent effects on the 1,3-oxazine ring system. The N-unsubstituted monoterpene-based derivatives exhibited considerable antiproliferative activity against a panel of human adherent cancer cell lines (HeLa, MCF7 and A431). Conclusions: A mild and efficient method has been developed for the synthesis of 2-imino-1,3-oxazines by the ring closure of thiourea adducts of 1,3-amino alcohols. The resulting 1,3-oxazines exert marked antiproliferative action on a panel of human cancer cell lines.

Design, synthesis and anticancer evaluation of novel thiazine, pyrimidine and pyridine derivatives

Acylisothiocyanate (1) was allowed to react with benzylidenemalononitrile producing oxazine derivative (2). The reaction of acetylacetone with compound (1) gave pyridine derivative (5) by intramolecularcycloaddition while it was reacted with N-methyl aniline affording thiazine derivative (7). In addition, it was reacted with cyanoacetamide producing mercaptopyrimidine derivative (9). Finally, compound (1) was refluxed with phenylhydrazine, urea, guanidmum carbonate and anthranilic acid in the presence of dry acetone affording triazole derivative (11), N-substituted pyrimidine (15), compound (18) and thiopyrimidine derivative (19) respectively. The structures of the new compounds were confiimed on the basis of elemental and spectral data. Some of the synthesized compounds were screened as anticancer.

The compound 2-aryl-2,3-dihydro-4H- [1,3]thiazino[3,2-a]benzimidazol-4-one (C16H12N2OS) (III) was synthesized. The compound crystallized in a monoclinic crystal system with P2(1)/c space group as revealed by mono-crystal X-ray diffraction. The X-ray analysis and density functional theory complementary calculations showed that noncovalent C-H center dot center dot center dot pi interactions between the benzimidazole system and the thiazine ring form a dimerization along the direction of the b axes, additionally pi center dot center dot center dot pi interactions were found between the benzene rings. The C12, in the thiazine ring, has a distortion angle (41.69 degrees) respect to the plane of benzimidazole. The intermolecular interactions in the crystal structure were quantified and analyzed using Hirshfeld surface analysis. The predominant interaction within the crystalline structure was found to be H center dot center dot center dot H interaction. On the other hand, antioxidant activity of the C16H12N2OS system was studied using the DPPH center dot and ABTS center dot(+) assay. Using the energy profile for the reaction of DPPH center dot with III it was demonstrated that the antioxidant activity is carried out through HAT (H12) mechanism where conjugation of the radical can occur between the C12 of the thiazine ring and the aromatic ring, this was confirmed by a H function calculation. Organic Synthesis with N-Heterocyclic Carbenes of Indazole: Synthesis of Benzo(thio)imidates, Benzo[d][1,3]thiazines and Quinazoline-4-thiones

The N-heterocyclic carbenes of indazole, generated by deprotonation of indazolium salts, proved to be versatile starting materials for organic synthesis.
