The lipophilicity was expressed as the chromatographically derived descriptors: mean of k(mk), mean of logk (mlogk), logk(W), S, and phi(0), scores of k, logk, and R-M corresponding to the first principal component. Additionally, the authors propose to introduce a descriptor (logk(m)) corresponding to the residual specific interactions. Chromatographic parameters of lipophilicity were compared with the partition coefficient (logP) calculated by various softwares (milogP, clogP, AlogPs, AClogP, AlogP, MlogP, KOWWIN, XlogP2, XlogP3). We also describe techniques for blocking branches of the mandibular nerve: inferior alveolar nerve, mental (or incisive) nerve, lingual nerve, and long buccal nerve. Local anesthesia for the mandibular and maxillary nerves can be accomplished in rhesus macaques and is a practical and efficient way to maximize animal welfare during potentially painful orofacial procedures.

Background: Thiazoles and pyridines are versatile synthetic scaffolds possessing wide spectrum of biological effects including potential antimicrobial activity. Objective: In the efforts to develop suitable antimicrobia drugs, medicinal chemists have focused on thiazole derivatives. A novel series of 2-thiazolyl pyridines was prepared in a one pot three-component reaction using 2-bromoacetyl pyridine as a starting precursor. Method: Structure of the synthesized compounds was elucidated by spectral data (FT-IR, H-1 NMR, C-13 NMR, and mass) and elemental analyses. The prepared compounds were screened for their in vitro antimicrobial activity. Results: The results revealed that compounds 4a,b,e-g and 12 showed promising activity. Molecular docking studies using MOE software were carried out for compounds 4a and 4b which exhibited potent activities indicated by the diameter zones (4a; 3.6, 4.0, 1.2 mm) (4b; 4.2, 3.5, 1.5 mm) and the binding affinities (4a; -5.7731, -5.3576, -4.6844 kcal mol(-1)) (4b; -5.9356, -2.8250, -5.3628 kcal mol(-1)) against Candida albicans, Bacillus subtilis and Escherichia coli, respectively. Conclusion: This paper describes a facile and efficient MCR for synthesis of 2-thiazolyl pyridines from reaction of 2-bromoacetyl pyridine with different reagents. There was an agreement between the values of binding affinities and interactions and the data obtained from the practical antimicrobial screening of the tested compounds.

Synthesis of 16 new tetracyclic angularly fused azaphenothiazines, 8-, 9- and 10-substituted quinobenzo1,4-thiazines (benzo[a]-3-azaphenothiazines) was based on the reactions of dichlorodiquinolinyl disulfide and diquinodithiin with substituted anilines. Whereas the reactions with p-fiuoroaniline and p-methylthioaniline led to only one product, the reaction with m-triflouromethylaniline led to isomeric compounds. The obtained 8-10-substituted 12H-quinobenzothiazines were further transformed into 12 substituted derivatives through allcylation of the thiazine nitrogen atom. The structure analysis was based on 1D and 2D NMR (NOESY, COSY, HSQC and HMBC) spectra which enabled to distinguish the isomers and to exclude retro-Smiles rearrangement and the azine nitrogen atom alkylation pathways. This supposition was fully confirmed by X-ray analysis showing the quinobenzothiazine system to be folded and the substituent at the thiazine nitrogen atom in an equatorial position. Some compounds exhibited anticancer activity against MCF-7, MDA-MB-231 and SNB-19 cell lines similar to a reference drug cisplatin. One-pot cis-selective route to sugar-fused thiazines via a masking-unmasking strategy in basic ionic liquid

A novel sequential Knoevenagel condensation, thia-Michael, and amino/mercaptoacetylative ring transformation reaction cascade for cis-selective synthesis of sugar-fused 1,3-thiazine is reported. The expeditious one-pot multicomponent annulation was performed using masked amino acid viz. 2-phenyl-1,3-oxazol-5-one or masked mercaptoacid viz. 2-methyl-2-phenyl-1,3-oxathiolan-5-one, D-xylose/D-glucose, and N-aryldithiocarbamic acid in ionic liquid [bmim] OH. The acetophenone obtained as a by-product and [bmim] OH itself could be easily recycled for further use without loss of efficiency. The envisaged method is operationally simple, high yielding, and excellent diastereoselective in favor of the cis-isomer of fused thiazines.
