Radical cation and dication of a 4H-dithieno[2,3-b:3′,2′-e][1,4]-thiazine

Experimental and Quantum Chemical Analysis of New Pyrimidothiazine Derivative as Corrosion Inhibitor for Mild Steel in 1.0 M Hydrochloric Acid Solution

The corrosion inhibition efficiency of pyrimidothiazine compound, namely: 8-ethyl-3-hydroxy-4,6-dioxo-2-(p-tolyl)-4,6-dihydropyrimido [2,1-b][1,3] thiazine-7carbonitrile (EHDPTC), was evaluated in the system steel/1 M HCl. This compound is investigated experimentally using weight loss, potentiodynamic polarization curves and electrochemical impedance spectroscopy. The results showed that the inhibition mechanism involves blockage of the steel surface by the inhibitor molecules by a Langmuir-type adsorption process and that the structure of molecule plays an important role in the inhibition efficiency of the synthesized inhibitor. The thermodynamic parameters of activation and adsorption have been calculated and discussed in detail. Furthermore, the quantum chemical parameters have been calculated and discussed in view of the results earlier reported.

Synthesis and biological evaluation as well as in silico studies of arylpiperazine-1,2-benzothiazine derivatives as novel anti-inflammatory agents

Novel arylpiperazine-1,2-benzothiazine derivatives have been designed and synthesized as potential antiinflammatory agents. Their structure and properties have been studied using spectroscopic techniques (H-1 NMR, C-13 NMR, FT-IR), MS, elemental analyses, and single-crystal X-ray diffraction (SCXRD, for compound 7b). This study aimed to evaluate the inhibitory activity of new derivatives against both cyclooxygenase isoforms COX-1 and COX-2 due to the similarity of new compounds to oxicams drugs from the NSAIDs group. All new compounds were divided into two series – A and B – with a different linker between thiazine and piperazines nitrogens. Series A included the three-carbon aliphatic linker and series B – two-carbon with a carbonyl group. According to in vitro and molecular docking studies all new compounds exhibited cyclooxygenase inhibitory activity. The series of A compounds included COX-1 inhibitors only. In contrast, the B series showed inhibition of both COX-1 and COX-2, which suggested the importance of the acetoxy linker for COX-2 inhibition. Moreover, the most selective compound 7b, towards COX-2, was non-toxic for the normal human cell line (in concentration of 10 mu M) comparable to reference drug meloxicam. Additionally, investigation of influence on model membranes confirmed the ability of the compound 7b to penetrate lipid bilayers which seemed to be important to the influence with membrane protein-cyclooxygenase.

Synthesis of 5,6-dihydro-4H-1,3-thiazine containing peptide mimics from N-(3-hydroxypropyl)thioamides and epimerization studies

The first synthesis of 1,3-thiazine fused peptide mimics is described from N-(3-hydroxypropyl)thioamides under MsCl/NEt3 conditions. L-Arginine/Nitric Oxide Pathway Is Altered in Colorectal Cancer and Can Be Modulated by Novel Derivatives from Oxicam Class of Non-Steroidal Anti-Inflammatory Drugs

Simple Summary Nitric oxide and arginine metabolism in colorectal cancer (CRC) holds potential for therapeutic intervention. We hypothesized that it can be modulated by oxicams, a class of non-steroidal anti-inflammatory drugs with documented chemopreventive and antineoplastic activity. The aim of this study was to determine the transcriptional patterns of pathway enzymes in CRC and evaluate the impact of classic and new oxicam analogues. Arginine metabolic pathways were altered not only in tumors but also in non-transformed mucosa from tumor vicinity, contributing to the phenomenon of tumor molecular margin. Classic oxicams, piroxicam and meloxicam, had negligible impact but their new analogues downregulated expression of dimethylarginine dimethylaminohydrolases and protein methyltransferases and upregulated asymmetric dimethylarginine. Those beneficial effects were accompanied by upregulation of arginase-2 and the potentially disadvantageous accumulation of arginine and symmetric dimethylarginine. Our findings provide novel insight into metabolic reprogramming in CRC and demonstrate that oxicam analogues are worth further consideration as novel anticancer agents. L-arginine/nitric oxide pathway metabolites are altered in colorectal cancer (CRC). We evaluated underlying changes in pathway enzymes in 55 paired tumor/tumor-adjacent samples and 20 normal mucosa using quantitative-PCR and assessed the impact of classic and novel oxicam analogues on enzyme expression and intracellular metabolite concentration (LC-MS/MS) in Caco-2, HCT116, and HT-29 cells. Compared to normal mucosa, ARG1, PRMT1, and PRMT5 were overexpressed in both tumor and tumor-adjacent tissue and DDAH2 solely in tumor-adjacent tissue. Tumor-adjacent tissue had higher expression of ARG1, DDAH1, and DDAH2 and lower NOS2 than patients-matched tumors. The ARG1 expression in tumors increased along with tumor grade and reflected lymph node involvement. Novel oxicam analogues with arylpiperazine moiety at the thiazine ring were more effective in downregulating DDAHs and PRMTs and upregulating ARG2 than piroxicam and meloxicam. An analogue distinguished by propylene linker between thiazine’s and piperazine’s nitrogen atoms and containing two fluorine substituents was the strongest inhibitor of DDAHs and PRMTs expression, while an analogue containing propylene linker but no fluorine substituents was the strongest inhibitor of ARG2 expression. Metabolic reprogramming in CRC includes overexpression of DDAHs and PRMTs in addition to ARG1 and NOS2 and is not restricted to tumor tissue but can be modulated by novel oxicam analogues.

The geometrical and non-linear optical parameters of the studied complexes 1-3 are investigated theoretically at the B3LYP/GENECP level of theory. The optimized geometries of the studied complexes are non-planner as indicated from the dihedral angles. The natural charge population (core, valence and Rydberg), exact electronic configuration, total Lewis, and total non-Lewis is computed and discussed in terms of natural bond orbitals (NBO) analysis. The calculated E-HOMO and E-LUMO energies at the same level of theory of the studied complexes were used to calculate the global properties; hardness (), global softness (S), electrophilicity () and electronegativity (). The total dipole moment ((tot)), total and anisotropy of polarizability (??), () and first hyperpolarizability (??) values were calculated and compared with urea as a reference compound. From the values of the computed first hyperpolarizability (??), the ligand and the studied complexes show promising optical properties.

5H-Benzo[d]Benzo[4,5]Imidazo[2,1-b][1,3]Thiazine as a Novel Electron-Acceptor Cored High Triplet Energy Bipolar Host Material for Efficient Solution-Processable Thermally Activated Delayed Fluorescence Organic Light-Emitting Diodes

Organic entities that can transport electrons are seldom available to develop adequate bipolar host materials applicable for solution-processable thermally activated delayed fluorescence (TADF)-organic light-emitting diodes (OLEDs). Therefore, the introduction of new electron-affine entities that plausibly demonstrate high triplet energy (E-T) is of urgent need. In this contribution, we introduced benzimidazo[1,2-a][3,1]benzothiazine (BBIT) as a novel electron-affine entity and developed two new bipolar host materials, CzBBIT and 2CzBBIT. Both host materials exhibit high E-T of 3.0 eV, superior thermal robustness with the thermal decomposition temperature of up to 392 degrees C, a glass transition temperature of up to 161 degrees C, and high solubility in common organic solvents. Consequently, the solution-processable OLEDs fabricated using a recognized IAcTr-out as the green TADF emitter doped into CzBBIT as the host, realized a maximum external quantum efficiency (EQE) of 23.3%, while the 2CzBBIT:IAcTr-out blend film-based device displayed an EQE of 18.7%. These outcomes corroborated that this work could shed light on the scientific community on the design of new electron-affine entities to establish the effective use of bipolar host materials toward proficient solution-processable TADF-OLEDs.
