Dipyrimido[4,5-b:5,4-e][1,4]thiazine: synthesis and their enzyme inhibitory activity assessment on soybean 15-lipoxygenase

A series of new derivatives of dipyrimido[4,5-b:5,4-e][1,4] thiazine were synthesized by treatment of 5-amino-6-methyl-2-morpholino-4-pyrimidinethiol (1) with 5-bromo-2,4-dichloro-6-methylpyrimidine (2) in the presence of triethylamine and ethanol and subsequently with various secondary amines in EtOH/DMF. The thioether derivative 3 and heterocyclization products 4a-f were characterized by elemental analysis and spectroscopic techniques. The 15-LO inhibitory activities of the new synthesized compounds were also evaluated. The results show that compound 4e has the best IC50 of 15-LO inhibition (IC50 = 14.4 A mu M) but when all the products were theoretically docked into 15-LO, the GOLD scores of compounds 4e and 4d are 43.67 and 43.39 respectively, which show the best results. We suggest that the hydrogen bond interaction between Ser489 of 15-LO and the nitrogen of piperazine ring of compound 4e appears to play major role in lipoxygenase inhibition.

A simple and efficient pathway to conjugate monosaccharides to thienopyrimidines via click chemistry establishing a 1,2,3-triazole linker was reported. Enhancement of nonlinear optical response of methylene blue and azure a during association with colloidal CdS quantum dots

The nonlinear optical response of thiazine dyes during association with colloidal CdS quantum dots (QDs) applying the z-scan technique at wavelength of 532 nm with use 10 ns pulses of Nd:YAG laser second harmonic were studied. Increasing in nonlinear absorption intensity in hybrid associates in comparison with the initial components were found. Moreover, UV-vis absorption spectrum shows the effective formation of dyes H-aggregates during association with colloidal QDs. Two-photon mechanism of observed nonlinear absorption in hybrid associates is proposed, based on a sharper dependence of the normalized transmittance on the incident intensity (distance between the sample and focal plane of focusing lens). And for pure solutions, the reverse saturated absorption is preferred nonlinear absorption mechanism. The nonlinear absorption coefficients for MB and AzA in the hybrid associates were found. They are beta approximate to 8.4 x 10(-9) cm W-1 (sic) approximate to 1.34 x 10(-8) cm W-1, respectively. It was shown that it is possible to control the parameters of nonlinear absorption of thiazine dyes via the hybrid association.

Structural Modifications of Surfactant-Assisted Alumina and Their Effectiveness for the Removal of Dyes

The current study focuses on the development of the modified surface of the alumina by anionic surfactant sodium dodecyl sulfate. The synthesized Surfactant Modified Alumina (SMA) effectively associated with dye molecules and amended their properties. The triphenylmethane (CBB) and thiazine dyes (MB) were selected as a simulated dye wastewater system. The removal was carried out by adsorption method under the optimized amount of adsorbent dosage, the concentration of adsorbate, contact time and temperature. The adsorption isotherm models like Langmuir, Freundlich and Dubinin Radushkevish were employed. The mechanism of the interaction represents the decolorized leuco dye molecules were formed after adsorption. Moreover, the thermodynamic parameters like (Delta S degrees), (Delta H degrees) and (Delta G degrees) were calculated represents the endothermic and spontaneous nature of the adsorption process. The pH at the point of zero charges (pHPZC) was determined. The photocatalytic degradation of respective dye systems was also observed. The surface morphology of (SMA) was determined by FT-IR and SEM techniques. Whereas the pseudo second order kinetics model was followed in the present system. The Pearson correlation Coefficient was conjointly applied. The removal efficiency was ascertained to be 99.50% for CBB(R-250) and 95.70% for (MB).

Synthesis of Benzo[1,4]thiazines via Ring Expansion of 2-Aminobenzothiazoles with Terminal Alkynes Under Metal-Organic Framework Catalysis

Copper-organic framework Cu-MOF-74 was synthesized, and consequently utilized as a heterogeneous catalyst for the synthesis of benzo[1,4]thiazines via ring expansion of 2-aminobenzothiazoles with terminal alkynes. Different from previous works, the reaction proceeded readily in the presence of lower catalyst concentration, at lower temperature, and under ligand-free conditions. The combination of 5 mol% framework catalyst, 20 mol% Cs2CO3, and 3 equivalents of di-tert-butyl peroxide led to high yields of benzo[1,4]thiazines. This copper-based framework demonstrated higher catalytic efficiency than a series of MOF-based heterogeneous catalysts and traditional homogeneous catalysts. In this system, the donation of soluble active copper species to the formation of benzo[1,4]thiazines was trivial. The copper-organic framework was reutilized without a remarkable decline in catalytic efficiency. To our best knowledge, this ring expansion reaction was not previously performed with a recyclable catalyst. [GRAPHICS] .

Media polarity and concentration roles on the third order nonlinear behaviors of thiazine dyes

Nonlinear optical materials play important roles in optics and photonics. In order to improve the performance of these materials, in this paper, we tried to find effective methods for controlling the nonlinear responses of thiazine dyes. Our experimental results indicate that molecular linear properties, media polarity, concentration and excitation beam properties with different contributions play significant roles on the saturable and reverse saturable absorption characteristics of thiazine dyes. In real, competition between these effective parameters leads to appearance of various third order nonlinear responses. Although by excitation wavelengths near the absorption peaks and at high powers, dye molecules tend to indicate reverse saturable absorption characteristics in polar protic solvents by increasing the concentration, saturable absorption characteristics were observed in polar aprotic solvents. Furthermore, the samples displayed the same behaviors in different solvent media at low powers and for excitation wavelengths far from absorption peaks. Therefore, by controlling the nonlinear responses of thiazine dyes, these materials can indicate both saturable and reverse saturable absorption characteristics that increase their applications in optics and photonics devices. Synthesis and oral hypoglycemic effect of novel thiazine containing trisubstituted benzenesulfonylurea derivatives

A new series of 3-(4-substituted phenyl)-1-(4-(4,6-dimethyl-6H-1,3-thiazin-2-yl)phenylsulfonyl)-1-substituted urea (5a-o) was synthesized by an effectual route via sulfonylcarbamates and explores the novel site for substitution in sulfonylurea as well as the way of thiazine can be prepared. The molecules were established by elemental analysis and spectroscopic viz. IR, H-1 NMR, C-13 NMR and MS techniques. All the fifteen derivatives were shown very prominent oral hypoglycemic effect at the dose of 40 mg/kg body weight (p.o.) in respect of standard drug glibenclamide and control. The hypoglycemic effect was studied using oral glucose tolerance test in normal and NIDDM in STZ-rat model. The compounds 5a, 5d, 5f, 5i, 5k and 5n were dominant out of fifteen derivatives for blood glucose lowering activity (more than 80%) when comparing with NIDDM control. These derivatives were either containing simply phenyl ring (5a, 5f and 5k) on to the second amine of sulfonylurea (R’ = H) or nitro group at the para position in compound 5d, Si and 5n (R’ = NO2) to produce significant oral hypoglycemic effect. Other structural activity relationship is also observed regarding the heteroaromatic and substituted aromatic group at R and R’ position respectively. Crystal structure of the 1 : 1 adduct of 2,3-diphenyl-3,4,5,6-tetrahydro-2H-1,3-thiazin-4-one with triphenyltin chloride

The title adduct, chlorido(2,3-diphenyl-3,4,5,6-tetrahydro-2H-1,3-thiazin-4-one-kappa O) triphenyltin, [Sn(C6H5)(3)Cl(C16H15NOS)], resulted from reaction of 2,3-diphenyl- 3,4,5,6-tetrahydro-2H-1,3-thiazin-4-one with triphenyltin chloride. The thiazine ring has an envelope conformation with the S atom forming the flap. The molecule has five phenyl rings, two of them attached to the thiazine ring at positions 2 and 3, and three in coordination with the Sn-IV atom. The three rings of the triphenyltin group are involved in intramolecular interactions of different types, C-H center dot center dot center dot O, edge-to-face (or T-type) pi-pi interactions with the 3-phenyl ring of the thiazine, T-type interactions with both phenyl rings of the thiazine etc. On the other hand, all the phenyl rings participate in intermolecular pi-pi interactions. There is one instance of a ‘parallel-displaced’-type interaction extending continuously along the a-axis direction and seven instances of T-type interactions stabilizing the crystal lattice.

Computational strategies to explore antimalarial thiazine alkaloid lead compounds based on an Australian marine sponge Plakortis Lita

In this work, an attempt was made to propose new leads based on the natural scaffold Thiaplakortone-A active against malaria. The 2D QSAR studies suggested that three descriptors correlate with the anti-malarial activity with an R-2 value of 0.814. Robustness, reliability, and predictive power of the model were tested by internal validation, external validation, Y-scrambling, and applicability domain analysis. HQSAR studies were carried out as an additional tool to find the sub-structural fingerprints. The CoMFA and CoMSIA models gave Q(2) values of 0.813 and 0.647, and http://www.w3.org/1999/ values of 0.994 and 0.984, respectively. Using the 2D-QSAR equation, the activity values of the seven modified compounds were calculated and it was found that three molecules showed good anti-malarial activity. Molecular docking of the 42 Thiaplakortone-A derivatives with Plasmodium falciparum calcium-dependent protein kinase 1 (PfCDPK1) was carried out to find out protein-ligand interactions. Data mining of the bioassay data-set AID: 504850 using the classifier based on Random Forest of Weka suggested that all of the eight molecules selected and three out of the seven virtual molecules were anti-malarial active. Both the virtual molecules and drug molecules were docked with CYP3A4, indicating that the virtual molecules could metabolize easily. Toxicity studies using Osiris shows that three molecules showed no toxic characters.
