Regioselective Synthesis of Functionalized 1,3-Thiazine-4-ones via Multicomponent Click Reaction Approach

Herein, we disclose a synthetic strategy for the preparation of functionalized thiazinones under exceptionally ambient condition via domino multicomponent click reaction. The protocol utilizes readily available starting materials: phenylisothiocyanates, hydrazine monohydrate and diethyl but-2-ynedioate or dimethyl but-2-ynedioate. The reaction condition merely requires mixing of substrates at room temperature without using any catalyst and/or solvent.

An efficient chemoselective synthesis of 1,3-thiazine-2-ylidenes was achieved via annulations of beta-aroyl-thioacetamide with propargyl alcohols using BF(3)OEt(2)as Lewis acid catalyst. A broad spectrum of substrates was well tolerated under the mild reaction conditions producing desired thiazine heterocyclics in good yields.

Synthesis of Pyrazine-1,3-thiazine Hybrid Analogues as Antiviral Agent Against HIV-1, Influenza A (H1N1), Enterovirus 71 (EV71), and Coxsackievirus B3 (CVB3)

A novel series of pyrazine-1,3-thiazine hybrid conjugates were synthesized in excellent yield. These derivatives were subsequently tested against human immunodeficiency virus (HIV-1); hemagglutinin type 1 and neuraminidase type 1’influenza’ A (H1N1) virus; enterovirus 71 (EV71); and coxsackievirus B3. The effect of these conjugates on the key enzymes responsible for the progression of these viral infections was also illustrated via enzyme-based assay, such as HIV-1 reverse transcriptase (RT) and neuraminidase, where entire tested molecules showed considerable inhibition. Particularly, among the tested derivatives, compound 3k was identified as most promising inhibitor of HIV-1 with 94% of inhibition (IC50 3.26 +/- 0.2 m). Moreover, the compound 3d was found to be the most potent analogue to inhibit the H1N1 virus with IC50 of 5.32 +/- 0.4 m together with inhibition of the neuraminidase enzyme (IC50 11.24 +/- 1.1 m). In regard to inhibitory activity against enterovirus 71 (EV71) and coxsackievirus B3 (CVB3), the tested derivatives showed considerable inhibition of infection. Molecular docking studies were also performed for the most promising inhibitors with their corresponding target protein to exemplify the structural requirement for better inhibitory activity. The results of inhibitory assay showed that designed molecules possess considerable inhibitory activity against the virus tested.

DECOLORIZATION KINETICS OF ACID AZO DYE AND BASIC THIAZINE DYE IN AQUEOUS SOLUTION BY UV/H2O2 AND UV/FENTON: EFFECTS OF OPERATIONAL PARAMETERS

The photochemical decolorization of two dyes, namely Acid Yellow 54 and Basic Blue 9, was studied using the UV/H2O2 and UV/Fenton processes. The effects of the amount of H2O2 and FeSO4 as well as the initial pH solution on decolorization kinetics of both the dyes were investigated. The pseudo-first order kinetic model was applied to predict the decolorization of the selected dyes at the different operational conditions and results showed that this model fitted very well with the experimental data. The obtained results also showed the efficiency of UV/Fenton process to quickly degrade aqueous effluents polluted by Acid Yellow 54 and Basic Blue 9 compared to the UV/H2O2 process.

Site-Selective Pd-Catalysed Fujiwara-Moritani type Reaction of N,S-Heterocyclic Systems with Olefins

Dihydro-1,4-thiazine skeletons bearing olefin fragment at their alpha-position were prepared through a Pd(OAc)(2)-catalysed Fujiwara-Moritani type reaction via C-H alkenylation with olefins. This approach is selective, generalizable to a wide range of olefins and requires only 1 eq. of Ag2CO3 without the need of co-oxidant. The C-H bond activation proved to be strongly dependent on the olefin’s substitution while unfused dihydro-1,4-thiazines seemed to be affected by the oxidation state of the sulfur atom. The utility of olefins obtained was demonstrated by their implication in the dipolar cycloaddition reaction with a non-stabilized azomethine ylide.

SYNTHESIS AND ANTICANCER ACTIVITY OF DIHYDROPYRIMIDINTHIONE, 1,3-THIAZINE AND 2-OXONICOTINONITRILE DERIVATIVES

A series of 2 -imino-1,3-thiazine, dihydropyrimidinthiones was synthesized by Que.:pot reaction of cuminaldehyde, thiourea and:ketone derivatives in the presence of HCl. Reaction of cuminaldehyde, ethyl cyanoacetate and ketone derivatives in the presence of ammonium acetate afforded different products of 2-oxonicotinonitrile derivatives. The structure of new synthesized compounds was elucidated by IR, NMR and elemental analysis. Some of the new compounds Were screened for anticancer activity and significant results were found in condensed 1,3-thiazine with five-membered ring, but not higher than the Doxorubicin.

Synthesis and antihypotensive properties of 2-amino-2-thiazoline analogues with enhanced lipophilicity

In a search of nitric oxide synthase inhibitors with prolonged vasoconstrictive activity, a series of lipophilic cyclohexafused 2-amino-2-thiazolines was obtained via cyclization of tert-butyl- or benzoyl-substituted N-(cyclohex-2-en-1-yl)thioureas. The crystal structure of intermediate N-[(3aRS,7aSR)-3a,4,5,6,7,7a-hexahydro-1,3-benzothiazol-2-yl]benzamide hydrobromide was determined by X-ray analysis. One compound was found to cause pronounced and prolonged vasoconstrictive effect after single injection to the Wystar rats with lipopolysaccharide induced acute endotoxic (vasodilatation) shock.

Residualization Rates of Near-Infrared Dyes for the Rational Design of Molecular Imaging Agents

Near-infrared (NIR) fluorescence imaging is widely used for tracking antibodies and biomolecules in vivo. Clinical and preclinical applications include intraoperative imaging, tracking therapeutics, and fluorescent labeling as a surrogate for subsequent radiolabeling. Despite their extensive use, one of the fundamental properties of NIR dyes, the residualization rate within cells following internalization, has not been systematically studied. This rate is required for the rational design of probes and proper interpretation of in vivo results. In this brief report, we measure the cellular residualization rate of eight commonly used dyes encompassing three core structures (cyanine, boron-dipyrromethene (BODIPY), and oxazine/thiazine/carbopyronin). We identify residualizing (half-life > 24 h) and non-residualizing (half-life < 24 h) dyes in both the far-red (similar to 650-680 nm) and near-infrared (similar to 740-800 nm) regions. This data will allow researchers to independently and rationally select the wavelength and residualizing nature of dyes for molecular imaging agent design. A comparative study for removal of different dyes over M/TiO2 (M = Cu, Ni, Co, Fe, Mn and Cr) photocatalysts under visible light irradiation

The purpose of this work is to design and develop a series of stable and effective TiO2 photocatalysts prepared by a modified precipitation method. In order to improve the photo response of the TiO2 to the visible region, the effect of addition of six successive transition metal elements of fourth period, namely chromium (Cr), manganese (Mn), iron (Fe), cobalt (Co), nickel (Ni), and copper (Cu) was studied. Two different types of dyes, methyl orange (MO, azo dye) and methylene blue (MB, thiazine dye group), were used to investigate the influence of structure of dyes on the photocatalytic degradation rates. The results showed that loading titania with 0.3 wt.% of different transition metal elements was found to have significant influences on the crystallographic structure, physical properties and optical absorption properties of titania based catalysts, as well as catalytic activity during dye degradation reactions. The optical reflection edge was obviously shifted to the visible light range with the transition metal additives. The band gap energies of the doped TiO2 samples were considerably narrower than that of plain TiO2. The different activity levels of the catalysts for both dyes clearly show that the photocatalytic activity of samples strongly depended on the metal dopant used. Among all the catalysts, Cu/TiO2 sample exhibited the highest photocatalytic activity under visible light for both of the dyes owing to the low band gap energy and delayed electron-hole recombination.
