A series of isomeric dithieno[1,4]thiazines is accessible through an intermolecular-intramolecular Buchwald-Hartwig amination starting from dihalodithienyl sulfides. The electronic properties of dithieno[1,4]thiazine isomers differ conspicuously over a broad range depending on the thiophene-thiazine anellation: a large cathodic (340 mV) or an anodic shift (130 mV) of the redox potentials relative to corresponding phenothiazines is possible. Structure-property relationships of the dithieno[1,4]thiazine constitution derived from DFT calculations and cyclic voltammetry not only reveal increased electron density but also different delocalization of the radical cations that determines the electrochemical properties significantly. Effect of Substituents in Hydrolyzed Cephalosporins on Intramolecular O-H center dot center dot center dot N Bond

Model molecular systems structurally similar to the transition state of the limiting step of the hydrolysis of cephalosporin antibiotics by the L1 metallo-beta-lactamase are studied. The series of fluorinated compounds show that the nature of substituents in thiazine and beta-lactam rings have a great impact on the strength of the intramolecular O-H center dot center dot center dot N hydrogen bond that determines the catalytic parameters in real biological systems. The strengthening or weakening of the O-H center dot center dot center dot N bond is registered via a quantum topological analysis of the electron density, supplemented with various bonding descriptors’ study. The obtained data are confirmed by the analysis of the vibrational frequency shift relatively to the nonfluorinated compound for the O-H stretching mode of the carboxylic group involved in the O-H center dot center dot center dot N bond formation. The absence of the monotonic dependence of the hydrogen bond strength on the donor-acceptor effects of substituents shows that considered bonding descriptors do not provide a complete understanding of the bonding mechanisms in the active center of L1 metallo-beta-lactamase.

One-pot two-step synthesis of fused thiazinofuranone linked geminal bis 1,2,3-triazole hybrids and their in vitro cytotoxic screening

In this study, a series of novel geminal bis 1,2,3-triazoles linked to 2H-furo[2,3-d][1,3]thiazine-2,4,5(1H,6H)-trione (3a-3m) were prepared in one pot starting from 1,3-thiazine-2,6-dione (1) to 6,6-diazido-2H-furo[2,3-d][1,3]thiazine-2,4,5(1H,6H)-trione (2) followed by Cu(I)-catalyzed azide-alkyne cycloaddition. The synthesized compounds were further explored for in vitro cytotoxic activity against PC3, A549, MCF-7, and HeLa cell lines and results revealed that the five compounds 3c, 3d, 3g, 3l, and 3m have displayed comparable in vitro cytotoxic activity with the standard drug Etoposide.

Selected azaphenothiazines inhibit delayed type hypersensitivity and carrageenan reaction in mice

Several, previously selected azaphenothiazines, as strongly antiproliferative agents in in vitro models, were subjected to evaluation for their potential immunosuppressive effects in the model of delayed type hypersensitivity (DTH) to ovalbumin (OVA) in BALB/c mice and in foot pad inflammation induced by carrageenan in CBA mice. In the DTH model the compounds were given to mice intraperitoneally (i.p.) in 50 mu g or 250 mu g doses, 1 h before the elicitation of the response. In the carrageenan-induced foot pad inflammation the compounds were given i.p. in 50 mu g or 250 mu g doses, 24 h and 2 h before administration of carrageenan. Among the compounds, the significantly suppressive activities in both models were exhibited only by compound 5 (6-chloroethylureidoethyldiquino[3,2-b;2′,3′-e][1,4]thiazine) and compound 4 (6-acetylaminobuty1-9-chloroquino[3,2-blbenzo[1,4]thiazine). Versatile one-pot synthesis of benzo-fused thiacycles by copper catalysis

A novel, one-pot synthesis of structurally diverse benzo-fused thiacycles via a Cu-catalysed intermolecular C-S coupling/cyclisation tandem process, employing the same catalytic system, has been developed. Thus, 3,4-dihydro-2H-benzo[e][1,3]thiazines and 4H-benzo[e][1,3] thiazines were selectively obtained by this one-pot tandem process from (2-iodophenyl) methanamine and aldehydes or 1-(azido-methyl)-2-iodobenzene, respectively. These reactions proceeded in toluene at 100 degrees C with potassium thioacetate, thiobenzoate or ethyl xanthogenate in moderate to good isolated yields. The first one avoids the imine isolation step. A similar approach to benzo[b] thiophene derivatives from 2-(2- iodophenyl) acetonitrile was also developed.

Thiazine Red(+) platelet inclusions in Cerebral Blood Vessels are first signs in an Alzheimer’s Disease mouse model

Strong evidence shows an association between cerebral vascular diseases and Alzheimer’s disease (AD). In order to study the interaction of beta-amyloid (A beta) plaques with brain vessels, we crossbred an AD mouse model (overexpressing amyloid precursor protein with the Swedish-Dutch-Iowa mutations, APP_SweDI) with mice expressing green fluorescent protein (GFP) under the flt-1/VEGFR1 promoter in vessels (GFP_FLT1). Our data show, that only very few A beta plaques were seen in 4-months old mice, focused in the mammillary body and in the lateral septal nucleus. The number of plaques markedly increased with age being most prominent in 12-months old mice. Thiazine Red was used to verify the plaques. Several Thiazine Red(+) inclusions were found in GFP(+) vessels, but only in non-perfused 4-months old mice. These inclusions were verified by Resorufin stainings possibly representing cerebral amyloid angiopathy. The inclusions were also seen in non-crossbred APP_SweDI but not in wildtype and GFP_FLT1 mice. In order to characterize these inclusions Flow Cytometry (FACS) analysis demonstrated that platelets were specifically stained by Thiazine Red(+), more pronounced when aggregated. In conclusion, our data show that Thiazine Red(+) inclusions representing aggregated platelets are a first pathological sign in AD before plaque development and may become important therapeutic targets in early AD.

6-Substituted 9-fluoroquino[3,2-b]benzo[1,4]thiazines display strong antiproliferative and antitumor properties

6-Substituted 9-fluoroquino[3,2-b]benzo[1,4]thiazines – a new type of tetracyclic azaphenothiazines -were obtained from of 6H-9-fluoroquinobenzothiazine by the introduction of appropriate substituents to the thiazine nitrogen atom (alkyl, aminoalkyl, amidoalkyl, sulfonamidoalkyl and nitrogen half-mustard groups). The compounds displayed differential cytotoxic as well as antiproliferative actions against human peripheral blood mononuclear cells (PBMC) stimulated with phytohemagglutinin A (PHA). In addition, they suppressed lipopolysaccharide (LPS)-induced tumor necrosis factor alpha (TNF-alpha) production by whole blood human cell cultures. Two compounds (4 and 15, with the propargyl and methanesulfonamidopropyl groups) were selected for further experiments because of lack of cytotoxicity and strong antiproliferative actions. Compound 4 showed strong suppressive actions on growth of L1210, SW948, A-431 and CX-1 tumor cell lines which were close to those of cisplatin, the reference drug (e.g. GI(50) of 2.28 mu g/mL vs. 1.86 mu g/mL for L1210 cells). Further, the compound appeared to be equally effective as cyclosporine A (CsA) in the inhibition of human two-way mixed lymphocyte reaction (MLR). The compound did not significantly inhibit interleukin 2 (IL-2)-induced growth of CTLL-2 cell line. In addition, inhibition of prostaglandin (PG) synthesis by indomethacin or block of PG receptors did not interfere with the inhibitory effect of the compound on PHA-induced cell proliferation. Therefore, it is likely that the compound acts by inhibiting cell cycle as proposed for other phenothiazines. Further studies are required for the elucidation of the mechanism of action and therapeutic utility of these compounds in more advanced in vivo models. (C) 2014 Elsevier Masson SAS.
