Widely Electronically Tunable 2,6-Disubstituted Dithieno[1,4]thiazines-Electron-Rich Fluorophores Up to Intense NIR Emission

2,6-Difunctionalized dithieno[1,4]thiazines were efficiently synthesized by (pseudo)five- or (pseudo)three-component one-pot processes based on lithiation-electrophilic trapping sequences. As supported by structure-property relationships, the thiophene anellation mode predominantly controls the photophysical and electrochemical properties and the electronic structures (as obtained by DFT calculations). From molecular geometries and redox potentials to fluorescence quantum yields in solution, the interaction of the dithieno[1,4]thiazine-core with the substituents causes striking differences within the series of regioisomers. Most interestingly, strong acceptors introduced inanti-antidithieno[1,4]thiazines nearly induce a planarization of the ground-state geometry and a highly intense NIR fluorescence (phi(F)=0.52), whereas an equally substitutedsyn-syndithieno[1,4]thiazine exhibits a much stronger folded molecular structure and fluoresces poorly (phi(F)=0.01). In essence, electrochemical and photophysical properties of dithieno[1,4]thiazines can be tuned widely and outscore the compared phenothiazine with cathodically shifted oxidation potentials and redshifted and more intense absorption bands.

The Metabolism of Methylene Blue and Its Derivatives in Japanese Eel (Anguilla Japonica)

Methylene blue (MB) is commonly used in aquaculture as a fungicide and antidotes. This study was designed to explore the pharmacokinetics of MB in Japanese eel (Anguilla Japonica) immersed in 10 mg L-1 and 20 mg L-1 MB for 0.5 h. The concentrations of MB and its derivatives in the blood, liver, kidney, skin and muscle were determined by HPLC after immersion. The results showed that the changes of drug concentrations in Japanese eel were basically the same in different dose groups, showing a general trend of increasing at first and then decreasing, but the peak time was slightly different. The peak concentration of the drug was positively correlated with the dose level. The peak concentrations of MB in MB (20 mg L-1) group, MB, azure A and azure B in the tissues of Japanese eel were significantly higher than those in MB (10 mg L-1) group. Moreover, MB, azure A and azure B remained for a long time and could still be detected at 64 days, and azure C was not detected in tissues.

Synthesis of 2,6-dicarbethoxy-3,5-diaryltetrahydro-1,4-thiazine-1, 1-dioxide derivatives as potent anticonvulsant agents

An efficient synthesis of 2,6-dicarbethoxy-3,5-diaryltetrahydro-1,4-thiazine-1,1-dioxide derivatives has been achieved under aqueous medium for the first time in good to excellent yields. All the synthesized compounds were tested for anticonvulsant activity using the maximal electroshock (MES), subcutaneous pentylenetetrazole (scPTZ) screens, which are the most broadly employed seizure models for early identification of candidate anticonvulsants. Their neurotoxicity was determined applying the rotarod test. Seven compounds 4a, 4d, 4f, 4h, 4o, 4p and 4q showed promising anticonvulsant activities in both models employed for anticonvulsant evaluation. The most active compound 4d showed the MES-induced seizures with ED50 value of 10.2 mg/kg and TD50 value of 288.6 mg/kg after intraperitoneal injection to mice, which provided compound 4d with a protective index (TD50/ED50) of 28.3 in the MES test. Synthesis and biological evaluation as well as in silico studies of arylpiperazine-1,2-benzothiazine derivatives as novel anti-inflammatory agents

Novel arylpiperazine-1,2-benzothiazine derivatives have been designed and synthesized as potential antiinflammatory agents. Their structure and properties have been studied using spectroscopic techniques (H-1 NMR, C-13 NMR, FT-IR), MS, elemental analyses, and single-crystal X-ray diffraction (SCXRD, for compound 7b). This study aimed to evaluate the inhibitory activity of new derivatives against both cyclooxygenase isoforms COX-1 and COX-2 due to the similarity of new compounds to oxicams drugs from the NSAIDs group. All new compounds were divided into two series – A and B – with a different linker between thiazine and piperazines nitrogens. Series A included the three-carbon aliphatic linker and series B – two-carbon with a carbonyl group. According to in vitro and molecular docking studies all new compounds exhibited cyclooxygenase inhibitory activity. The series of A compounds included COX-1 inhibitors only. In contrast, the B series showed inhibition of both COX-1 and COX-2, which suggested the importance of the acetoxy linker for COX-2 inhibition. Moreover, the most selective compound 7b, towards COX-2, was non-toxic for the normal human cell line (in concentration of 10 mu M) comparable to reference drug meloxicam. Additionally, investigation of influence on model membranes confirmed the ability of the compound 7b to penetrate lipid bilayers which seemed to be important to the influence with membrane protein-cyclooxygenase.

A facile access and evaluation of some novel thiazole and 1,3,4-thiadiazole derivatives incorporating thiazole moiety as potent anticancer agents

Background: Many heterocyclic compounds containing thiazole or 1,3,4-thiadiazole ring in their skeletons have been reported to possess various pharmacological activities especially anticancer activities. Results: 4-Methyl-2-phenylthiazole-5-carbohydrazide (2) was used as a synthon to prepare 2-(4-methyl-2-phenylthiazole-5-carbonyl)-N-phenylhydrazinecarbothioamide (3) and 2-(2-(4-methyl-2-phenylthiazole-5-carbonyl) hydrazono)-N’-phenylpropane hydrazonoyl chlorides 5a-c. In addition, thioamide 3 was used as starting material for preparation of a new series of thiadiazole derivatives via its reaction with hydrazonoyl chlorides 5a-c in dioxane using triethylamines as catalyst. In addition, a series of thiazole derivatives was synthesized by reaction of thioamide 3 with a number of a-halo compounds, namely, 3-chloropentane-2,4-dione (8) or 2-chloro-3-oxo-N-phenyl butanamide (10) phenacyl bromide 12 ethyl chloroacetate (14) in EtOH in the presence of triethylamine. The structures assigned for all the new products were elucidated based on both elemental analyses and spectral data and the mechanisms of their formation was also discussed. Moreover, the new products was evaluated in vitro by MTT assays for their anticancer activity against cell lines of Hepatocellular carcinoma cell line (HepG-2). The best result observed for compounds 7b (IC50 = 1.61 +/- 1.92 (mu g/mL)) and 11 (IC50 = 1.98 +/- 1.22 (mu g/mL)). The structure-activity relationships have been suggested based on their anticancer results. Conclusions: A novel series of new pharmacophores containing thiazole moiety have been synthesized using a facile and convenient methods and evaluated as potent anticancer agents.

A multicomponent access to 1,3-thiazine-6-phenylimino-5-carboxylates

The multicomponent reaction of ethyl 3-aminocrotonate (1), substituted phenylisothiocyanates (2a-i) and acetic anhydride (7), afforded facile access to a series of substituted 1,3-thiazine-6-phenylimino-5-carboxylates under mild conditions in 15-65% yields. Synthesis of Some Novel Thiazolo[3,2-a]pyrimidine and Pyrimido[2,1-b][1,3]thiazine Derivatives and their Antimicrobial Evaluation

A 2-(2-Mercapto-4-(4-phenoxyphenyl)-6-(thiophen-2-yl)-1,6-dihydropyrimidin-5-yl) acetic acid was used as a reactive key precursor to design various pyrimidine derivatives such as thiazolo[3,2-a]pyrimidines and pyrimido[2,1-b][1,3]thiazines. The chemical structures of the newly synthesized products were confirmed by their elemental analyses and spectral data (IR, H-1 NMR, C-13 NMR, and mass spectra). The antibacterial and antifungal activities of some of the synthesized products were also evaluated, and it was found that compounds 3, 5, 9, and 11 exhibited potent activity against tested microorganisms in comparison with standard drugs.

An unusual electrochemical oxidation of phenothiazine dye to phenothiazine-bi-1,4-quinone derivative (a donor-acceptor type molecular hybrid) on MWCNT surface and its cysteine electrocatalytic oxidation function

Phenothiazine (PTZ), a thiazine class heterocyclic compound, is a well-known electron donating system and has been widely used as a starting compound to prepare various phenothiazine dyes and pharmaceutically important compounds. Quinones and its derivatives are constituents of biologically active molecules serve as excellent electron-acceptor systems. Oxidation of PTZ by chemical and electrochemical methods often resulted into monohydroxylation of benzene ring moiety, S-oxidized and polymerized compounds as end products. Electrochemical oxidation of PTZ on a multiwalled carbon nanotube (MWCNT) modified glassy carbon electrode in pH 7 phosphate buffers solution (PBS) has been investigated in this work. A highly redox active surface confined PTZ-bi-1,4-quinone derivative (PTZ-biQ) on MWCNT modified glassy carbon electrode, designated as GCE/MWCNT@PTZ-biQ, as a product was unusually observed. The GCE/MWCNT@PTZ-biQ showed well-defined redox peaks at E-1/2 = -0.07 and +0.29 V vs Ag/AgCl corresponding to surface confined electron-transfer behavior of the bi-quinone (acceptor) and PTZ-cationic radical species (donor) respectively. No such electrochemical characteristics were noticed when unmodified GCE was subjected to the electrochemical oxidation of PTZ. Existence of PTZ-biQ was confirmed by XRD, Raman spectroscopy, FT-IR and GC-MS (methanolic extract of the active layer) analyses. Position of biQ in PTZ-biQ as 1,4-quinone isomer was confirmed by observation of absence of copper-complexation with 1,4-quinone and H2O2 electrochemical reduction reactions at -0.1 V vs Ag/AgCl unlike to the specific copper-complexation and H2O2 reduction with 1,2-quinone isomer in pH 7. Cysteine (CySH) oxidation was studied as a model system to understand the electron-transfer function of the MWCNT@PTZ-biQ. A highly selective electrocatalytic oxidation and sensing by amperometric i-t and flow injection analysis of CySH at low oxidation potential, 0.3 V vs Ag/AgCl in pH 7 PBS with detection limit values (signal-to-noise ratio = 3) of 11.10 mM and 110 nM respectively, without any interference from other biochemicals like uric acid, dopamine, nitrite, citric acid and H2O2, unlike the conventional chemically modified electrodes with serious interference’s, have been demonstrated. A comparative study of media polarity effects on the linear and third-order nonlinear optical responses of thiazine dyes

In this paper, solvent effects on the linear and nonlinear optical responses of thiazine dyes were investigated. Spectroscopic and open aperture Z-scan techniques were used for studying linear and nonlinear responses of azure A and methylene blue dyes. The experimental results show that linear and nonlinear optical properties of used dyes depend highly on the molecular structure and their surrounding media characteristics. The calculated contribution of various media-induced interactions indicate that solvent hydrogen bond acceptor and solvent dipolarity and polarizability abilities have significant effects on the linear and nonlinear absorption characteristics of thiazine dyes. Moreover, by transition of the linear optics domain to nonlinear optics, similar resonance structures were obtained for thiazine dyes. So, the spectroscopic technique can be considered as a simple method for prediction of dominant resonance structures and effective solvent polarity parameters on the third-order nonlinear properties of azure A and methylene blue dyes.
