Cresyl Violet Stains Mast Cells Selectively: Its Application to Counterstaining in Immunohistochemistry

The thiazine dye toluidine blue (TB) is well known to stain mast cells and hyaline cartilage metachromatically, and thus is mostly often used for their identification. However, TB is not suitable for counterstaining in immunohistochemistry, because of its high-background staining in the cytoplasm of other cell species and in extracellular structures. To expand the knowledge about dyestuffs staining mast cells in consideration with their usage in immunohistochemistry, we determined the stainability of several thiazines and oxazines, which are structurally related compounds to TB, using sections of mast cell-containing tissues. We found that all azine dyes used metachromatically stained mast cells and cartilage. Among these dyes, an oxazines cresyl violet (CV) stained mast cells with lower background, suggesting that those are useful for detecting mast cells and for counterstaining in immunohistochemistry. To ascertain its utility, CV was used in immunostaining of bHSDs in sections from adult rat ovary. Immunopositive signals reflected by DAB development in brown were clearly detected even after CV staining. We conclude that, similar to thiazines, oxazines stain mast cells metachromatically, and that of these, CV is more useful as a counterstain in immunohistochemistry than TB.

Synthesis of New Azoles and Azolopyrimidines Incorporating Morpholine Moiety as Potent Anti-Tumor Agents

A new series of morpholinyl-chalcones 2a-d was prepared by reaction of 2-oxo-N,4-diarylbut-3-enehydrazonoyl chlorides 1a-d with morpholine. These chalcones were used as a building block for constructing pyrazoles 3a-d and 3,4-dihydropyrimidine-2(1H)-thione 6 via their reaction with phenylhydrazine and thiourea, respectively. Moreover, a new series of azolopyrimidine derivatives 11a,b, 15, 17, 19, and 21 incorporating morpholine moiety were synthesized by reaction of 1-morpholino-4-phenyl 1 (2 phenylhydrazono)but-3-en-2-one (2a) with a number of heterocyclic amines in the presence of a catalytic amount of acetic acid. The assigned structures for all the newly synthesized compounds were confirmed on the basis of elemental analyses and spectral data and the mechanisms of their formation were also discussed. All the synthesized compounds were tested for in vitro activities against two antitumor cell lines, human lung cancer (A-549) and human hepatocellular carcinoma (HepG-2) compared with the employed standard antitumor drug (cisplatin) and the results revealed that compounds 6, 8c and 17 have promising activities compared with cisplatin.

The ant Lasius niger is a new source of bacterial enzymes with biotechnological potential for bleaching dye

Industrial synthetic dyes cause health and environmental problems. This work describes the isolation of 84 bacterial strains from the midgut of the Lasius niger ant and the evaluation of their potential application in dye bioremediation. Strains were identified and classified as judged by rRNA 16S. The most abundant isolates were found to belong to Actinobacteria (49%) and Firmicutes (47.2%). We analyzed the content in laccase, azoreductase and peroxidase activities and their ability to degrade three known dyes (azo, thiazine and anthraquinone) with different chemical structures. Strain Ln26 (identified as Brevibacterium permense) strongly decolorized the three dyes tested at different conditions. Strain Ln78 (Streptomyces ambofaciens) exhibited a high level of activity in the presence of Toluidine Blue (TB). It was determined that 8.5 was the optimal pH for these two strains, the optimal temperature conditions ranged between 22 and 37 degrees C, and acidic pHs and temperatures around 50 degrees C caused enzyme inactivation. Finally, the genome of the most promising candidate (Ln26, approximately 4.2 Mb in size) was sequenced. Genes coding for two DyP-type peroxidases, one laccase and one azoreductase were identified and account for the ability of this strain to effectively oxidize a variety of dyes with different chemical structures.

Synthesis and oral hypoglycemic effect of novel thiazine containing trisubstituted benzenesulfonylurea derivatives

A new series of 3-(4-substituted phenyl)-1-(4-(4,6-dimethyl-6H-1,3-thiazin-2-yl)phenylsulfonyl)-1-substituted urea (5a-o) was synthesized by an effectual route via sulfonylcarbamates and explores the novel site for substitution in sulfonylurea as well as the way of thiazine can be prepared. The molecules were established by elemental analysis and spectroscopic viz. IR, H-1 NMR, C-13 NMR and MS techniques. All the fifteen derivatives were shown very prominent oral hypoglycemic effect at the dose of 40 mg/kg body weight (p.o.) in respect of standard drug glibenclamide and control. The hypoglycemic effect was studied using oral glucose tolerance test in normal and NIDDM in STZ-rat model. The compounds 5a, 5d, 5f, 5i, 5k and 5n were dominant out of fifteen derivatives for blood glucose lowering activity (more than 80%) when comparing with NIDDM control. These derivatives were either containing simply phenyl ring (5a, 5f and 5k) on to the second amine of sulfonylurea (R’ = H) or nitro group at the para position in compound 5d, Si and 5n (R’ = NO2) to produce significant oral hypoglycemic effect. Other structural activity relationship is also observed regarding the heteroaromatic and substituted aromatic group at R and R’ position respectively. Tuning the Electronic Structure of Fe(II) Polypyridines via Donor Atom and Ligand Scaffold Modifications: A Computational Study

Fe(II) polypyridines are an important class of pseudo-octahedral metal complexes known for their potential applications in molecular electronic switches, data storage and display devices, sensors, and dye-sensitized solar cells. Fe(II) polypyridines have a d(6) electronic configuration and pseudo-octahedral geometry and can therefore possess either a high-spin (quintet) or a low-spin (singlet) ground state. In this study, we investigate a series of complexes based on [Fe(tpy)(2)](2+) (tpy = 2,2′;6′,2 ”-terpyridine) and [Fe(dcpp)(2)](2+) (dcpp = 2,6-bis(2-carboxypyridyl)pyridine). The ligand field strength in these complexes is systematically tuned by replacing the central pyridine with five-membered (N-heterocyclic carbene, pyrrole, furan) or six-membered (aryl, thiazine-1,1-dioxide, 4-pyrone) moieties. To determine the impact of ligand substitutions on the relative energies of metal-centered states, the singlet, triplet, and quintet states of the Fe(II) complexes were optimized in water (PCM) using density functional theory at the B3LYP+D2 level with 6-311G* (nonmetals) and SDD (Fe) basis sets. It was found that the dcpp ligand scaffold allows for a more ideal octahedral coordination environment in comparison to the tpy ligand scaffold. The presence of six-membered central rings also allows for a more ideally octahedral coordination environment relative to five-membered central rings, regardless of the ligand scaffold. We find that the ligand field strength in the Fe(II) polypyridines can be tuned by altering the donor atom identity, with C donor atoms providing the strongest ligand field.

Crystal structure of cyclic sulfin- and sulfonamides of the thiazine series: Conformation, intra- and intermolecular interactions

An X-ray crystallographic study of a new compound is performed and the structure of five sulfin- and sulfonamides of the thiazine series is discussed. The conformation of the thiazine ring in all structures (a distorted boat) is stabilized by the intramolecular interaction of the C-HaEuro broken vertical bar N type. The nitrogen atom of the thiazine ring has a pyramidal configuration. The geometry of isolated molecules is calculated at density functional theory level (PBE1PBE, 6-31G(d,p)) and compared to that observed in the crystals. In the crystal structures different packing motifs are implemented with the formation of supramolecular associates of different types due to classical hydrogen bonds such as N-H center dot center dot center dot O.

Synthesis of new hybrid 1,4-thiazinyl-1,2,3-dithiazolyl radicals via Smiles rearrangement

The condensation reaction of 2-aminobenzenethiols and 3-aminopyrazinethiols with 2-amino-6-fluoro-N-methylpyridinium triflate afforded thioether derivatives that were found to undergo Smiles rearrangement and cyclocondensation with sulphur monochloride to yield new hybrid 1,4-thiazine-1,2,3-dithiazolylium cations. The synthesized cations were readily reduced to the corresponding stable neutral radicals with spin densities delocalized over both 1,4-thiazinyl and 1,2,3-dithiazolyl moieties.

Removal behaviour of a thiazine, an azo and a triarylmethane dyes from polluted kaolinitic soil using electrokinetic remediation technology

In this study, we investigated the potentiality of the electro-kinetic remediation (EKR) technique for the removal of organic dyes polluted soil. Methylene blue (MB), methyl orange (MO), and phenol red (PR) are representing a thiazine, an azo, and a tryarilmethane dye respectively, which was spiked with kaolinite were selected as a model for pollutant dyes tests. An EKR tool (15 cm length) equipped with a DC electric current with the maximum values of 30 V. Graphite electrodes were used for both anode and cathode was set up for two weeks operation. As a result, only 40-55 % of dye was removed from the soil sections by using distilled water. However, by the addition of some electrolytes; the percentage of dyes removed from the soil increased from 73-76 % and 85-89 % for sodium sulphate, and monosodium dihydrogen phosphate, respectively. It resulted that 55-64 % of dyes was removed without controlling the pH. The significant improvement was achieved by controlling the pH of the system. By controlling the pH in the cathode chamber, only 23 % of MB, 25 % of MO, and 18 % of PR dyes remain in the soil sections, respectively. While by controlling the pH in the anode chamber, almost 90 % of tested dyes could be removed from the kaolinite chamber effectively. The movement of a thiazine dye, from the anode to the cathode chamber was controlled by electro-migration and electro-osmosis phenomena. An azo dye transported from the cathode to the anode chamber by a similar process. However, a triarylmethane dye was removed from the soil sections by only electro-osmosis process. For three kinds of tested dyes, it were found that electro-osmotic flow moving from the anode to the cathode directions. The ageing of dye affects the removal percentage of the dye. Synthesis and In Vitro Anticancer Evaluation of Symmetrically Bridged 1,3-thiazine Derivatives

A series of bis-1,3-thiazine derivatives 3a-o were synthesized from the condensation reactions of symmetric dialdehydes 1a-c possessing aliphatic ether spacer units with 3-substituted-amino-2-cyano-3-mercaptoacrylamides 2a-e. The chemical structures of the products were fully characterized by using different spectroscopic techniques, such as H-1 NMR, C-13 NMR, IR, electron impact mass spectrometry, and elemental analysis. Compounds 3a, 3f, and 3k underwent ring opening followed by recyclization and alkylation in basic medium to afford bis-pyrimidinones 4a-c and 5a-c. The anticancer potential of the new bis-1,3-thiazines was assessed in vitro against six different human cell lines, including lung A549, colon HCT116, breast MCF-7, prostate PC3, liver HepG2, and normal melanocyte HFB4. The results revealed a potent activity of compounds 3e and 3k against breast and liver cancer cell lines in comparison with the reference drug doxorubicin with no noticeable toxicity on normal cells.

Laccase mediator system obtained from a marine spore exhibits decolorization potential in harsh environmental conditions

Laccases play a significant role in remedying dye pollutants. Most of these enzymes are originated from terrestrial fungi and bacteria, thus they are not proper to be used in the environments with neutral/alkaline pH, or they may require laborious extraction/purification steps. These limitations can be solved using marine spore laccases through high stability and easy to use application. In the current study, laccase activity of the marine spore-forming Bacillus sp. KC2 was measured according to the guaiacol and syringaldazine oxidation. Abiotic stresses like pH of 6, temperature of 37 degrees C and 0.3 mM CuSO4 (in comparison with optimal sporulation conditions: pH of 8, temperature of 20 degrees C and 0.0 mM CuSO4) enhanced laccase formation in sporal coat. Maximum activity of enzyme was observed at 50 degrees C and pH 7, which did not change in the alkaline pH and temperature range of 20-70 degrees C. Results indicated ions, inhibitors and solvent stability of the enzyme and its activity were stimulated by Co2+, Mn2+, PMSF, acetone, acetonitrile, ethanol, and methanol. The spore laccase could decolorize synthetic dyes from various chemical groups including azo (acid orange, amaranth, trypan blue, congo red, and amido black), indigo (indigo carmine), thiazine (methylene blue, and toluidine blue), and triarylmethane (malachite green) with ABTS/syringaldazine mediators after 5 h. Degradation products were not toxic against Sorghum vulgare and Artemia sauna model organisms. The enzyme mediator system showed high potentials for dye bioremediation over a wide range of harsh conditions.
