Determination of Lipophilicity of Allyl Thiosemicarbazide, N-1-Thiocarbamylamidrazone Derivatives, and their Cyclic Products by RP-HPLC, RP-TLC, and Theoretical Methods: Effects of Selected Compounds on the CNS of Mice

The allyl thiosemicarbazide, N-1-thiocarbamylamidrazone derivatives, and the products of their cyclization: the 1,2,4-triazole, 1,3,4-thiadiazole, 1,2,4-triazole[3,4-b]1,3-thiazine, 1,2,4-triazole[3,2-b]1,3-thiazine, 1,3,4-thiadiazole[3,2-a]pyrimidine, 5,6-dihydrothiazolo[2,3-c][1,2,4]triazole, and benzoic acid derivatives have been analyzed by RP-HPLC and RP-TLC methods using the methanol-water mixtures as the mobile phase and the octadecyl stationary phase. The lipophilicity was expressed as the chromatographically derived descriptors: mean of k(mk), mean of logk (mlogk), logk(W), S, and phi(0), scores of k, logk, and R-M corresponding to the first principal component. Additionally, the authors propose to introduce a descriptor (logk(m)) corresponding to the residual specific interactions. Chromatographic parameters of lipophilicity were compared with the partition coefficient (logP) calculated by various softwares (milogP, clogP, AlogPs, AClogP, AlogP, MlogP, KOWWIN, XlogP2, XlogP3). The matrices were created with logk(W) or R-MW and logP and they have been the subject of PCA analysis. The effects of the selected compounds on the central nervous system (CNS) of mice were studied.

Studies on Synthesis of Novel Triazole Tagged Pyrazole Fused Naphthalene 5-Thiazine-5,5-dioxide Derivatives, Their Antimicrobial, and Antioxidant Activity

A series of novel triazole tagged pyrazole fused naphthalene-5-thiazine-5,5-dioxide derivatives 8 and 9 were synthesized starting from sodium salt of saccharin 1. The structure of each intermediate and products was established on the basis of spectroscopy data. All the synthesized compounds 8 and 9 were screened against various bacterial and fungal strains but found to show no activity up to 150-mu g/mL concentration. Further screening for antioxidant property resulted promising compounds.

Synthesis and selected immunological properties of 10-substituted 1,8-diazaphenothiazines

A new type of tricyclic azaphenothiazines-1,8-diazaphenothiazines-was obtained in the reaction of 2,3- and 3,4-disubstituted pyridines. The reaction ran as the Smiles rearrangement. The 1,8-diazaphenothiazine system was determined using NOE experiment and 2D NMR spectra (COSY, HSQC, HMBC). 10H-1,8-diazaphenothiazine was transformed into 10-derivatives with alkyl, aminoalkyl, amidoalkyl, sulfonamidoalkyl, and nitrogen half-mustard groups. The compounds were tested for their effects on phytohemagglutinin A-induced proliferative response of human peripheral blood mononuclear cells (PBMC) and lipopolysaccharide-induced tumor necrosis factor alpha production by human whole blood cultures. The compounds exhibited differential, dose-dependent inhibitory activities in these tests. All the compounds were low toxic against PBMC. The compounds showing the highest antiproliferative activity strongly inhibited the growth of leukemia L-1210 and colon cancer SW-948 cell lines, similarly as cisplatin, a reference drug.

Synthesis, Cytotoxicity Evaluation, Molecular Docking and Utility of Novel Chalcones as Precursors for Heterocycles Incorporating Pyrazole Moiety

Background: Chalcones, 2-pyrazolines and thiazoles have been reported to possess various pharmacological activities. Objective: Synthesis of new chalcones and utilizing them as a building block for constructing a series of thiazole derivatives and evaluating some of them as anticancer agents. Method: The new compounds were synthesized via stirring at room temperature or thermal heating. Cytotoxic evaluation of the new synthesized compounds was tested using the method of Skehan et al. Moreover, the computational studies were performed using MOE 2014.09 software. Result: A series of new chalcones were prepared by the reaction of ethyl 3-acetyl-1-aryl-5-methyl-1H-pyrazole-4-carboxylate with a number of substituted benzaldehydes. One of these chalcones was used as a building block for constructing a pyrazoline ring via its reaction with thiosemicarbazide. The produced carbothioamide derivative was used for the preparation of two series of thiazole derivatives by its reaction with a number of hydrazonoyl chlorides. Moreover, reaction of 3-acetylpyrazole thiosemicarbazone derivative with a number of N-aryl-2-oxopropane hydrazonoyl chlorides afforded 5-arylazothiazole derivatives. The assigned structures for all the newly synthesized compounds were confirmed on the basis of elemental analyses and spectral data. Some of the newly synthesized chalcones and thiazoles were tested for their cytotoxicity against human colon carcinoma cell line (HCT-116) and the molecular docking was carried out on the most active compound 3f. Conclusion: The results of the anticancer activity revealed that compounds 3f, 3e, 3c and 3b have promising activities compared with the standard drug Doxorubicin. Moreover, the computational studies confirm the results of biological activity. Also, the ADME profile study showed that compound 3f can be considered as a promising drug by conducting good pharmacokinetic and medicinal chemistry tests.

Two new compounds from Xanthium strumarium

One new lignan, fructusol A (1), and one new thiazine derivative, 2-hydroxy-xanthiazone (2), along with eight known ones, were isolated from the seeds of Xanthium strumarium.
