Pathological conformations involving the amino terminus of tau occur early in Alzheimer’s disease and are differentially detected by monoclonal antibodies

Conformational changes involving the amino terminus of the tau protein are among the earliest alterations associated with tau pathology in Alzheimer’s disease and other tauopathies. This region of tau contains a phosphatase-activating domain (PAD) that is aberrantly exposed in pathological forms of the protein, an event that is associated with disruptions in anterograde fast axonal transport We utilized four antibodies that recognize the amino terminus of tau, TNT1, TNT2 (a novel antibody), Tau12, and Tau13, to further study this important region. Using scanning alanine mutations in recombinant tau proteins, we refined the epitopes of each antibody. We examined the antibodies’ relative abilities to specifically label pathological tau in non-denaturing and denaturing assays to gain insight into some of the mechanistic details of PAD exposure. We then determined the pattern of tau pathology labeled by each antibody in human hippocampal sections at various disease stages in order to characterize PAD exposure in the context of disease progression. The characteristics of reactivity for the antibodies fell into two groups. TNT1 and TNT2 recognized epitopes within amino acids 7-12 and specifically identified recombinant tau aggregates and pathological tau from Alzheimer’s disease brains in a conformation-dependent manner. These antibodies labeled early pre-tangle pathology from neurons in early Braak stages and colocalized with thiazine red, a marker of fibrillar pathology, in classic neurofibrillary tangles. However, late tangles were negative for TNT1 and TNT2 indicating a loss of the epitope in later stages of tangle evolution. In contrast, Tau12 and Tau13 both identified discontinuous epitopes in the amino terminus and were unable to differentiate between normal and pathological tau in biochemical and tissue immunohistological assays. Despite the close proximity of these epitopes, the antibodies demonstrated remarkably different abilities to identify pathological changes in tau indicating that detection of conformational alterations involving PAD exposure is not achieved by all N-terminal tau antibodies and that a relatively discrete region of the N-terminus (i.e., amino acids 7-12, the TNT1 and TNT2 epitope) is central to the differences between normal and pathological tau. The appearance of PAD in early tau pathology and its disappearance in late-stage tangles suggest that toxic forms of tau are associated with the earliest forms of tau deposits. Collectively, these findings demonstrate that the TNT antibodies are useful markers for early conformational display of PAD and provide information regarding conformational changes that have potential implications in the toxic mechanisms of tau pathology. New pyrimidothiazine Derivative as Corrosion Inhibitor for Carbon Steel in Acidic Media

2,8-bis(4-chlorophenyl)-3-hydroxy-4,6-dioxo-4,6-dihydropyrimido[2,1-b][1,3]thiazine-7-carbonitrile (CHPTC) was tested as corrosion inhibitor for carbon steel in 2.0 M H3PO4 by using polarization, electrochemical impedance spectroscopy (EIS) and computational calculations. Potentiodynamic polarization curves indicated that the pyrimidothiazine derivativeas mixed-type inhibitor. Impedance measurements showed that the double-layer capacitance decreased and charge-transfer resistance increased with increase in the inhibitor concentration and hence increasing in inhibition efficiency. The effect of temperature on the corrosion behavior of carbon steel in 2.0 M H3PO4 with and without addition of CHPTC was studied in the temperature range 313-333 K. The adsorption of the inhibitor molecules was in accordance with the Langmuir adsorption isotherm. Quantum chemical approach used to calculate electronic properties of the molecule to ascertain the relation between inhibitive effect and molecular structure.

DFT Calculations, Spectroscopic Studies, Biological Activity and Non Linear Optical Properties (NLO) of Novel Ternary Cu(II)-Chelates Derived from 5-Acetyl-4-hydroxy-2H-1,3-thiazinedione

Characterization of the synthesized complexes was established based on elemental analysis, molar conductance, magnetic susceptibility measurements, spectral (infrared, electronic, mass, H-1-NMR and ESR) as well as thermal gravimetric analysis (TGA). The complexes exhibited octahedral and square planer geometry. The antimicrobial activity for the studied complexes was tested for different kind of organisms. The geometrical and non-linear optical parameters of the studied complexes 1-3 are investigated theoretically at the B3LYP/GENECP level of theory. The optimized geometries of the studied complexes are non-planner as indicated from the dihedral angles. The natural charge population (core, valence and Rydberg), exact electronic configuration, total Lewis, and total non-Lewis is computed and discussed in terms of natural bond orbitals (NBO) analysis. The calculated E-HOMO and E-LUMO energies at the same level of theory of the studied complexes were used to calculate the global properties; hardness (), global softness (S), electrophilicity () and electronegativity (). The total dipole moment ((tot)), total and anisotropy of polarizability (??), () and first hyperpolarizability (??) values were calculated and compared with urea as a reference compound. From the values of the computed first hyperpolarizability (??), the ligand and the studied complexes show promising optical properties.

Selected azaphenothiazines inhibit delayed type hypersensitivity and carrageenan reaction in mice

Several, previously selected azaphenothiazines, as strongly antiproliferative agents in in vitro models, were subjected to evaluation for their potential immunosuppressive effects in the model of delayed type hypersensitivity (DTH) to ovalbumin (OVA) in BALB/c mice and in foot pad inflammation induced by carrageenan in CBA mice. Eco-Friendly Acaricidal Effects of Nylon 66 Nanofibers via Grafted Clove Bud Oil-Loaded Capsules on House Dust Mites

Acaricidal nylon 66 fabrics (AN66Fs) grafted with clove oil-loaded microcapsules (COMCs) were developed against Dermatophagoides farina (D. gallinae). The average diameter was about 2.9 mu m with a range of 100 nm-8.5 mu m. COMCs carried clove oil loading of about 65 vol %. COMCs were chemically grafted to electrospun nylon nanofibers by the chemical reactions between -OH groups of COMCs and -COOH end groups of nylon fabrics to form ester linkages. AN66Fs had an effect on D. farinae depending on COMCs loadings. The increase in COMCs loading of AN66Fs from 5 to 15 wt % increased from 22% to 93% mortality against D. farinae within 72 h. However, AN66Fs containing over 20 wt % COMCs were more effective, showing up to 100% mortality within 24 h because the large amount of monoterpene alcohol, eugenol. This research suggests the use of clove oil and its major constituent eugenol as eco-friendly bioactive agents that can serve as a replacement for synthetic acaricides in controlling the population of D. farinae.

Ultrasmall Plasmonic Nanoparticles Decorated Hierarchical Mesoporous TiO2 as an Efficient Photocatalyst for Photocatalytic Degradation of Textile Dyes

Hierarchical mesoporous TiO2 was synthesized via a solvothermal technique. The sonochemical method was adopted to decorate plasmonic nanoparticles (NPs) (Ag, Au) on the pores of mesoporous TiO2. The crystallinity, structure, and morphology were determined to understand the physicochemical nature of the nanocomposites. The catalytic efficiency of the plasmonic nanocatalysts was tested for the azo dyes (congo red, methyl orange, acid orange 10, and remazol red) under solar and visible light irradiations. The generation of hydroxyl radicals was also studied using terephthalic acid as a probe molecule. An attempt was made to understand the influence of size, work function and Fermi level of the metal NPs toward the efficiency of the photocatalyst. The efficiency of the nanocomposites was found to be in the order of P25 < mesoporous TiO2 < mesoporous Ag-TiO2 < mesoporous Au-TiO2 nanospheres under both direct solar light and visible light irradiation. The results indicated that the adsorption of dye, anatase phase, and surface plasmon resonance of NPs favored the effective degradation of dyes in aqueous solution. Further, the efficiency of the catalyst was also tested for xanthene (rose bengal), rhodamine (rhodamine B, rhodamine 6G), and thiazine (methylene blue) dyes. Both TiO2 and NPs (Ag & Au) possess a huge potential as an eco-friendly photocatalyst for wastewater treatment.
