Widely Electronically Tunable 2,6-Disubstituted Dithieno[1,4]thiazines-Electron-Rich Fluorophores Up to Intense NIR Emission

2,6-Difunctionalized dithieno[1,4]thiazines were efficiently synthesized by (pseudo)five- or (pseudo)three-component one-pot processes based on lithiation-electrophilic trapping sequences. As supported by structure-property relationships, the thiophene anellation mode predominantly controls the photophysical and electrochemical properties and the electronic structures (as obtained by DFT calculations). From molecular geometries and redox potentials to fluorescence quantum yields in solution, the interaction of the dithieno[1,4]thiazine-core with the substituents causes striking differences within the series of regioisomers. Most interestingly, strong acceptors introduced inanti-antidithieno[1,4]thiazines nearly induce a planarization of the ground-state geometry and a highly intense NIR fluorescence (phi(F)=0.52), whereas an equally substitutedsyn-syndithieno[1,4]thiazine exhibits a much stronger folded molecular structure and fluoresces poorly (phi(F)=0.01). In essence, electrochemical and photophysical properties of dithieno[1,4]thiazines can be tuned widely and outscore the compared phenothiazine with cathodically shifted oxidation potentials and redshifted and more intense absorption bands.

Sol-gel (template) synthesis of macroporous Mo-based catalysts for hydrothermal oxidation of radionuclide-organic complexes

Molybdenum compounds are industrially demanding as heterogeneous catalysts for oxidation of various organic substances. Highly porous structure of molybdenum-containing catalysts avoids surface’s colmatation and prevents blocking catalytic sites that makes these materials play a key role in processes of hydrothermal oxidation of radionuclide organic complexes. The study presents an original way of sol-gel synthesis of new macroporous molybdenum compounds using core-shell colloid template (polymer latex) as poreforming agent. We have described three individual routs of template removal via thermal decomposition to obtain porous materials based on molybdenum compounds. Thermal treatment conditions (temperature, gaseous atmosphere) have been studied with respect to their influence on composition, structure and catalytic properties of synthesized molybdenum systems. The optimal way to synthesis of crystal molybdenum (VI) oxide with ordered porous structure (mean pore size 100-160 nm) has been suggested. Catalytic properties of macroporous molybdenum materials have been investigated in the process of liquid phase and hydrothermal oxidation of such organic substances thiazine and stable Co-EDTA complex. It was shown that macroporous molybdenum oxides could be applied as prospective catalysts for hydrothermal oxidation of organic radionuclide complexes during the processing of radioactive waste. Computational strategies to explore antimalarial thiazine alkaloid lead compounds based on an Australian marine sponge Plakortis Lita

In this work, an attempt was made to propose new leads based on the natural scaffold Thiaplakortone-A active against malaria. The 2D QSAR studies suggested that three descriptors correlate with the anti-malarial activity with an R-2 value of 0.814. Robustness, reliability, and predictive power of the model were tested by internal validation, external validation, Y-scrambling, and applicability domain analysis. HQSAR studies were carried out as an additional tool to find the sub-structural fingerprints. The CoMFA and CoMSIA models gave Q(2) values of 0.813 and 0.647, and http://www.w3.org/1999/ values of 0.994 and 0.984, respectively. Using the 2D-QSAR equation, the activity values of the seven modified compounds were calculated and it was found that three molecules showed good anti-malarial activity. Molecular docking of the 42 Thiaplakortone-A derivatives with Plasmodium falciparum calcium-dependent protein kinase 1 (PfCDPK1) was carried out to find out protein-ligand interactions. Data mining of the bioassay data-set AID: 504850 using the classifier based on Random Forest of Weka suggested that all of the eight molecules selected and three out of the seven virtual molecules were anti-malarial active. Both the virtual molecules and drug molecules were docked with CYP3A4, indicating that the virtual molecules could metabolize easily. Toxicity studies using Osiris shows that three molecules showed no toxic characters.

Microwave-assisted regioselective reaction of furanone derivative supported by DFT stimulation and molecular docking to afford controlling insecticidal agents

The furanone 1 was allowed to react with different nitrogen nucleophiles such as hydrazine hydrate, hydroxyl amine, anisidine, anthranilic acid, glycine, ethylglycinate and isatin and some carbon nicleophiles under Michael reaction conditions assisted by microwave irradiation. They afforded new synthesized some important heterocyclic compounds 2-16. The structures of the synthesized new compounds were characterized by spectral data and elemental analysis. DFT calculation is serving to clarify not only the active electrophilic sites attacked by suitable nucleophiles in the organic reaction but also the bioactivity of the larvicidal compounds. The larvicidal activities of the synthesized compounds were tested against Plutella xylostella and Helicoverpa armigera larvae in vitro. The results revealed that the synthesized compounds 4, 13 and 14 showed good insecticidal activity against Plutella xylostella larvae with 100%, 87.2% and 91.4% activities, respectively, while 13 and 14 displayed 92% and %, 94.8 activities, respectively, against Helicoverpa armigera larvae. Therefore, they exhibited good larvicidal activity. The executed molecular docking simulations evinced the selective binding of the novel synthesized compounds toward acetylcholinesterase (AchE) of Drosophila Melanogaster, which signifies their role as a prominent larvicides. Compound 4, 13 and 14 showed clear docking affinity and to the assumed binding sites in terms of both calculated binding energies and the types of interactions, which provided a solid rationalization for the conducted experimental results.

Stereoselective Polymer-Supported Synthesis of Morpholine- and Thiomorpholine-3-carboxylic Acid Derivatives

Herein we report the polymer-supported synthesis of 3,4-dihydro-2H-1,4-oxazine-3-carboxylic acid derivatives using immobilized Fmoc-Ser(tBu)-OH and Fmoc-Thr(tBu)-OH as the starting materials. After the solid-phase-synthesis of N-alkyl-N-sulfonyl/acyl intermediates, the target dihydrooxazines were obtained using trifluoroacetic acid-mediated cleavage from the resin. This approach was also studied for the preparation of dihydrothiazines from immobilized Fmoc-Cys(Trt)-OH. Inclusion of triethylsilane in the cleavage cocktail resulted in the stereoselective formation of the corresponding morpholine/thiomorpholine-3-carboxylic acids. Stereochemical studies revealed the specific configuration of the newly formed stereocenter and also the formation of stable N-acylmorpholine rotamers.

A novel metal-free multicomponent cascade reaction was developed for the construction of thiazine imides. This four-component cascade reaction had advantages of mild reaction conditions, wide substrate scope and good atom economy. An unusual electrochemical oxidation of phenothiazine dye to phenothiazine-bi-1,4-quinone derivative (a donor-acceptor type molecular hybrid) on MWCNT surface and its cysteine electrocatalytic oxidation function

Phenothiazine (PTZ), a thiazine class heterocyclic compound, is a well-known electron donating system and has been widely used as a starting compound to prepare various phenothiazine dyes and pharmaceutically important compounds. Quinones and its derivatives are constituents of biologically active molecules serve as excellent electron-acceptor systems. Oxidation of PTZ by chemical and electrochemical methods often resulted into monohydroxylation of benzene ring moiety, S-oxidized and polymerized compounds as end products. Electrochemical oxidation of PTZ on a multiwalled carbon nanotube (MWCNT) modified glassy carbon electrode in pH 7 phosphate buffers solution (PBS) has been investigated in this work. A highly redox active surface confined PTZ-bi-1,4-quinone derivative (PTZ-biQ) on MWCNT modified glassy carbon electrode, designated as GCE/MWCNT@PTZ-biQ, as a product was unusually observed. The GCE/MWCNT@PTZ-biQ showed well-defined redox peaks at E-1/2 = -0.07 and +0.29 V vs Ag/AgCl corresponding to surface confined electron-transfer behavior of the bi-quinone (acceptor) and PTZ-cationic radical species (donor) respectively. No such electrochemical characteristics were noticed when unmodified GCE was subjected to the electrochemical oxidation of PTZ. Existence of PTZ-biQ was confirmed by XRD, Raman spectroscopy, FT-IR and GC-MS (methanolic extract of the active layer) analyses. Position of biQ in PTZ-biQ as 1,4-quinone isomer was confirmed by observation of absence of copper-complexation with 1,4-quinone and H2O2 electrochemical reduction reactions at -0.1 V vs Ag/AgCl unlike to the specific copper-complexation and H2O2 reduction with 1,2-quinone isomer in pH 7. Cysteine (CySH) oxidation was studied as a model system to understand the electron-transfer function of the MWCNT@PTZ-biQ. A highly selective electrocatalytic oxidation and sensing by amperometric i-t and flow injection analysis of CySH at low oxidation potential, 0.3 V vs Ag/AgCl in pH 7 PBS with detection limit values (signal-to-noise ratio = 3) of 11.10 mM and 110 nM respectively, without any interference from other biochemicals like uric acid, dopamine, nitrite, citric acid and H2O2, unlike the conventional chemically modified electrodes with serious interference’s, have been demonstrated.
